Melanoidin pigments prepared by heating an aqueous solution of glycine and xylose at 100°C for 2 hr were chromatographically fractionated into 8 color components (designated as P1, P2, P3, P4, P5, P6, P7 and P8, in order of elution with DEAE-cellulose). It is suggested that these color components of melanoidin were produced by oxidation in the following way; P1→P2→P3→P4→P5→P6→P7→P8.
Color components of melanoidin were separated by utilizing their change of elution position on DEAE-cellulose due to oxidation. The color tones of the separated color com ponents became darker and
1%1cm E450 values became higher in the order of elution on DEAF-cellulose. It is concluded that the amount of browning increased and the color tone darkened with increasing oxidation of the color components. According to spectral measurements, elemental analysis and amino acid analysis, all the color components appear to be very similar in chemical structure only differing as regards molecular weight, or degree of polymerization.

Effect of a phenylalanine (Phe)-free diet on body weight, nitrogen balance and plasma free Phe levels were studied with male White Leghorn aged 2 weeks, 6, 12 and 15 months. Effects of feeding an isoleucine (Ile)- or amino acid-free diet and fasting on plasma amino acid levels in 15-month-old roosters were also studied.
Two-week-old chicks lost appetite for the Phe-free diet and their body weights decreased gradually. Six-month-old cockerels consumed the Phe-free diet completely but could not maintain normal growth rate and positive nitrogen balances. Twelve- and 15-month-old roosters maintained their body weights and nitrogen equilibrium on the Phe-free diet.
Plasma Phe levels of 2-week- and 6-month-old fowls at the 6th hr after the last feeding of the Phe-free diet did not decrease significantly compared to those of the control groups. However, plasma Phe levels of 12- and 15-month-old roosters decreased rapidly within a day on the Phe-free diet. Fasting resulted in a significant increase in plasma amino acid concentrations whereas feeding an Ile- or amino acid-free diet resulted in concentrations of plasma amino acids which were not lower than those observed in the control roosters.

Effect of feeding 4.23, 16.94 and 27.53mg of retinol daily for 10 days on the liver lipids of adult rats has been studied. Feeding of different amounts of retinol produced dose dependent toxicity symptoms in rats. Retinol feeding resulted in significant elevations of liver total lipids, total fatty acids, and glycerides. The amounts of liver esterified cholesterol were significantly raised in rats fed different amounts of retinol. Acetate-1-
14C incorporation was increased in liver total cholesterol of rats fed 27.53mg retinol and in free cholesterol of all retinol fed rats. Total
14C activity of hepatic triglycerides of retinol fed rats was the same as that of control, but their specific activity was decreased. Significant alterations were noted in phosphatidyl serine, lysophosphatidyl choline, lysophosphatidyl ethanolamine, sphingomyelin, phosphatidyl choline, phosphatidyl ethanolamine and phosphatidic acid and polyglycerophosphate fractions in liver rats fed different amounts of retinol.

Growing rats fed a protein deficient diet showed an increase in acid and alkaline protease activity and a decrease in protease inhibitor activity. Similar results were found after two and four day fasting periods. Animals on diets of 10% and 20% casein showed parallel protease and protease inhibitor activity when compared to each other at similar body weight periods.

The rates of recycling and turnover of glucose in 5-day- and 21-day-old lambs and adult sheep were measured by the method of single simultaneous injection of glucose-6-
3H and glucose-6-
14C. The
3H/
14C ratio decreased linearly with time and was 0.58 and 0.60 in lambs of 5-day- and 21-day-old, respectively, and 0.82 in adult sheep at 120min after injection of the labeled glucose. The pool size and turnover rate of glucose considerably decreased with age. The rate of glucose recycling was significantly higher in lambs of both ages (22.0 and 26.2%, respectively) than in adult sheep (11.1%).

