The pectic substances differing from each other in molecular weight, or in content of neutral sugar or methoxyl group, were resolved by the DEAE-cellulose chromatography with a gradient of increasing concentration of acetate buffer, pH 6. Acid-soluble acid polysaccharides (YPG-ASAP) produced from pectic acid with endo-polygalacturonase of Saccharomyces fragilis (YPG) and pectic acid, which had been reported not to be resolved with carbonate-bicarbonate buffer, were separated from each other by using this gradient and 0.1N NaOH. Tailing could not be perfectly eliminated even by the gradient elution. When sodium acetate solution was used instead of acetate buffer, pectinic acid and pectic acid were eluted at the same concentration of the eluent.
