The production of actinomycin D was examined using
Streptomnyces antibioticus growing on a complex medium based on glucose and casein hydrolyzate. Purification of the medium to remove calcium increased actinomycin production by about one-third and prevented production of a characteristic brown pigment. The sensitivity to magnesium concentrations was less than was reported by Katz
et al.,
11) while similar requirements were found for zinc and iron.

Streptomyces fradiae IFO 3439 elaborated enzymes with macerating activity toward various plant tissues. The optimum pH of the macerating activity was about 8.0 when the crude enzyme preparation acted on disks of potato tuber or pieces of Ganpi (
Wikstroemia sikokiana Fr. et Sav.) bark. Pectolytic activities in this preparation toward free pectin or polygalacturonic acid were considerably lower than those of fungi or bacteria. However, when the crude enzyme preparation acted on native pectin in Ganpi bark, about 90 per cent of the galacturonic acid residues were recovered as the polygalacturonides having a still high degree of polymerization. These results suggested that the crude enzyme of
S. fradiae solubilized Ganpi pectin, degrading it to only a very small extent.

The rapid microbiological method for determination of amino acids was established. It is composed of 3 steps of culture; inoculum culture, intermediate culture, and assay culture. The inoculum culture is the same as that of ordinary method using
Leuc. mesenteroides P-60. For the intermediate culture, which is carried out between the inoculum and assay cultures, the basal medium supplemented with appropriate amount of the amino acid to be determined is employed. The large amount of cells at logarithm phase grown in the intermediate culture are dispersed and used as inoculum for the assay culture. By this technique the assay can be performed by 2.5 to 3.5 hr of assay culture after 2 to 3 hr-intermediate culture.
The technique can be applied to the determination of amino acids in the mixture and the results agree with those obtained by ordinary method.

Photosensitized oxygenation of α-pyran (1) (2, 5, 5, 8a-tetramethyl-6, 7, 8, 8a-tetrahydro-5H-1-benzopyran), in methanol using rose bengal, rapidly formed a stable peroxide (2).
The peroxide (2) gave 6, 6-dimethyl-8-undecene-2, 7, 10-trione (11) at 140°C in xylene, and 8 (or 9)-methoxy or hydroxy derivatives of 6, 6-dimethyl-undecane-2, 7, 10-trione (12a or b) by hydrochloric acid.
These triketones (11 and 12a) were also obtained by a photo-reaction from the peroxide (2).

The effect of thiamine on the D-ribulose production from gluconate by a thiamine-requiring
Corynebacterium species was investigated. The D-ribulose production by the cells previously grown in a thiamine-deficient medium was higher than that by the cells grown in a thiaminerich medium and supplementation of the thiamine-deficient cells with thiamine resulted in a significant depression of the D-ribulose production. Gluconokinase and NADP-linked phosphogluconate dehydrogenase were detected in the cell-free extract of this organism. Oxidation and anaerobic dissimilation of n-ribose 5-phosphate by the cell-free extract of the thiaminedeficient cells are reduced and the addition of thiamine pyrophosphate to the extract enhanced the catabolic activities for D-ribose 5-phosphate. These results suggest that the accumulation of D-ribulose by the thiamine-deficient cells is a consequence of a reduction of transketolase activity.

The washed cells of a gluconate-utilizing
Corynebacterium strain grown in a gluconatexylose medium produced xylitol from D-xylose in the presence of gluconate. The amount of xylitol was progressively increased with increasing gluconate concentration.
An extract of cells grown in the gluconate-xylose medium showed NADPH-dependent D-xylose reductase activity and NADP-dependent 6-phosphogluconate dehydrogenase activity.
These enzymes in the cell-free extract were purified by Sephadex G-100 gel filtration.
The reduction of D-xylose to xylitol was demonstrated by the coupling the D-xylose reductase activity to the 6-phosphogluconate dehydrogenase activity with NADP as a cofactor using the cell-free extract and the fractionated enzymes.

