Anti-inflammatory action of the proprietary drug, Rokusingan (musk, bezoar, bufotalis, borneol, pearl, realgar=2 : 3 : 2 : 2 : 3 : 2), and its componental crude drugs was examined by the filter pellet method. Although the effect of Rokusingan was about 6% of that of hydrocortisone acetate in this action, the complex effect of the combined use of musk, bezoar and bufotalis in 2 : 3 : 2 ratio was found to be responsible for 50% of this action. ID50 values of each of these crude drugs were 388 mg/kg (282-523) in musk, 433 mg/kg (246-767) in bezoar, and 159 mg/kg (114-223) in bufotalis. Based on these values, additive theoretical value was calculated as 284 mg/kg. Since the complex effect of the three drugs, 69.4 mg/kg, is smaller than the theoretical value, this complex effect is considered to be due to the concerted action of these three drugs. It was revealed that the concerted effect of musk and bezoar in 2 : 3 ratio was the principle one in this complex effect. Combined effect of borneol, pearl, and realgar in 2 : 3 : 2 ratio is extremely weak and these drugs probably inhibit the anti-inflammatory action of the foregoing three crude drugs.
