Isolation of circulating tumor cells (CTCs) at a single cell level is crucial for molecular analyses of CTCs in blood, which contains large amounts of blood cells. Here, we report development of two types of new microfluidic chip for size-based selection and isolation of CTCs from blood samples. One is device that use drag force and capillary force to trap CTCs on a microfluidic chip with open channels. CTCs are then carried to the meniscus of air-liquid interface due to the drag force generated by a syringe pump and CTCs are stained and the number is counted. The other is a slide glass-sized CTC isolation device using 3D electroforming metal filter and a syringe pump. CTC trapped in the filter is stained and counted. Cell spike experiments with these device demonstrated that the recovery rate from blood was more than 85%. These devices would be useful tools for the rapid and sensitive enumeration, isolation of CTC from blood.
