In this study, we propose a closed microfluidic chip for cell trapping, chromosome stretching, and fluorescence observation on a single chip in a quick and simple operation using centrifugal force. The proposed chip was fabricated using a mold fabricated in batches by 3D lithography with multiple masks. In cell experiments, more cells were trapped compared to the conventional open chip. Since the closed chip did not allow cells to drop out of the structure for rinsing to remove excess cells remaining on the chip, the rinse process improved the trapping rate in the case of the closed chip. In addition, a large number of chromosome stretching was observed on the closed chip.
