Ehrlich ascites cancer cells gradually took up Zn65 from their enviroment of 0.9% NaCl and Tris buffer medium, on incubation, at the temperature of 37°C.
Change of temperature on incubation, pH of medium, and addition of carrier, amino acids, chelating agents and metal ions, changed the uptake of Zn65.
1. Lower temperature of 19°C and 11°C on incubation, diminished the uptake.
2. Within pH 4.9 to 8.1 range of medium, no changes occurred.
3. Carrier inhibited the uptake.
4. Some amino acids were added to the medium of 0. 9% NaCl to the final concentration of 10-3M. L- glutaihione, L-aspartic acid, and DL-and D-penillamine markedly inhibited, L-cysteine and glycine inhibited moderately.
5. Some chelating agents were added to the medium of 0. 9% NaCl to various concentration. Only Ca EDTA at final 10-3M and 10-5M markedly inhibited. Dihidrothioctic acid at final 10-3M and riboflavin at final 200μg, modera tely inhibited, and Na citrate and α-thiola-ctoylglycine inhibited sligtly.
6. Zn2+, Cu2+, Ni2+, Co2+, Fe2+ and Mn2+ at final 10-4M concentration in medium of Tris buffer, inhibited the uptake to various degree.