D-Arabinose(L-fucose) isomerase (D-arabinose ketol-isomerase, EC 5. 3. 1. 3) was purified from the extracts of D-arabinose-grown cells of
Aerobacter aerogenes, strain M-7 by the procedure of repeated fractional precipitation with polyethylene glycol 6000 and isolating the crystalline state. The crystalline enzyme was homogeneous in ultracentrifugal analysis and polyacrylamide gel electrophoresis. Sedimentation constant obtained was 15.4s and the molecular weight was estimated as being approximately 2.5×10
5 by gel filtration on Sephadex G-200.
Optimum pH for isomerization of D-arabinose and of L-fucose was identical at pH 9.3, and the Michaelis constants were 51 mM for L-fucose and 160mM for D-arabinose. Both of these activities decreased at the same rate with thermal inactivation at 45 and 50°C. All four pentitols inhibited two pentose isomerase activities competitively with same
Ki values: 1.3-1.5mM for D-arabitol, 2.2-2.7mM for ribitol, 2.9-3.2mM for L-arabitol, and 10-10.5mM for xylitol. It is confirmed that the single enzyme is responsible for the isomerization of D-arabinose and L-fucose.

Corynebacterium cultures, isolated from soil by the use of
n-paraffin as the sole carbon source have been shown to bring about significant oxidation of a new compound, dl-cis tetrahydro-2-oxo-4-
n-pentyl-thieno-(3, 4-d)-imidazoline (
dl-TOPTI), in the presence of hydro-carbon.
The co-oxidation products were isolated from culture broth and identified as
dl-biotinol and
dl-biotin by IR, NMR and MS spectroscopies.

The systematic identification and determination of many kinds of pesticides were tried by a combination of column, thin-layer and gas chromatography. Pesticides in the first division,
1) e. g., aldrin, DMC ethylene, CPAS, DDDS,
o, p'-DDT,
p, p'-DDT, heptachlor, quintozene, Telodrin and tetrasul, were separated from pesticides in the second division,
1) e. g., α-BHC, β-BHC, γ-BHC, trifluralin, CPA, CNP, nitrofen, endrin, dieldrin, dicofol,
etc. by column chromatography. Further, pesticides in the first division were separated from each other and determined approximately by thin-layer and gas chromatography.
The recoveries of these pesticides, except for aldrin and CPAS, were about 100%. The recoveries of aldrin and CPAS were about 70%, due to decomposition during the procedures, or to adsorption on the adsorbent during the column chromatography. It was presumed from GC-MS data that CPAS was decomposed to DDDS, tetrasul and other compounds during the procedures.

This experiment was conducted to confirm the reason why roosters adapt themselves to a phenylalanine(Phe)-free diet. The results that recovery % of
14C of injected DL-Phe-2-
14C in expired air reduced drastically for the Phe-free diet and the ratio of specific activity of tyrosine (Tyr) to that of Phe in tissue proteins was also much lower than that for the control diet possibly indicate that the pathway from Phe to Tyr is blocked to a larger extent for adaptation to the Phe-free diet in adult roosters.

Extracellular accumulation of high molecular weight DNA was further studied using
Pseudomonas species. More efficient production was obtained by the use of glucose-grown seed culture and by controlling the broth-pH at around 6.0 for first 24hr and then around 8.0 during the fermentation. The maximum yield was 5 to 6g per liter of the broth culture, which corresponded to 10-fold of that reported in the previous work.
Purified DNA (4×10
6 daltons) was obtained successfully by applying an aqueous biphase system of dextran-polyethyleneglycol and dextranase.
Significant release of DNA occurred only with cell lysis of
n-paraffin-grown bacteria. The primary cause of rapid lysis was explained by the exhaustion of cellular glucose pool. Relation of DNA accumulation to the effect of rhamnolipids on cell membrane was also investigated.

Methylenedioxyphenyl derivatives and the compounds structurally related to piperonyl butoxide, which has been found to be a potent protectant against ozone injury to plants, were tested for effectiveness against the same injury to tobacco leaves. Five of 90 materials tested, namely 3, 4-methylenedioxyphthaldehyde, benzimidazole, safroxane, xanthone and piperonal, showed high protective activity though the effectiveness was inferior to piperonyl butoxide. The protective mechanism was discussed.

Visible and chemical changes of sterilized concentrated skim milk prepared laboratorially were studied over a long period of storage, and on the basis of the results the changes of the calcium-caseinate-phosphate complex were discussed.
The results obtained are summarized as follows. (1) The formation of visible sediment and “whey off” were observed on the 50th day of storage. The increment of the viscosity was not so large during storage. (2) The remarkable increase of the soluble casein was observed, and its proportion to total casein amounted to about 2/3 on the 120th day of storage. Only small amount of calcium bound to the soluble casein. (3) Each amount of calcium and inorganic phosphorus in the ultracentrifugal whey decreased gradually, and that of magnesium increased during storage. (4) The ratios of Ca/N and P/N in the casein complex increased extremely after storage.

