The extracellular galactomannan of
Neurospora crassa has been characterized. The purified polysaccharide preparation consisted of more than 98% carbohydrate (D-mannose, 49%; D-galactose, 43%; D-glucose, 8%) and trace amounts of protein (<1%) and phosphate (<1%). The native polymer ([α]
D, + 28°) was resistant to bacterial exo-α-D-mannanase, while the mannan ([α]D, + 85.9°) after mild acid hydrolysis was partially hydrolyzed by the enzyme. Methylation analysis, Smith degradation, acetolysis, and enzymic and acid hydrolysis indicated that the polysaccharide had a linear α-mannan backbone consisting of α-1, 2-tetrasaccharide, α-1, 2- and α-1, 3-disaccharide units which might be joined by α-1, 6-bonds. Galactose residues linked through a β-1, 5-bond were located exclusively in the side-chains. The side-chains were attached to the mannan backbone by a 1, 2- or 1, 6-linkage.

The amino acid compositions of alginate lyases, SP1 and SP2 were slightly different from each other. They contained about five mol of sugar per molecular weight (25, 000). The reaction with DTNB or PDS suggested that the enzymes contained one sulfhydryl group buried in the molecule and two disulfide bonds. It was obvious from the NBS-oxidation of the enzymes and the protection by the substrate of SP1 that one residue out of 11 Trp residues for SP1 and of 9 Trp residues for SP2 was essential for the activity. Treatment with TNBS rendered the enzymes inactive when 7 of 13 Lys residues for SP1 and 4 of 13 Lys residues for SP2 were trinitrophenylated in 20mM phosphate buffer, pH 8.0. However, kinetic analysis showed that one Lys residue in the active unit was possibly involved in enzyme function.

The absorption efficiency and biological half-life of individual chlorobiphenyls in Kanechlor products were determined in carp (
Cyprinus carpio) after acute dietary exposure. The absorption efficiences of individual chlorobiphenyls ranged from 67 to 93% and decreased as the number of chlorine atoms substituted in biphenyls increased. The metabolized PCBs in carp were confined to lower chlorine substitutes, and they had more chlorine atoms substituted in
ortho positions.
The absorption efficiences of PCBs by carp were nearly the same as the rat data reported earlier. However, the biological half-lives of PCBs in carp were much longer than those of rats. These results suggest that the absorption mechanisms through digestive tract are not dependent on specific physiological differences in vertebrates but on the physicochemical properties of individual chlorobiphenyls. On the other hand, the elimination of PCBs seems to be strongly dependent on the metabolic capacity of the species.
The absorption efficiency and biological half-life of individual chlorobiphenyls appear to provide useful information on the behavior and fate of PCBs in the ecosystems.

Twelve strains of lactose-fermenting yeast isolated from raw milk were evaluated on β-galactosidase producing ability. The enzymes from the four strains (
Tolulopsis versatilis M6,
Tolulopsis sphaerica J28,
Candida pseudotropicalis B57 and A60), selected by high productivity, showed very similar properties and were characterized by a pH optimum of 7.0 or 7.5 and a relatively low optimal temperature of 30°C. The molecular weights were estimated by gel filtration to be 200, 000 - 233, 000. The Km values for ο-nitrophenyl-β-D-galactopyranoside were 3.45 HIM, 2.09 mm, 3.45 mM and 2.82mM for enzymes from M6, J28, B57 and A60, respectively. All enzymes were activated by Mn
2+ and inhibited by Mg
2+, Zn
2+ and Ca
2+. The enzymes are sulfhydryl dependent and were completely inhibited by Hg
2+ and sulfhydryl reagents. The yeasts may be a potential source for the enzyme for industrial use.

Measuring the impedance was found to be a highly reliable and practical technique for identifying irradiated potatoes. Impedance was measured by puncturing a potato tuber with a steel electrode and passing a 3 - 5 mA alternating current through it. Three parameters were determined: Z

5k was the most favourable index. The technique allowed not only differentiation between unirradiated and irradiated potatoes but an estimation of the irradiation dose for up to six months after irradiation, independent of the potato storage condition.

A new type of lysine derivative connected with dicarboxylic acid was synthesized. The acid was coupled with ε-benzyloxycarbonyl-lysine ethyl ester (Lys(Z)-OEt) by conventional methods of peptide synthesis. The removal of the Z-group from the protected compounds by hydrogenation gave dihydrochlorides of the final products: succinyl-Lys-OEt, glutaryl-Lys-OEt, adipoyl-Lys-OEt, pimeloyl-Lys-OEt, suberoyl-Lys-OEt, azelaoyl-Lys-OEt and sebacoyl-Lys-OEt. Among the eight products, azelaoyl-Lys-OEt dihydrochloride showed the highest inactivating effect on several phages.

