  • Part I. Substrate Specificity and Transglucosidation Action of an α-Glucosidase of Brewer's Yeast

    The properties of brewer's yeast α-glucosidase have been investigated. The enzyme was capable of hydrolyzing various α-glucosides and was active especially on aryl-α-glucosides in comparison with other α-glucosides and sugars. The rate of hydrolysis decreased in following order: phenyl-α-glucosides, sucrose, matlose and isomaltose.
    The range of opt. temp. was 40-45°C and opt. pH, 6.5-7.0.
    Cu++and Hg++inhibited strongly the enzyme activity and Zn++, moderately. The enzyme was suggested to be a sulfhydryl enzyme from the inhibition experiments by SH-reagents and the effects of glutathione on the activity.
    The enzyme synthesized some oligosaccharides from maltose. As the transglucosidation products, nigerose, isomaltose, kojibiose and maltotriose were detected by paperchromatography.
    Pure nigerosc was separated by splitting maltose with amyloglucosidase from the mixture of maltose and nigerose and by use of successive carbon column chromatography.

    Pure inositol tetra-and hexaphosphate were isolated as the main components of rice phytin by Dowex 1 column chromatography. Phytase of wheat bran was purified more than 1, 500 folds. This phytase preparation showed 1) broad substrate specificity, 2) pH optimum at about 5.0, 3) no activation by magnesium ion, and 4) inhibition by fluoride ion for all the substrates tested; and was concluded as nonspecific acid phosphomonoesterase rather than phytin-specific phosphatase. The effects of metalic ions, thiol reagents, chelating agents, arsenate, alkylarsenate and hydrolysis products for this enzyme were studied. Michaelis constants and activation energy by this enzyme to various substrates were also calculated.

    Cultural conditions for polyalcohol production by Pichia miso were examined in Waldhof type 20 liter-fermentor scale. The best result was obtained under conditions where the aeration rate was 1 volume per volume of the medium per minute with the stirring rate of 500 r.p.m., (Kd=5×10-6 [g-mol of O2/atm. min. ml.]); in 5 days incubation, Pichia miso completely dissimilated the glucose of a high concentration, 30%, and produced glycerol, D-arabitol and erythritol in a very high yield, 50% of sugar consumed. The greatest advantage compared with the shake flask culture is that the required fermentation time is shortened to half.

    Nitrate reductase from the leaves of Burley 21 tobacco (Nicotiana tabacum L.) was partially purified by ammonium sulfate fractionation, protamine sulfate treatment and calcium-phosphate gel adsorption.
    The enzyme has optimum pH at 7.4 and is specific for reduced diphosphopyridine nucleo - tide (DPNH) as the electron donor. The nitrite formed increased in proportion to the rate at which DPNH disappeared in the reaction mixtures. Addition of flavin adenine dinucleotide (FAD) to the assay system enhanced the activity. FAD content in the “highly purified” enzyme was also determined. The enzyme was sensitive to heavy metals and SH-group inhibitors.
    Discussions are presented on the metal and the properties of the enzyme in comparison to those published on other higher plants.

    The growth rate of the α-crystal of L-glutamic acid was measured under various degrees of supersaturation and temperatures. The rate constants and the activation energies for (0 0 1) and (1 1 1) faces were measured and the latter values were 6.7 and 11.5kcal/mol, respectively. The controlling process of the α-crystal growth was investigated by comparison of Sherwood numbers of dissolution and crystallization, and the crystallization process was found to be controlled by the surface reaction.
  • Part III. Influences of Some Factors on the Solubility

    The influences of some factors at the both sides of the solid and liquid phases on the solubilities of L-glutamic acid in a and β-forms in water were examined.
    As differences in the conditions of the liquid phase, the effects of co-existing inorganic neutral salts and amino acids on the solubilities were investigated; and as for the inorganic neutral salts applied here, salting-in effects were recognized, the degrees of which were in the following order, i.e., NH4Cl, (NH4) 2SO4>KCl>NaCl. The effect in the α-form was larger than that of in the β-form. The influence of co-existing amino acids was very little if the amount was not so much, but the tendency to increase the solubility could be recognized.
    As a difference in the condition of the solid phase, the purity of the crystals was picked up, and it was clarified that the crude crystal of L-glutamic acid in α-form gave a larger value of the solubility than the purified one.

    From 2-anisylideneamino-1, 3, 4-tri-O-acetyl-2, 6-dideoxy-6-S-acetyl-6-thio-β-D-glucopyranose, three derivatives of 2-amino-2-deoxy-1, 6-dithio-D-glucose were prepared, i. e., 2-anisylideneamino-3, 4-di-O-acetyl-1, 2, 6-trideoxy-1, 6-di-S-acetyl-1, 6-dithio-β-D-glucopyranose, 2-amino-3, 4-di-O-acetyl-1, 2, 6-trideoxy-1, 6-di-S-acetyl-1, 6-dithio-β-D-glucopyranose hydrochloride and 2-acetamido-3, 4-di-O-acetyl-1, 2, 6-trideoxy-1-mercapto-6-S-acetyl-6-thio-β-D-glucopyranose; and some of their properties were described.

    Some oximes reacted with cyclic phosphorus esters derived from o-hydroxybenzyl alcohol and its analogues to give phenolic and acidic products. The phenolic products were O-o-hydroxybenzyl derivatives of the oximes and the acidic products were phosphoric-oximinic anhydrides. The rate of reaction decreased in the order; phenylphosphonate>phenyl phosphate>alkyl phosphate. Phosphoramide ester was not reactive. Three of eight tested oximes, monoisonitrosoacetone, pyridine-2-aldoxime methiodide and salicylaldoxime, were reactive. The reaction mechanism was discussed.
  • Part VIII. Proteinase Productions of Various Pseudomonas Species, Especially Ps. aeruginosa

    Proteinasc productions of various species of the Pseudomonas genus were comparatively examined. Among them, Ps. aeruginosa was found to be the most suitable species for obtaining a proteinase in a high yield. The proteinase production of the organism was slight in natural medium, but remarkably high in shaking culture of synthetic medium containing glucose in a high concentration and a limited content of organic nitrogen. This medium found to be suitable for the species Ps. aeruginosa, also was remarkably similar with the medium suitable for Ps. myxogenes sp., which had been identified by us and used for this series of studies on the protease of Pseudomonas.
