Tomaymycin, a new antiphage antibiotic is produced by cultivation of
Streptomyces achromogenes var.
tomaymyceticus in a lactose-bouillon medium enriched with yeast extract and phosphate salts. The active substance can be extracted from the culture filtrate by carbon adsorption and further purified by silica column chromatography. Tomaymycin is obtained in crystalline form as a molecular complex with methanol. The complex occurs as colorless platelets, m.p. 145-146°C, [α]
20D + 423° (
c 0.5, pyridine), λ
max 224, 237, 260, 320 mμ (in methanol). The molecular formula is C
16H
20O
4N
2. The antiphag°e activity is strong against
Escherichia coli T
1, T
3 phages and
Bacillus subtilis M-2, SP-10 phages. Tomaymycin has also antimicrobial activity against Grampositive bacteria. The LD
50 (i.p.) of tomaymycin for mice is estimated at 3.0 mg/kg.

7-O-Demethylcelesticetin is an antibacterial agent produced by
Streptomyces caelestis strain 22227 a, a mutant of
S. caelestis. This antibiotic was found to be as active as celesticetin
in vitro. Treatment of 7-O-demethylcelesticetin with base yielded 7-O-demethyldesalicetin which also has antibacterial properties. Hydrazinolysis of 7-O-demethylcelesticetin gave 2'-hydroxyethyl thiolincosaminide, indicating that
S. caelestis strain 22227a produces lincosamine, the aminosugar present in the lincosaminide antibiotics.

In this study, 58 samples of water and mud were collected at 19 different locations in the Japan Sea, and from these 142 strains of microorganisms were isolated. In all 126 isolates were bacteria and 16 isolates were yeast and fungi. No actinomycetes were isolated. The isolates were placed into 10 groups in accordance with SHEWAN'S scheme. A total of 35 strains (28.5%) of 123 isolates were dependent to sea water, and 25 of these strains (20.3%) showed antimicrobial activities.

In all, 136 strains of actinomycetes were isolated from 37 samples collected at three inlets in Sagami Bay. The salt tolerance of terrestrial actinomycetes was examined. Most of actinomycetes tested were able to grow in media containing 3.0-3.5% NaCl eqivalent to the average salt concentration of sea water, with the exception of two strains of
Microbispora, which failed to grow in media containing less than 2% NaCl. A strain of
Actinomyces aureoverticillus, of
Chainia violens and of
Nocardia asteroides were able to grow in media containing more than 10% NaCl. The production of antibiotic substances by four strains of these isolated actinomycetes was studied in various media with varied temperatures. It was remarkable that the antibacterial spectra changed with varied cultural conditions. In all, 27% of the isolates had antimicrobial activity and 17% showed inhibition against YOSHIDA sarcoma cell.