The sol of konjac mannan (KM) was gelatinized with an alkali such as sodium carbonate. The turbidity and the viscosity of sol, the infrared spectra of KM, and the consumption of alkali by KM in the course of gelation were measured. Then, experiments were undertaken in order to elucidate the major role of alkalies in gelation and the mechanism of gel-formation. It was presumed that the alkalies eliminate a moiety containing C=O group (probably an organic acid) from KM, and then the molecules of KM which lost the moiety crystallize in part through a linkage such as hydrogen bonding, and a network structure is formed.

Carboxylic acids found in the cultured broth of Sporobolomyces odorus AHU 3246 which produces γ-lactones as principles of the aromatic flavor, were analyzed. The concentrate of methylated acids was steam-distilled and in the residue, succinic acid, nonanedioic acid (azelaic acid), undecanedioic acid and 2-hydroxy-3-phenylpropionic acid (β-phenyllactic acid) were identified as their methyl esters by GLC and spectroscopic methods. Phthalic acid and its mono-
n-butyl ester were also found, but these compounds were thought to arise from di-
n-butyl phthalate, one of impurities of deionized water.

An enzyme which degrades yeast glucan and yeast cells in the logarithmic phase of growth (log yeast cells) and produces protoplasts from log yeast cells has been crystallized from the culture filtrate of a strain belonging to Fungi Imperfecti.
Analyses by ultracentrifugation and disc gel electrophoresis showed the crystalline enzyme to be homogeneous. Its molecular weight was found to be 24, 500. The hydrolysis of laminarin, pachyman and yeast glucan was catalysed by the enzyme to produce a mixture of laminaridextrins. The conversion of log yeast cells to protoplasts was obtained by the addition of only this enzyme, the addition of mercaptoethanol or phosphomannanase to the enzyme promoted the conversion.

Condensation of 4'-benzylapigenin with α-acetobromorutinose in the presense of silver carbonate or potassium hydroxide followed by deacetylation afforded 4'-benzylapigenin-7-β-rutinoside (VI). Debenzylation of VI resulted in the formation of Isorhoifolin (α-form) (I). I was isolated from the leaves of Tengu orange.

Infrared spectra of film specimens of the cell wall of
Nitella were recorded in the untreated state, after acid treatment, and after treatment for removal of pectic substances and hemicellulose. Assignment of the bands in the spectrum of the wall was made. Polarization measurements on the wall indicate that in addition to cellulose, carboxylate ions, which are attributable to pectic substances, are oriented in the wall. The nature of the bonds holding the oriented carboxylate ions is described.

Crystalline β-glucanase able to disrupt the cells of living yeast was studied from enzymic, structural and immunological view points. The enzyme degraded yeast glucan and laminarin rapidly, but it produced only very little reducing sugar and, hence, was supposed to be endo β-1, 3-glucanase of a “more random” type, such as the C
1, component of cellulase. The enzyme did not act on oligosaccharides composed of 2 to 9 glucose units.
Serological differentiation among some yeast cell lytic enzymes using γ-immunoglobulin against the crystalline enzyme revealed the absence of an immunologically common structural unit among them. C-Terminal sequence of -Asp-Ala-Gly-Ser was established for the enzyme protein which was composed of a single polypeptide with 196 residues of 17 amino acids.

