Pharmacological actions of 3-isopropyl-5, 7-dipiperidinomethyltropolone hydrochloride (IDPT) have been investigated and following results wire obtained. 1) On rat blood pressure, IDPT showed a temporary hypotensive effect and little influenced on the effects of norepinephrine, acetylcholine, and isoproterenol. The hypotensive effect was not affected by the pretreatment with atropine, propranolol, and phentolamine. 2) IDPT decreased the contractile amplitude of heart temporarily and this was recovered to 80-90% of the control level in isolated guinea-pig heart. 3) IDPT caused a remarkable dilatation, in guinea-pig coronary vessel, in accordance with the dose used. 4) IDPT caused a dose-dependent relaxation in isolated rabbit and guinea-pig thoracic aorta strip. 5) IDPT antagonized contraction of intestine induced by histamine, BaCl₂ and a high concentration of acetylcholine. 6) IDPT inhibited spontaneous motility of rabbit intestine and weakly affected relaxation of intestine induced by electrical stimulation to adrenergic nerve fiber. 7) IDPT caused a dilatation of vessel of rabbit ear according to the dose used. 8) In the hypogastric nerve-vas deferens preparation isolated from guinea pig, IDPT had no influence on the hypogastric nerve, but showed inhibition of the vas deferens. 9) IDPT had no influence on the sleeping action induced by hexobarbital. 10) IDPT (50 mg/kg, s.c.) had no effect on normal body temperature. 11) It is concluded that relaxation of smooth muscle by IDPT was due mainly to the papaverine-like action of this compound.