A strong Cotton effect, which practically govern the sign of the optical rotation at 589 nm ([M]
D), was studied in phenyl 1-thio-α (and β)-D-glycopyranosides with our new chiroptical technique. The proposal optical rotatory dispersion (ORD) method, with calculations based on a one term Drude equation, showed the presence of a strong Cotton effect at 200-210nm. Circular dichroism (CD), with accumulation technique, also gave the same Cotton effect. Agreement in these two methods suggests the usefulness of the proposed ORD calculation method. The rotational strengths and the signs were shown to reflect the anomeric configurations and conformations (α-anomer gave positive and β-anomer gave negative signs; axial gave strong and equatorial gave weak bands). This result is an extension of the ring oxygen helicity rule of alkyl and alkyl thioglycosides to phenyl 1-thioglycopyranosides, and probably to other aromatic glycosides.

The Cotton effect below 200 nm was first studied in aromatic glycopyranosides with our new technique of optical rotatory dispersion (ORD). The phenyl and
p-nitrophenyl α- and β-D-glycopyranosides showed a strong Cotton effect at around 180nm which practically governed the optical rotations in the visible wavelength region. The rotational strength and sign of this Cotton effect were shown to reflect the anomeric configurations and conformations, which led to an extension of the ring oxygen helicity rule for alkyl and alkyl 1-thioglycosides to aromatic glycopyranosides.

Outdoor batch cultures of
Rhodopseudomonas sphaeroides B5 were carried out from April 20 to May 15, 1981. The species was isolated in the Bangkok area. One culture (culture A) was placed vertically to the ground and the other (culture B) was inclined at 30° to receive more sunlight. In each culture, 33 liters of culture medium was used which contained 53mM DL-lactate and 5 mM L-glutamate, respectively, as electron donor and nitrogen source. During the cultural period of 25 days, total hydrogen production was 154.7 liters in culture A and 177.2 liters in culture B. These values corresponded to 69.1% and 78.0% of the theoretical value (6 mol of hydrogen formed per mol of lactate), respectively. The maximum rates of hydrogen production per hour were 2.85 liters in culture A and 4.11 liters in culture B. These results indicated that illumination was the limiting factor in photoproduction of hydrogen under these conditions. The results also suggested the importance of the culture medium temperature in hydrogen production.

The distribution of multiple forms of β-amylase in some varieties or species of soybean seeds was examined by the gel isoelectric focusing method. Seven components (1', 1, 2, 3, 4, 5 and 6) were found. Their respective isoelectric points were 5.07, 5.15, 5.25, 5.40, 5.55, 5.70 and 5.93±0.04. The varieties or species of soybean seeds were separated into two types by their zymograph: the low pI type and high pI type. Component 6 was purified from commercial defatted soybean meal containing all seven components by ion-exchange column chromatography and by gel filtration, and compared with previously purified components 2 and 4. Components 2, 4 and 6 had the same molecular weight and immunological properties but some differences were found in their amino acid compositions and enzymatic properties. The C-terminal amino acid of components 2 and 6 was glycine but that of component 4 was alanine. It was concluded from these results that differences between components 2, 4 and 6 were caused by charged amino acid substitution.

Fermentative production of pyruvic acid by yeasts was studied using extracts from citrus natsudaidai peel as a carbon source. Many yeasts showed good growth. Of these yeasts,
Debaryomyces coudertii IFO 1381 produced pyruvic acid at high yield. Pretreatment of the peel extract with Amberlite IR-120B (Na
+) led to increased production of pyruvic acid. Under optimum conditions, the accumulation of pyruvic acid reached a maximum of 970mg/100ml at 48hr-fermentation. The pyruvic acid from the fermentation broth was identified with lactic acid dehydrogenase and by comparisons of properties of its 2, 4-dinitrophenylhydrazone with those of authentic pyruvic acid in paper chromatography, IR spectrometry and elemental analysis.

Zymolyase was purified from Zymolyase-60, 000 by ion-exchange chromatography with sugar and gel filtration. Zymolyase was separated into the two protein fractions A and B. Neither alone could lyse yeast cells, but together showed high lytic activity. Zymolyase A and B were β-1, 3-glucanase and alkaline protease, respectively. On stepwise treatment of yeast cells with the enzymes, yeast cells were lysed only by treatment with Zymolyase A after pretreatment with Zymolyase B. Zymolyase A lysed yeast cells in the presence of 2-mercaptoethanol, but B could not even at high concentration. Zymolyase B decreased the turbidity of a yeast cell suspension by about 13%.

