    To clarify the mechanism by which β-adrenergic blockade in asthma is induced, the microviscosity of lymphocyte cell membranes in asthmatic patients was investigated by the fluorescence polarization method. For fluorescence hydrocarbon (DPH) was used as a probe for monitor membrane fluidity in the cell membrane lipid layers. For fluorescence labeling, the peripheral lymphocytes were incubated with 2×10^<-6>M DPH for 60 min at 37℃. Fluorescence polarization was measured by a fluorescence spectrophotometer (Hitachi model 1650-40) with 361nm wavelength for excitation and 431nm for emmision. The degree of fluorescence polarization (P), the total intensity (F) and the fluorescence anisotropy (γ) were obtained by measurements of the fluorescence intensities polarized parallel (I_<11>) and perpendicular(I_1). The polarization fluorescence relates to the viscosity of the medium according to the Perrin equation. The fluorescence anisotropy was used as substitute for the viscosity. The fluorescence anisotropy ratio of 22 asthmatic patients was 0.257±0.027(mean±S.D.)and that of 20 healthy subjects was 0.266±0.028. There was no remarkable difference between asthmatic patients and healthy subjects in the degree of fluorescence polarization, the total intensity and the fluorescence anisotropy. These results suggest that β-adrenergic blockade in asthma is not mainly caused by the cell membrane fluidity. It may be caused in the post receptor site of the β-adrenergic system.

    A total of 150 asthmatic children were investigated by prick test for the presence of allergenicities against chironomid antigens. The percentage of cases who showed positive prick tests to the antigens of Polypedilum kyotoense, Chironomus yoshimatsui, Tokunagayusurika akamusi(adlt & larvae)and Dermatophagoides farinae were 22.0%, 16.7%, 8.0%, 14.7%, 94.0%, respectively. In addition, Phadezyme RAST were made with the previously tested 3 midge antigens on a total of 51 asthmatic cases. There were positive-correlations between the results of RAST and those of prick tests. The correlation between the wheal size in the prick test, the threshold dilution in the intradermal test, the RAST score, and the result of the provocation test were examined in 16 asthmatics. In 9 asthmatic children with the positive prick test, the threshold dilutions were all less than 10^<-4>, RAST scores were 2 or above in 6, and the provocation tests were positive in 7. This clearly show that strong positive-correlations exist. The cross reactivity between Dermatophagoides farinae and Polypedilum kyotoense was investigated by RAST inhibition tests. The antigens causing allergic reactions were quite independent of each other. In conclusion, extracts of chironomid midges have strong allergenicities, and constitute one of the more important inhalant antigens together with Dermatophagoides farinae.

    We investigated the effect of sulfuric acid mist on the breathing curve pattern and pulmonary β-adrenergic receptor of guinea-pigs. The guinea-pigs were exposed to sulfuric acid mist (average concentration of 2.49 mg/m^3)for 4 hours at a rate of 6 times a week, totaling 25 exposures. After the guinea-pigs were exposed to mist 16 times repeatedly, they were also exposed to albumin spray solution for 30 minutes at a rate of 3 times a week, totaling 5 exposures. Their breathing curves were recorded with the autorecording bodyplethysmograph system and β-receptor assay was carried out by the ^3H-dihydroalprenolol binding technique. The results were as follows. 1) Exposure to sulfuric acid mist alone did not affect the number and dissociation constant of β-receptors. 2) Albumin inhalation followed by exposure to sulfuric acid mist induced severe dyspneic attacks. Those guinea-pigs which had definitely developed severe dyspneic attacks showed apparant decrease in the number of pulmonary β-receptors, although dissociation constrant did not show any changes.

    Atomospheric pollen grains of Cryptomeria japonica and Cupressaceae(mainly Chamaecyparis obtusa)in Sakado City, Saitam Prefecture were collected by Durham's pollen trap from 1981 to 1985. The relationship between pollen count data and the number of cases of Sugi pollinosis as well as the relationship between pollen count data and meterological factors was studied and the following results were obtained. 1. Dispersion of Cupressaceae pollen occurred of the end of March or in early April and overlapped the end of the dispersal period of Cryptomeria japonica pollen. The pollen count of Cupressaceae in 1985(8862)was about twice that of Cryptomeria japonica(4122). 2. There was a closer correlation between the number of cases of Sugi pollinosis and total count of Cryptomeria japonica and Cupressaceae pollen than between the number of cases of Sugi pollinosis and pollen count of the Cryptomeria japonica only. This result suggests the neccessity of pollen count forecasts for Cupressaceae in addition to pollen count forecasts for Cryptomeria japonica. 3. The total count of Cryptomeria japonica pollen was correlated significantly with the mean humidity and sunshine duration from July 11 to August 10 in the preceding year, whereas the Cupressaceae pollen count tended to be correlated with the mean temperature and sunshine duration in July and August of the preceding year as well as sunshine duration in February and March in the actual year.

    Both culture supernatants of unstimulated lymphocytes and sera from atopic patients were tested for IgE-binding factors (IgE-BFs) displaying the inhibition of rosetting of RPMI 8866 cells expressing Fc receptors for IgE with IgE-coated ox erythrocytes. The culture supernatants of T and B cells as well as the sera of asthmatic patients with elevated serum IgE of more than 2,000IU/ml contained rosette-inhibiting factors. Furthermore, the rosette-inhibiting activity of the samples tended to be higher in the asthmatic patients with elevated serum IgE level. Rosette-inhibiting factors determined in the preparations could be removed by absorption with IgE-Sepharose from which they were subsequently eluated with acid buffer, indicating that the rosette-inhibiting activity was indeed mediated by IgE-BFs. In addition, such IgE-BFs derived from atopic patients had affinity for concanavalin A. Furthermore, rosette-inhibiting factors which had been also binding to serum IgE molecules were detected in the sera from patients with atopic dermatitis.

    Cianidanol, a flavonoid compound, enhanced lymphocyte transformation of peripheral blood mononuclear cells from patients with HBe antigen-positive active hepatitis induced by stimulation with either HBs antigen or purified protein derivative(PPD). This increased response was presumed to be attributable at least partially to the activation of macrophages, since (^3H)-glucosamine uptake into macrophage activating factor(MAF)-activated macrophages and production of interleukin-1(IL-1) were also increased by cianidanol. These results suggest that cianidanol may act on the immune system as an immunopotentiator.
