The fluorescence of xanthurenic acid was found to be greatly enhanced after incubating at 37°C in the aqueous solution of dimethyl sulfate and sodium hydroxide This finding was assay procedure of xanthurenic acid in urine. The fluorescence intensity of xanthurenic acid by dimethyl sulfate-sodium hydroxide method was measured at 425nm with excitation at 338 nm. The calibration curve for xanthurenic acid showed a linear relation in the range of 0.25nmol/ml to 7.5nmol/ml. The relative standard deviation at 7.5nmol was 1.1% (n=8). In this procedure, recovery of xanthurenic acid in the six samples of urine was found to be 99.8±11.2% (n=6). Xanthurenic acid contents in urine of healthy adults which were collected for 24h were 3.45±0.89umol/day (male, 32 years old, n=4).
