Peripheral stem cell assay was done in order to examine the hematopoietic status of myelodysplastic syndorome (MDS) . A patient with refractory anemia, 2 with refractory anemia with excess of blasts (RAEB) and 2 with RAEB in transformation (RAEBt) were studied. Efficiencies of colony-cluster formation in 2 MDS (RAEB, RAEBt) were higher and in 3 MDS (RA, RAEB, RAEBt) were lower than normal subjects. Some of the colony and cluster in 3 MDS patients were composed of myeloperoxidase-negative neutrophils, furthermore, in one of these patients colony and cluster were composed of cells with blastlike morphology. These findings indicate that quantitative and qualitative studies of colony and cluster are useful for the analysis of pathophysiological aspects of MDS.