The organic solvent extract of Kumazasa leaves (
Sasa albo-marginata) showed antimicrobial activity against bacteria, fungi and yeast. Kumazasa at a concentration of 0.2-1.0% showed stronger antimicrobial activity than potassium sorbate or sodium benzoate at the same concentration. Both acidic and phenolic fractions of the extract showed strong antimicrobial activity. Thirty acidic and phenolic compounds were identified by GC and GC-MS analysis. Acetic, propionic, benzoic, phenylacetic, salicylic, 3-hydroxybenzoic and
o-anisic acids, and guaiacol, phenol, 4-ethylphenol, xylenol and 4-vinylphenol were the main components. It was estimated that these components play an important role in the formation of the antimicrobial activity of Kumazasa extract.

Casein and egg albumin were allowed to react with methyl linoleate (ML) at a relative humidity (RH) of 0% or 80% at 50°C for 10 days (protein : ML=1:0.2 or 1:1, w/w). Changes in the molecular sizes of the reacting proteins were examined by gel filtration and gel electrophoresis. Both proteins showed similar changes, whereas the reaction at RH 80% (protein: ML=1:1) resulted in insolubilization because of polymerization. Changes in the amino acid residues of the reacting proteins were investigated after acid (6 N HCl) and enzymatic (pepsin-pancreatin, followed by aminopeptidase-prolidase) hydrolyses. Insignificant changes were observed in the amino acid composition of proteins reacted at RH 0%. After reaction at RH 80% (protein: ML=1:1), Lys, His and Met were the only amino acids affected. The percentage loss of these amino acids after acid hydrolysis was Lys (22%), His (41%), Met (9%) for casein and Lys (22%), His (31%), Met (1%) for egg albumin. This percentage loss after enzymatic hydrolysis was Lys (41%), His (49%), Met (94%) for casein and Lys (37%), His (42%), Met (88%) for egg albumin. Some differences between our results and other researchers were also discussed.

Lentinus edodes, and the structure was confirmed by enzymatic synthesis. The presence of lactam of saccharopine and γ-glutamylglutamic acid was demonstrated in the fraction.

Diphenyl,
o-phenylphenol and thiabendazole were analyzed in citrus fruits. The peel and edible parts were separately homogenized. These fungicides were extracted with dichloromethane from the homogenate, and they were fractionated with Sephadex LH-20 columns. Gas chromatography was used to determine the presence of these fungicides. The fungicides found in edible parts of citrus fruits were confirmed by gas chromatography-mass spectrometry.
Diphenyl,
o-phenylphenol and thiabendazole were detected in imported grapefruits, lemons and oranges. Almost all fungicides were found in the peel. The concentrations of the three fungicides in the edible parts were very low. Some samples contained all three fungicides in the edible parts.

Rats fed a low protein diet containing 3% or more of tyrosine developed external pathological lesions, accompanied by marked depression in growth and food intake. Their liver tyrosine (Tyr) transaminase activity increased in proportion to dietary tyrosine level, but
p-hydroxyphenylpyruvic acid (pHPP) hydroxylase activity was not elevated. The excreted amounts of tyrosine and
pHPP in urine were increased markedly, and the tyrosine concentration in plasma and tissues was markedly elevated. There was a correlation between free tyrosine concentration in body fluid and the development of pathological lesions.
When rats were fed a low protein diet containing graded levels of
pHPP up to 5%, they did not develop external lesions, but their growth and food intake were depressed more severely than in rats fed the same levels of tyrosine. Neither Tyr transaminase nor
pHPP hydroxylase activity in liver was increased, and tyrosine and
pHPP concentrations in plasma and tissues were not elevated by excessive intake of
pHPP. From these results, it is concluded that
pHPP was not responsible for the development of eye and paw lesions characteristic of tyrosine toxicity.

D-Glucose dehydrogenase purified from the membrane of
Pseudomonas fluorescens was shown to be highly hydrophobia in amino acid analysis, with a polarity of 39.7%. The purified enzyme was inactivated upon removal of detergent by acetone treatment. The detergent-depleted enzyme was activated partially with Triton X-100, and the activity was restored almost completely upon addition of both phospholipids and Triton X-100, followed by sonication. The purified enzyme, in spite of being a single polypeptide dehydrogenase, directly reduced not only short-chain ubiquinone but also long-chain homologs. It should be noted that coenzyme Q-6 or Q-9 incorporated in phospholipid vesicles was efficiently reduced with the enzyme. These results show that, in the cytoplasmic membrane of
Pseudomonas fluorescens, the glucose dehydrogenase may be linked to an electron transport chain
via ubiquinone.

