Promotion of in vitro Shoot Formation from Protocorm-like Bodies of a Hybrid between Tropical and Temperate Cymbidium Species

Abstract

Three different treatments, which are known to promote shoot formation in temperate Cymbidium species, were applied to the protocorm-like bodies (PLBs) of Cymbidium Sleeping Beauty × Cymbidium sinense (Andr.) Willd, a hybrid between tropical and temperate Cymbidium species. Shoot formation from the PLBs was promoted on Murashige and Skoog (MS) medium in which NH₄NO₃ and KNO₃ were reduced to 25 and 50%, respectively. The highest shoot formation rate (77%) was obtained on MS medium supplemented with 0.1 mg·L⁻¹ AgNO₃. The most favorable combination for shoot formation was 10 mg·L⁻¹ of 6-benzyladenine (BA) and 0.1 mg·L⁻¹ α-naphthaleneacetic acid (NAA).
