Aspirin inhibits the platelet cyclooxygenase by acetylation. Stuart et al. reported a technique for the determination of platelet survival based on the assay of platelet malonaldehyde (MDA) after one single oral intake of aspirin. In this study, the platelet survival time was measured simultaneously by 51Cr labeling method and MDA method in 3 normal subjects and 8 patients with ITP. In 3 normal cases, platelet survival times gained by 51Cr-labeling method (9.1±0.71 days) well corresponded to those by MDA method (9.3±0.9 days). In ITP, however, the assay by 51Cr-labeling method gave short survival times (T1/2 1.11±0.81 days), compared with those by MDA method (T1/2 1.54±0.84 day). This difference by two methods in ITP, which was reported previously by several workers, was less in our new analysis. MDA method will become useful for measurement of platelet survival time because of simple and non-radioisotopic method, but in the cases with random destruction of platelets, obtained values must be calculated by mathematical analysis.