Purple nonsulfur bacteria,
Rhodospirillum rubrum and
Rhodopseudomonas spheroides were found to possess coenzyme B
12-dependent glutamate mutase activity. Cell-free extracts of these bacteria grown on Co
2+-containing media catalyzed the conversion of glutamate to β-methylaspartate and further to mesaconate. The activity of the cell-free extracts of these organisms cultivated on Co
2+-deficient media was markedly lower than that of the normal cells. Addition of coenzyme B
12 to the former reaction mixture enhanced the mesaconate formation
via β-methylaspartate. These results indicate the involvement of coenzyme B
12-dependent glutamate mutase of these bacteria in the dissimilation of glutamate to acetyl-CoA and pyruvate through the following pathway.
glutamate→β-methylaspartate→mesaconate→citramalate→→acetyl-CoA, pyruvate
On the other hand, a greater part of glutamate was converted to α-hydroxyglutarate and succinate with the cell-free extracts of these photosynthetic bacteria. This fact, taking account of the presence of propionyl-CoA carboxylase in these bacteria, implies the participation of coenzyme B
12-dependent (R)-methylmalonyl-CoA mutase in the formation of succinate
via the following route.
glutamate→α-ketoglutarate→α-hydroxyglutarate→propionate→propionyl-CoA→
(S)-methylmalonyl-CoA→(R)-methylmalonyl-CoA→succinyl-CoA

Diethylpyrocarbonate (DEPC) exhibited a bacteriostatic effect at concentrations between 0.5 mM and 8 mM and a bactericidal effect at concentrations above 10 mM on
Lactobacillus casei S-1. At the bacteriostatic concentrations the bacteria began to grow after the added DEPC had disappeared from the medium by self-decomposition, whereas at the bactericidal concentrations such growth was not observed even after long-term incubation.
DEPC inactivated free phage J1 within several min; 10, 20 and 30mM of DEPC inactivated the phage by 90, 99.9 and 100%, respectively. The inactivation by DEPC was enhanced slightly by high temperature and markedly by low pH. Metal ions and chelating agents rendered negligible effects on the inactivation percentage.
DEPC did not cause strand scissions in the phage DNA, as exhibited by sucrose gradient centrifugation analysis. It changed the serum-blocking power of the phage, and this change was thought to be due to the structural alteration of phage tail-proteins.
The results are discussed in relation to the elimination of active phages from the dairy cultures.

A bitter peptide has been isolated from a bitter Cheddar cheese by chromatography and high voltage electrophoresis. Its molecular weight was approximately 2500 and it originated from β-casein, its N-terminus being residue 46 (glutamine) of that protein. The glutamine cyclised readily to pyrrolidone-carboxylic acid in neutral solutions of ionic strength 0.1 or less, and this change was accompanied by a loss of bitter taste.

An ultracentrifugally homogeneous preparation of the cyclodextrin glycosyltransferase was obtained from the culture broth of a strain of
Bacillus species (No. 5 strain) by a purification methods including adsorption on corn starch, ammonium sulfate precipitation, chromatography on DEAF-cellulose column, and gel-filtration on Sephadex G-100. This enzyme preparation (No. 5 enzyme) was separated into two fractions in disc-electrophoresis and Ampholine-electrophoresis (Fr. 1: pI 6.07 and Fr. 2: pI 6.80) which had the ability to form cyclodextrin. These two fractions were similar to each other in the enzymic properties except the isoelectric point. These enzymes differ from the amylase from
Bacillus macerans (BMA) in the enzymic properties. Furthermore, No. 5 enzymes differ from BMA in the action on starch. The authors consider that No. 5 enzymes mainly produce the cycloheptaamylose from starch and on the contrary, BMA mainly produces the cyclohexaamylose from starch.

Effects of pH, sodium chloride, temperature, and particle size were studied on the adsorption of proteins onto air bubbles. The results suggested that the froth flotation could be applied to the recovery of dissolved proteins with a comparable efficiency to the active sludge method. Such a strategy was proposed as to strip the dissolved hemoglobin under alkaline conditions or in the presence of sodium chloride, and to concentrate it under acidic conditions. Temperature was of little effect. Flotation of the precipitated proteins was much more efficient than that of the dissolved proteins. Particle s_??_ze greatly affected the efficiency of flotation of the precipitated proteins.