A xylose-utilizing bacterial strain was isolated from soil.
The strain, No. 553, was identified as
Enterobacter liquefaciens from the result of the taxonomical studies. This bacterium grew well on D-xylose as a sole carbon source and accumulated pentitol extracellularly in shaking culture.
Pentitol produced was isolated from the culture broth and identified as xylitol.
The xylitol production reached the maximum after the cessation of the cell growth with a yield of 33.3mg per ml in a medium containing 10% D-xylose as a sole carbon source and no significant decline of the amount of xylitol was observed through the period of the cultivation.

A highly γ-ray resistant bacterium, which has not been described hitherto, has been isolated from water containing mud, fur and moss at a radioactive hot spring, Misasa, Tottori Prefecture, Japan. This bacterium was Gram-positive, non-sporulating, pink to red colored and pleomorphic rod at young stage and predominantly coccoid or small short rod at old. The radiosensitivity of this bacterium was lower than that of well-known bacterium
Micrococcus radiodurans. When its exponentially growing cells were irradiated in buffer solution aerated sufficiently at room temperature, shoulder dose and D

6 rads, respectively. The morphological, cultural and physiological characteristics of the bacterium have been studied. These attributes suggested that the organism was a new species, and the name
Arthrobacter radiotolerans has been assigned with respect to its high radioresistance.

Acid hydrolysis of the antibiotic YA-56 X (Zorbamycin) and Y belonging to the phleomycin-bleomycin group was carried out and the following constitutional amino acids were isolated from the hydrolyzate of YA-56 X: β-Amino-β-(4-amino-6-carboxy-5-methylpyrimidine-2-yl)-propionic acid, β-aminoalanine, L-erythro-β-hydroxyhistidine and 3 unidentified amino acids. Though the former 3 amino acids were known to be constituents of phleomycins and bleomycins, the latter three were not found in phleomycins and bleomycins. YA-56 Y gave one more unidentified amino acid.
Furthermore, isolation of β-alanine and 2-acetylthiazole-4-carboxylic acid from the hydrolyzate indicated the presence of 2-(2-(2-aminoethyl)-Δ
2-thiazoline-4-yl)-thiazole-4-carboxylic acid in YA-56 X and Y as in phleomycins.

From the acid hydrolyzate of the antibiotic YA-56 X (Zorbamycin) and Y belonging to the phleomycin-bleomycin group, 3 unique amino acids were isolated and their structures were determined to be β-hydroxy-L-valine and 2-(3'- amino-tetrahydrofuryl-2'-yl)-propionic acid and an isomer of the latter compound.
Considerations of NMR of the antibiotic YA-56 and of the reaction conditions led us to a view that the latter two new amino acids were not an integral part of the YA-56 molecule but were artifacts derived from 4-amino-3, 6-dihydroxy-2-methylhexanoic acid constituting of YA-56 X and Y.

The mode of action of the toxic protein isolated from wheat on brewing yeast was investigated, and the following results were obtained: (1) The toxin inhibits respiration and fermentation of the yeast, and causes death of the cell in a few min (6min) at a concentration of 4ppm. (2) At a lower concentration (0.4ppm), the toxin inhibits incorporation of sugars without causing death of the cells. (3) Potassium ion, phosphate ion, protein and nucleotides leak from the cell upon treatment with toxin at a lower concentration (0.4ppm). (4) A directly proportional relationship exists between the lowest lethal concentration of the toxin and the yeast cell population. (5) The toxin is adsorbed onto the cell wall and cell membrane.
According to these results, the toxin seems to react with functional site(s) of the cell membrane causing changes in the permeability of the membrane and resulting in cell death.

