A delayed hypersensitivity reaction to sheep red blood cells (SRBC) was elicited in the footpads of the mice sensitized by subcutaneous (sc) injection of the antigen emulsified in Freund's complete adjuvant (FCA) ; injection of the test antigen to the footpad induced a strong reaction with its peak at 24 hr. The delayed hypersensitivity was first detected on day 3 and reached its maximum on day 5 after sensitization, accompanying no serum antibody production. On the other hand, a humoral response with no delayed hypersensitivity was induced by intraperitoneal (ip) injection of SRBC without FCA.
The delayed hypersensivity was suppressed by ip injection of SRBC given during the period of 7 days before to 2 days after the sensitization by sc injection of the antigen in FCA. However, the humoral response induced by ip injection of SRBC was not affected by sc injection of SRBC in FCA. It was suggested that even if the appearance of the delayed hypersensitivity reaction was suppressed by ip injection of the antigen, sc injection of SRBC in FCA was capable of bestowing the immunological memory for delayed hypersensitivity on the mouse.
