Similarity searching approach in identification of bioactive Sabinene as potential anti-microbial, anti-oxidant and cytoprotective molecule

Bioactive compounds are emerging as alternative therapies with great potential to reduce the disease burden. They usually have no adverse effects and can be easily incorporated into daily diets. Pharmaceutical companies also rely on pure bioactive compounds for new drug development. Cardamom, a well known spice is used to add flavors to food since ancient times and is also known to possess many health benefits. The main objective of our study was to identify and characterize the bioactive compound present in cardamom. We used ligand based similarity searching and molecular docking approach to identify the biological activities of cardamom phytochemicals Data mining approach identified Sabinene, a natural monoterpene abundantly present in Cardamom, to possess potential biological activities. Ligand based similarity searching demonstrated that (+)-3-carene possessed the maximum similarity to Sabinene (similarity-index >50%). Molecular docking studies elucidated that binding modes of the Sabinene were similar to (+)-3-carene against Cytochrome CYP101D1 (PDB ID: 4C9K). Further, by in vitro studies we demonstrated that Sabinene possess anti-microbial activity . We also observed increased DPPH inhibition by increasing Sabinene concentration. Sabinene also protect yeast cells from H2O2 induced cytotoxicity as analyzed by MTT assay. Our results further highlight that Sabinene treatment in yeast cell decreased generation of oxidative stress and showed decreased activity of antioxidant enzymes Glutathione-S-transferase, Catalase and Lipid Peroxidase as compared to untreated cells. To best of our knowledge this is first report which demonstrates the anti-microbial, anti-oxidant and cyto-protective effects of Sabinene.