The susceptibilities of various strains of yeast to a yeast cell wall lytic enzyme produced by
Arthrobacter luteus were examined. Twenty six strains of yeasts, mainly in the genera
Saccharoinyces and
Candida were tested. They were tested after growth attained to the logarithmic or to the resting stage in different media (malt extract medium or
n-paraffin medium) and various culture conditions (shaking or stationary liquid cultures or agar slopes).
The effects of various treatments, such as heating, or treatment with 2-mercaptoethanol or sodium dodecylsulfate on their susceptibility were also examined.
These various conditions and treatments greatly influenced the susceptibilities of the yeast cells, suggesting that they affected the composition and/or structure of the yeast cell walls.

From defatted
n-paraffin-assimilating yeast cells, a crude protein was obtained by alkaliextraction followed by acid-precipitation. Then the protein was treated with ether until extractable substances were removed exhaustively at this stage. However, at the next stage where the ether-treated protein had been partially hydrolyzed with pepsin, when the hydrolysate was retreated with ether, it was found that ether-extractable substances totalling 270mg/100 g were obtainable additionally. Chromatographic investigations demonstrated that the substances included significant amounts of aliphatic and aromatic hydrocarbons, some indoles, and a ubiquinone (
n=8).
From the protein hydrolysate (substrate) after the above ether-treatment, a plastein was synthesized with Bioprase under the specific conditions. The plastein was obtained as a precipitate when the whole reaction mixture was treated with aqueous ethanol or acetone. The quantity and quality (nitrogen content) of the plastein depended on the ethanol or acetone concentration. Roughly speaking, the higher the concentration, the more the plastein quantity. The converse relation held for the quality; a plastein precipitated by treatment solely with water showed a higher quality than any other case.

A mutant of
Streptomyces fradiae which requires oleic acid for neomycin formation was isolated and the effects of exogenous fatty acids and other additives on the formation of neomycin were studied. Palmitic acid and high concentration of sodium ions could replace oleic acid in neomycin formation. The fatty acid spectrum of the mutant strain ST-5B was quite different from that of the parent strain 3123. The major fatty acid components of the mutant and the parent were anteiso 15:0 and iso 16:0, respectively. However the fatty acid composition of the mutant was changed from the anteiso 15:0-type to the parental iso 16:0-type by the supplement of oleic acid or high concentration of sodium ions in the medium. In the case of palmitic acid, the major fatty acid component of the mutant cells was changed from anteriso 15:0 to normal 16:0. The role of these additives in neomycin formation by the mutant is discussed.

Neomycin-producing strains of
Streptomyces fradiae, whose cellular fatty acid spectra are of iso 16:0-type or normal 16:0-type, had about two to six times larger amino acid and hexosamine pools than a neomycin-nonproducing strain which has the anteiso 15:0-type cellular fatty acid spectrum. About 50 to 80 percent of the amount of extractable free amino acids were L-glutamic acid in either type of cells. The difference of pool size in these strains seems to be explained by the difference in ability for amino acid uptake. That is, the ability for L-glutamate uptake of anteiso 15:0-type cells was markedly reduced and accumulated glutamate was easily washed out by buffer. Glucose, magnesium ions and L-glutamate were essential for the formation of neomycin by washed cells and, therefore, even the mutant ST-5B of anteiso 15:0-type could accumulate a large amount of glutamate and produce neomycin as far as it was grown in a medium containing a high concentration of glutamate. These results indicate that a large pool of glutamate is essential for the formation of neomycin and the fatty acid spectrum is a factor governing the capacity to accumulate L-glutamic acid.

