    Brush border-linked disaccharidases, peptidases, phosphatases and lyases have been investigated in the intestine of rats, mice and guinea-pigs by means of light microscopic histochemistry. Distribution and activity differences exist not only between various parts of the intestine and the villi but also between various cell types of the intestinal epithelium and between various animal species. From these results it becomes obvious that the intestinal brush border, which seems to be a uniform zone from the morphological point of view, in reality represents a highly differentiated structure the functional meaning of which is not yet completely understood.

    The enzymes relating to phosphoric acid metabolism, oxido-reduction system, and carbohydrate metabolism were histochemically demonstrated in the Purkinje cells, RBB, LBB, HIS, and A-V node. Generally, the enzyme activity in the tissue cells of the conduction system was lower than in the tissue cells of the heart muscle. Only the case, phosphorylase, was the activity of the conduction system higher than that of the heart muscle. The polyglucose particles synthesized by phosphorylase and brancher were larger and more numerous under the electronmicroscopic level in the CS. The activities of glucose-phosphate isomerase, phosphoglucomutase, and ribose-5-phosphate isomerase were newly demonstrated in CS and HM. The activities of LDH, G-6-P DH, and R-5-P isomerase in the CS were the same as or slightly lower than their activities in the HM. The activities of G-6-P and SDH in the CS were apparently lower than those in the HM, as previous reports were already indicated. Acid-P and Alk-P activities were very slight or almost negative in both CS and HM. The author presumes that higher phosphorylase activity and considerable ATP, G-6-P DH, and R-5-P isomerase activities in the CS are related to anaerobic metabolism and to the chemical energy from ATP and NADPH.

    ACTH·anti-ACTH immune complex was found to have steroidogenic activity, and to bind with rat adrenal frozen sections in a characteristic manner by means of immunofluorescence technique.
    It specifically bound with the adrenal cortex, and competed with native ACTH, but not with insulin. The adrenal glands from hypophysectomized rats showed stronger affinity to the complex than the untreated. The outer zona fasciculata bound most prominently, and also occasionally the inner zona fasciculata and reticularis, but the zona glomerulosa never bound. The cellular sites of binding were not restricted on the cell membrane, but also cytoplasm of the cortical cells showed positive staining.
    These binding characteristics of ACTH·anti-ACTH complex were discussed, comparing with those of ACTH receptor biochemically established.

    Steroid 3β-ol dehydrogenase activity in some transplantable tumors of mice was reported in the previous paper. In this study, activities of this enzyme and of certain related enzymes were examined histochemically in their host organs.
    Steroid 3β-ol dehydrogenase activity was highly positive in the renal distal convoluted tubular epithelium of mice with Gardner 107 testicular interstitial cell tumor or ovarian granulosa cell tumor, both of which always showed a positive activity of the enzyme. However, the activity was undemonstrable in the kidneys of non-tumor or osteosarcoma-bearing mice.
    ALPase activity did not increase in the kidneys of osteosarcoma-bearing mice, despite a marked elevation of serum ALPase of the host.
    The degree of the activity of steroid 3β-ol DH, of G-6-PDH, of LDH or of ALPase varied in the lungs and livers, but no any difference was found between the activities in the similar organs of mice with or without tumors.

    G6PD and ICD activity within the hepatic lobule was determined quantitatively by the ultramicrochemical method of Lowry in CCl4-injured liver of rats and their controls. The activity of G6PD, ICD and NADPH diaphorase was evaluated simultaneously by two histochemical staining methods: the Meijer method using phenadine methosulfate (PMS) and menadione as intermediate electron acceptors and the Pearse method without using PMS or menadione. By the ultramicrochemical method, the G6PD activity increased remarkably and ICD activity decreased in the central area which was most severely affected in CCl4-injured rats. By the Pearse method, the activities of these three enzyme decreased or diminished in the central area. The Meijer method, however, demonstrated considerable G6PD and ICD activity in the central area. Further examination confirmed that such discrepancies were related to NADPH diaphorase which is generally considered the rate-limiting factor in the staining of NADP-linked dehydrogenases. Increased G6PD activity and considerable ICD activity were detected even in necrotic cells by the ultramicrochemical method and the Meijer method.

    Ultrastructural observations and the histochemical methods for phosphatases (ACPase, ALPase, TPPase, G6Pase, and ATPase), dehydrogenases (SDH, LDH, and G6PDH), and fats were performed on the mammary and adenohypophyseal tissues of adult virgin, diestrous rats, within 48hr after a single administration of Sulpiride and in a week of the daily injection. A rapid release and the following production of secretory granules could be seen in the prolactin cells of the anterior pituitary gland 6-24hr after the injection. Acute hyperactivity of the mammary acini and ductuli could be detected 12-24hr after the single dosage, and a prominent secretion and a marked proliferation could be observed on the mammary gland during the daily injection. Milk congestion in some mammary acini caused finally a pressure atrophy in the corresponding epithelial cells. Both radioimmunoassay for prolactin contents in the serum and the adenohypophysis and radioautography for thymidine uptakes into the mammary and pituitary parenchyma did serve to confirm the “hypothalamic lactation” with the drug.
