This paper proposed a high-speed control system of air-liquid two phase flow to change osmotic pressure in a microfluidic device. In particular, we measured the osmoadaptation of the single cell at the high time resolution over 1 ksps. Conventional studies utilized optical density and stopped-flow spectrophotometry methods, which only showed the response of cell population. Another representative method utilized a microfluidic chip having patterned hydrogel to measure the osmoadaptation for each of single cell. However, it was still difficult to measure the osmoadaptation for a single cell with high time resolution. Therefore, we developed the high-speed solution replacement system to change cell suspension into different osmolality solution using at the volume of 10 pL order droplet. We sought the design strategy of droplet formation and high-speed solution replacement in the microchannel. From the experiment, the solution replacement time of 3.3 ms was achieved. Finally, we demonstrated the proposed microfluidic system that could measure osmoadaptation of single cell under sudden osmotic pressure change.