Brevibacterium fiavum, prephenate dehydratase in the phenylalanine specific biosynthetic pathway was strongly inhibited by phenylalanine and activated by tyrosine. Furthermore, the inhibition by phenylalanine was completely reversed by tyrosine. Inhibition by tyrosine of prephenate dehydrogenase in the tyrosine specific pathway was very weak. Overall regulation mechanism of the aromatic amino acid biosynthesis in
B. flavum was proposed on the bases of these results and the previous findings on 3-deoxy-D-
arabino-heptulosonate-7-phosphate synthetase(DAHP synthetase
*) of the common pathway and on anthranilate synthetase of the tryptophan specific pathway. Two types of
m-fluorophenylalanine(
mFP) resistant mutants which accumulated phenylalanine alone or both phenylalanine and tyrosine, respectively, were derived. The accumulation in the former mutants was inhibited by tyrosine, but that in the latter was affected neither by tyrosine nor by phenylalanine. DAHP synthetase of the latter mutants had been desensitized from the synergistic feedback inhibition by tyrosine and phenylalanine, while prephenate dehydratase of the former mutants had been desensitized in the feedback inhibition by phenylalanine. Tyrosine auxotroph accumulated phenylalanine under tyrosine limitation and its accumulation was inhibited by the excessive addition of tyrosine. Phenylalanine auxotroph accumulated tyrosine under phenylalanine limitation and its accumulation was inhibited by the excessive addition of phenylalanine. These results
in vivo strongly supported the proposed regulation mechanism in which synthesis of phenylalanine in preference to tyrosine was assumed.

Among about 500 strains tested, a newly isolated soil bacterium,
Brevibacterium sterolicum nov. sp. KY 3463 (ATCC 21387) showed the highest potency in production of 3β-hydroxysteroid oxidase in the culture fluid.
The 3β-hydroxysteroid oxidase was purified from the culture filtrate by a procedure involving ammonium sulfate fractionation, DEAE-cellulose and hydroxyapatite column chromatographies and Sephadex G-75 gel filtration. Crystals of the enzyme were obtained from solutions of the purified preparation by the addition of ammonium sulfate. The crystals appeared as fine rods, with a bright yellow color.
The enzyme is homogeneous by disc gel electrophoresis and ultracentrifugation. Sedi mentation velocity yields a value of
s020, w=3.3S. It exhibits a typical flavoprotein spectrum of absorption maxima at 280, 390, and 470mμ.

Potent bacteria for production of chillproofing enzyme were isolated during screening tests on 1670 strains of microorganisms.
All but one of these bacteria were classified as
Serratia marcescens and the exceptional strain was tentatively designated as B-103. These bacteria produced an extracellular proteolytic enzyme which prevented chill haze of beer.

A bacterium was isolated which produced an extracellular proteolytic enzyme preventing chill haze of beer. This strain was named B-103.
Taxonomical studies on strain B-103 indicated that it was Gram-negative, aerobic, motile, and catalase-positive. Cells were single rods (0.2_??_0.6×1.0_??_1.8μ) with a peritrichous flagella. The bacterium produced nitrite from nitrate, extracellular DNase and 5'-nucleotide from nucleoside. It was capable of nitrate respiration and of oxidative or fermentative cleavage of carbohydrates.
From these characteristics, it was identified as a strain of
Serratia marcescens.

A facile synthetic method of variously substituted α-methylene-δ-valerolactones was devised and the growth inhibiting effects of synthesized samples were tested on rice seedlings. Alkylation of enamines (II) with methyl α-bromomethylacrylate (III) provided key intermediates, unsaturated keto esters (IV) which were readily converted to substituted α-methylene-δ-valerolaetones (V) by a sequence involving saponification, reduction and lactonization. The growth inhibitory activities of the 6-membered α-methylene lactones synthesized closely paralleled that of α-methylene-γ-phenyl-γ-butyrolactone (XIII) which had already been shown in our previous paper to possess the inhibitory activity comparable to that of the natural inhibitors, heliangine and pyrethrosin.

From the methanolysis product of the antibiotic YA-56 X (Zorbamycin) and Y belonging to phleomycin-bleomycin group, two monosaccharides and one disaccharide were isolated as their fully acetylated derivatives. The structures of these compounds were determined to be methyl 2, 3, 4-tri-O-acetyl-6-deoxy-β-L-gulopyranoside, methyl 2, 4, 6-tri-O-acetyl-3-O-carbamoyl-α-D-mannopyranoside and methyl 2-O-(2, 4, 6-tri-O-acetyl-3-O-carbamoyl-a-D-mannopyranosyl)-3, 4-di-O-acetyl-6-deoxy-β-L-gulopyra noside.
Based on these results, it was concluded that 2-O-(3-O-carbamoyl-α-D-mannosyl)-6-deoxy-L-gulose is present as a sugar moiety of the antibiotic YA-56.

