Hyoscyamine and scopolamine in Scopolia extract were determined by gas chromatography. The stationary phase used was 1.5% SE-30 coated on acid-washed and silanized Chromosorb W (60-80 mesh), and the determination was carried out under the chromatographic conditions of 2.5 mm×1 m glass tube, temp of column 200°, injection 230-250°, carrier gas N2 25 ml/min. Hyoscyamine and scopolamine were quantitatively converted into O-trimethylsilyl derivatives using N, O-bis (trimethylsilyl) acetamide in chloroform for gas chromatography at room temperature. Determination was made successfully by the internal standard method using santonin. Results obtained by this method were in good agreement with those of Scopolia extract in Pharmacopeia Japonica (Seventh Edition).
