Application of the retentions analysis of amino acids and hydroxy-dicarboxylic acids to thin-layer chromatography was examined. Silica gel was used as the adsorbent, and 10% hydrous tetrahydrofuran containing 0.1% cupric acetate and 10% hydrous 2-propanol added with 0.5% of acetic acid were used as the developing solvents. Development was stopped when copper (II) moved 2cm. above the original point, copper (II) ion was detected with Rubeanic acid test solution, and its area was measured to effect quantitative determination. As a result, it was found possible to carry out retentions analysis of five kinds of amino acid and three kinds of hydroxy-dicarboxylic acid in the range of 2∼16 μg. It was also made possible to carry out retentions analysis of a mixture of these substances by initial separation by one-dimensional development, followed by two-dimensional development.
