Purification of β-glucosidase from the seeds of Japanese cycad, and properties of the purified preparation are described. The enzyme activity was determined by colorimetry using ONPG as substrate. Crude preparation was obtained easily by adsorption on fibrous CMC pulp. It was further purified by chromatography on CMC powder, and a preparation which showed an activity of 135-folds of the original extract was obtained. Influences of pH, temperature, and substrate concentration upon the enzyme activity were examined. Michaelis constant of the enzyme for ONPG was 3.3×10
-3M.

Part II. Existence of Free Aglycon of Cycasin in Its Enzymatic Hydrolysis

Some evidences indicating existence of free aglycon of cycasin are described. Cycasin was hydrolysed with purified cycad β-glucosidase, and the freed sugar component and aglycon were analyzed by manometry or quantitative paper chromatography and by spectrophotometry or polarography, respectively. When the hydrolysis was stopped by ice-cooling or byacidification, the reaction mixture showed the characteristic absorption spectrum and polarogram similar tothose of cycasin in spite of the liberation of glucose. However, if the reaction was stopped by heating, the spectrum and polarogram were not observed. This fact was considered to indicate that the free aglycon of cycasin formed in its enzymatic hydrolysis was decomposed by the heat treatment.

The culture filtrate of a strain of
Bacillus subtilis decomposed ribonucleic acid into 5'-nucleotides and into other intermediates which released orthophosphate by an arsenateresistant phosphatase. Under the best conditions examined in these experiments, about 50 per cent of ribonucleic acid was converted into 5'-nucleotides.
The culture filtrate of a strain of
Bacillus brevis showed slight activities of ribonuclease and/or phosphodiesterase which produced 5'-nucleotides from ribonucleic acid, but showed predominant activity of 5'-adenylic acid degrading phosphatase.

The culture filtrate of
Bacillus subtilis decomposed ribonucleic acid into adenosine and orthophosphate without passing through 5'-adenylic acid. It hydrolyzed neither 2'-nor 5'-nucleotides but only 3'-nucleotides into nucleoside and orthophosphate, while the cell suspension hydrolyzed 2'-, 3'-and 5'-nucleotides non-specifically. Thus 3'-nucleotidase was proved to exist in the culture filtrate of
Bacillus subtilis.

When treated with Raney nickel, S-bisbenzoyl derivative of dihydronereistoxin gave N, N-dimethylisopropylamine together with dimethylamine. Supported by other evidences, especially on the presence of 1, 2-dithiolane ring, this led us to postulate that nereistoxin is 4-N, N-dimethylamino-1, 2-dithiolane. The structure was also unequivocally supported by the NMR spectrum. Some additional data on chemical property of nereistoxin and its derivatives were described in succession to the previous paper.

Investigation was made on the mycological and antibacterial properties of a strain No.46408 isolated from a soil sample collected in Shimoueda, Wakayama Prefecture. The strain No.46408 was compared with similar strains,
St. hygroscopicus and
St. halstedii, and it was judged to belong to a new species and therefore named
Streptomyces atratus nov. sp. Also a strain A-165-Z1, which produces ilamycin, was referred to on its mycological properties and assumed to resemble
St. atratus. St. atratus was found to produce new antibiotics, rufomycin A and B, specially active against acid-fast bacteria.

Investigation was made on the isolation procedure, physico-chemical properties and biological activities of antibiotic 46408 produced by
Streptomyces atratus nov. sp. The solvent extract from the whole culture of this strain gave two antibiotic substances. Both are yellow, neutral and sparingly soluble in water and active against acid-fast bacteria, especially
Mycobacterium tuberculosis and
Mycobacterium smegmatis but inactive against most other bacteria, fungi and yeast examined.
From the physico-chemical and microbiological properties they were found to be new substances and named rufomycin A and B, respectively.
Recently Umezawa et al. reported on new antibiotics, ilamycins, active against acidfast bacteria. The properties of rufomycins and ilamycins suggest that they really are very much like. Rufomycin A was nearly as effective as streptomycin in experimental tuberculosis in mice.

Polarographic reduction-wave of vitamin K
3 showed two inflexions in acetonitrile as solvent. The first step inflexion is caused by reduction of quinone to semiquinone stabilized anion free-radical in acetonitrile, and the second by reduction of semiquinone to hydroquinone dianion. With small addition of water, the dismutation of semiquinone is accelerated and the reduction-wave becomes only one step inflexion. The reduction-wave in acetonitrile is controlled by liffusion. The total wave-height is strictly proportional to concentration of vitamin K
3. The measurement is not disturbed by coexistent fatty substances.

Besides O-ethyl-O-(4-nitrophenyl) phenylphosphonothioate, which is utilized in these days, another compound, O-ethyl-O-(4-cyanophenyl) phenylphosphonothioate, proved to have excellent properties as an insecticide.

In our previous paper, it was reported that O, O-dimethyl-O-(4-cyanophenyl) phosphorothioate (S-4084) has only a low toxicity towards mammals though possessing a high activity towards rice stem borers. In this paper, the chemical and biological properties of S-4084 and its phenylphosphonothioate derivative (S-4087) will be reported in detail. It should especially be noted that S-4084 had a higher activity towards rice stem borers than S-4087 by topical or pot test in a room or in a green-house, but by field test S-4087 was stronger than S-4084.