As a result of screening of
n-alkylbenzenes assimilating yeasts, it was shown that the yeasts which grew well on
n-alkane (C
15) showed also good growth on
n-alkylbenzenes (from C
7 to C
19 of side chain). Among four
Candida strains selected,
C. tropicalis S131Y1 produced 4-(
o-hydroxyphenyl)-butyric acid,
o-hydroxyphenylacetic acid and phenylacetic acid from
n-alkylbenzenes with even-carbon side chain and cinnamic acid from
n-alkylbenzenes with odd-carbon side chain.
On the other hand, another three strains produced only phenylacetic acid from
n-alkyl-benzenes with even-carbon side chain. In addition, as for the products from
n-alkylbenzenes with odd-carbon side chain, two of three strains,
C. parapsilosis IFO-1068 and
C. hydrocarbofumarica Et 15-2 produced benzoic acid.
From these oxidation products and oxygen uptake experiment, a new metabolic pathway of
n-alkylbenzenes was assumed.

An unidentified bacterial strain S107B1, isolated from soil by use of isopropylbenzene as a carbon source, was shown to bring about oxidation of α-methylstyrene and β-methylstyrene.
One of the oxidation products produced from α-methylstyrene was identified as the new compound, (-)-
cis-2, 3-dihydroxy-1-isopropenyl-6-cyclohexene.
The same strain S107B1 also oxidized β-methylstyrene and produced 3-phenylpropionaldehyde and benzoic acid.
From these results, the existence of reductive step for the aerobic degradation of these aromatic hydrocarbons by this strain was made clear. The initial attack on these aromatic hydrocarbons and a cyclohexenediol compound formed from a-methylstyrene were discussed.

An extracellular acidic polysaccharide produced by
Serratia piscatorum IFO 12527 was found to exhibit a marked anti inflammatory activity. The polysaccharide was purified by fractional precipitation with cetyltrimethylammonium bromide and then by gel filtration on Sepharose 2B to give two homogeneous fractions, PLS N-I and PLS N-II, the former exhibiting the anti inflammatory activity.
PLS N-I was a complex polysaccharide composed of L-rhamnose, D-galactose and D-galacturonic acid in the molar ratio of 2:1:1, together with small portions of D-glucosamine, D-galaetosamine, protein and fatty acids such as acetic, lauric, myristic, β-hydroxymyristic and palmitic acids. Physicochemical and biological properties of PLS N-I and PLS N-II were also described.

Bacterial production of 4-hydroxy(3, 4-
d)pyrazolopyrimidine riboside (AP-R) was studied. Seven among 73 tested strains were found to produce AP-R through N-ribosyl transfer reaction between uridine and 4-hydroxy(3, 4-
d)pyrazolopyrimidine (allopurinol). AP-R was produced by the cell-free extract of
Erwinia carotovora and was isolated in crystals from the reaction mixture. The crystalline AP-R was characterized by spectroscopic data and was confirmed to be β-1-ribosyl allopurinol. AP-R could not substitute for inosine to support the growth of a nonexacting purine base-requiring mutant of
Sarcina lutea.

A new synthetic procedure for dienone derivatives,
i.e., ionone and irone, is described. The key step is the pyrolytic rearrangement of allyl alcohols and 2, 2-dimethoxypropane in the presence of an acid anhydride and phosphoric acid.

α-Glucosidase has been isolated from
Bacillus cereus in ultracentrifugally and electrophoretically homogeneous form, and its properties have been investigated. The enzyme has a sedimentation constant of 1.4S and a molecular weight of 12, 000. The highly purified enzyme splits α-D-(1→4)-glucosidic linkages in maltose, maltotriose, and phenyl α-maltoside, but shows little or no activity toward polysaccharides, such as amylose, amylopectin, glycogen and soluble starch. The enzyme has α-glucosyltransferase activity, the main transfer product from maltose being maltotriose. The enzyme can also catalyze the transfer of α-glucosyl residue from maltose to riboflavin. On the basis of inhibition studies with diazonium-1-Htetrazole, rose bengal and
p-chloromercuribenzoate, it is assumed that the enzyme contains both histidine and cysteine residues in the active center.

Pseudomonas melanogenum ATCC 17806 required methionine, cysteine, cystine, cystathionine, homocysteine or homocystine for growth. However, the addition of these amino acids decreased remarkably L-DOPA (3, 4-dihydroxyphenyl-L-alanine) production by the bacterium. L-DOPA production by the bacterium was further affected by the amount of the substrate, the method of its addition and by the addition of antioxidants, as was the case with
Vibrio tyrosinaticus.
Under suitable conditions about 8mg/ml of L-DOPA were produced from 8.6mg/ml of L-tyrosine.