In addition to the previously reported six amino acids and two carbohydrates (one disac-charide), 3-aminopropionamidine was isolated as a constituent from the acid hydrolyzate of the antibiotic YA-56 X, one of the major component of antibiotic YA-56 complex. Based on the hitherto found informations, structural comparison of the antibiotic YA-56 X and Y was made with phleomycins, bleomycins, zorbamycin and zorbonomycin B to which antibiotic YA-56 closely related. Among those, zorbamycin was identical with antibiotic YA-56 X.

From the comparisons of mass spectral fragmentations and gas chromatographic retention times with reference compounds, volatile flavor products from UV-photolysis of S-(
cis-1-propenyl)-L-cysteine in oxygen-free aqueous solutions were identical with propanal, 2-methyl2-pentenal,
n-propyl mercaptan, allyl mercaptan, 1-propenyl mercaptan, 2-methylthiophene, 3-methylthiophene, 2, 3-dimethylthiophene, 2, 4-dimethylthiophene, 2, 5-dimethylthiophene, 3, 4-dimethylthiophene,
n-propyl 1-propenyl sulfide and di-l-propenyl sulfide. Moreover, from the comparison of two-dimentional thin-layer chromatograms with reference compounds, ninhydrin-positive products with interest in terms of the degradation mechanism were identical with alanine and cystine. It seems that above-mentioned thiophene derivatives are produced
via 1-propenyl thiyl radicals.

The addition of penicillin to cells of
Corynebacterium alkanolyticum No. 314 growing on
n-paraffins medium caused the simultaneous excretion of phospholipids, UDP-N-acetylhexosamine derivatives and L-glutamic acid.
Among many antibiotics which inhibit cell wall synthesis, only the inhibitors of peptideglycan transpeptidase such as penicillin G and cephaloridine were effective for inducing the excretion of phospholipids, UDP-N-acetylhexosamine derivatives and L-glutamic acid, while the others promoted only the excretion of UDP-N-acetylhexosamine derivatives.
From the close relationship between the excretion of L-glutamic acid and the excretion of phospholipids, it was suggested that the action of penicillins and cephalosporins on the cell membrane resulted in the excretion of L-glutamic acid.

The addition of penicillin (50u/ml) to the cells of
Corynebacterium alkanolyticum No. 314 growing on
n-paraffins medium at the logarithmic growth phase was the most suitable for the extracellular accumulation of L-glutamic acid and the excretion of phospholipids.
The relation between the extracellular accumulation L-glutamic acid and the excretion of phospholipids-in the presence of penicillin was very close and specific. The kinds of phospholipids excreted and their fatty acid components were the same as those of intracellular phospholipids.

N-Acetylhexosamine derivatives, which are intermediates of cell wall synthesis, were detected in UV-absorbing substances excreted by penicillin-treated
Corynebacterium alkanolyticum. Gel filtration, using Sephadex G-25, separated N-acetylhexosamine derivatives to three components, each of which was purified by Dowex 1×2 column and paper chromatographies.
From the analytical studies, N-acetylhexosamine derivatives were found to be composed of UDP-N-acetylmuramic acid-(diaminopimelic acid, glutamic acid, alanine), UDP-N-acetylhexosaminuronic acid and UDP-N-acetylglucosamine.

The structures of pentopyranines A and C have been established as 1-α-(2', 3'-dideoxy-L-arabinopyranosyl)-cytosine and 1-α-(3'-deoxy-L-arabinopyranosyl)-cytosine based on chemical and NMR spectral evidences. These compounds are considered to be shunt pathway products of blasticidin S biosynthesis.