The interaction of some anthracycline antibiotics (adriamycin, daunomycin, aclacinomycin-A) with bacteriophage øX174 was investigated. Adriamycin and daunomycin inactivated the infectivity of both free øX174 phage and naked single-stranded øX174 DNA without DNA strand scission, but aclacinomycin-A did not show this action. The phage inactivation reaction was reversibly inhibited by superoxide dismutase, catalase or other oxygen radical scavengers. The inactivation of øX174 by adriamycin and aclacinomycin-A was stimulated by the addition of Cu
2+, while the øX174 inactivation by daunomycin was inhibited by the addition of Cu
2+. The øX174 inactivation by adriamycin and aclacinomycin-A in the presence of Cu
2+ was caused by degradation of DNA, and this inactivation reaction was inhibited irreversibly by oxygen radical scavengers. These results indicate that anthracycline antibiotics bind to ΦX174 DNA in the form of free radicals and that during the auto-oxidation of these antibiotics in the presence of Cu
2+, oxygen radicals were generated to cause the degradation of øX174 DNA.

A chemically defined medium was devised to examine the growth, production and biochemical pathway of tetrocarcin A. The stereochemistry of 1, 3-eliminative cyclopropanation was established in the NaH-catalyzed condensation system involving carbanionic reagents with chlorine and phosphate as the leaving group.
It was proved by means of product analysis that the steric course of 1, 3-elimanative cyclopropanation was determined at the 1, 3-eliminative S
N2 displacement by the catalyst base, stability of the leaving group and adduct carbanion as well as by solvent polarity, The
cis/trans ratios, as well as the optical yeilds of cyclopropane products, agreed closely with the prediction based on the transition state topology at the 1, 3-eliminative cyclization step. It is therefore suggested that the asymmetric Michael type cyclopropanation could be applicable for the absolute assignment of configuration to chiral cyclopropanes.

Biosynthetic threonine deaminase was purified to an apparent homogeneous state from the cell extract of
Proteus morganii, with an overall yield of 7.5%. The enzyme had a s
020, w of 10.0 S, and the molecular weight was calculated to be approximately, 228, 000. The molecular weight of a subunit of the enzyme was estimated to be 58, 000 by sodium dodecyl sulfate gel electrophoresis. The enzyme seemed to have a tetrameric structure consisting of identical subunits. The enzymehad a marked yellow color with an absorption maximum at 415 nm and contained 2mol of pyridoxal 5'-phosphate per mol. The threonine deaminase catalyzed the deamination of L-threonine, L-serine, L-cysteine and β-chloro-L-alanine.
Km values for L-threonine and L-serine were 3.2 and 7.1mM, respectively. The enzyme was not activated by AMP, ADP and ATP, but was inhibited by L-isoleucine. The
Ki for L-isoleucine was 1.17 mM, and the inhibition was not recovered by L-valine. Treatment with mercuric chloride effectively protected the enzyme from inhibition by L-isoleucine.

Bovine casein micelles were fractionated on controlled pore granule (CPG-10/3000) chromatography by size and the chemical properties of the fractionated micelles were compared. The results indicated the presence of two types of micelles distinguishable as large and small micelles. In skim milk, 72.7% of casein was calculated to be in the form of small micelles, 13.6% in the form of large micelles and 13.8% in non-micellar casein form.
The α
s1-casein content decreased, but β- and Κ-casein content increased as the micelle size became smaller. Κ-Casein in large micelles had a much higher sialic acid content than in small micelles. It was found that this difference in sialic acid content was due to the presence of non-glycosylated Κ-casein in small micelles. In large micelles, non-glycosylated Κ-casein was almost undetectable.
The addition of wheat germ lectin to micelles resulted in the formation of aggregates through intermicellar bridges between the carbohydrate chains of Κ-casein located on the surface of the micelles. Both large and small micelles formed aggregates after the addition of wheat germ lectin. Large micelles were more sensitive to wheat germ lectin than small ones.

Myosin and reconstituted actomyosin free of regulatory proteins were mixed in 0.6M KCl and 20 HIM phosphate buffer at pH 6.0 and were tested quantitatively for thermally induced gelation properties by measuring the rigidity of the system at 65°C. Full enhancement of gelation was attained when the weight ratio of myosin-to-actomyosin was about 4. The addition of regulatory proteins to actomyosin could restore calcium sensitivity of the contractile system, but did not affect the heat-induced gelation of myosin in the presence of actomyosin, suggesting that regulatory proteins play no role in the heat-induced gelation of the system.
Neither the single and double headed subfragments, both capable of interacting with F-actin, nor the helical tail subfragments, devoid of the intracting site with F-actin, exhibited changes in thermogelling properties when mixed with F-actin. However, upon addition to F-actomyosin, the tail subfragments revealed a significant effect on the gelation of actomyosin, whereas the headed subfragments exerted no influence over gelation of the system. These results indicate that the enhancing effect of F-actin on the heat-induced gelation of myosin was brought about solely by the limited amount of F-actomyosin formed in the system, which acts as a cross-linker between the tail portion of bound and free myosin molecules.
