The relationship of the β-myrcene content with the susceptibility and/or resistance of
Pinus species against the pine wood nematode was investigated. For this purpose, the β-myrcene content of the wood of one-year-old shoots was determined. The resistant trees were found to contain a very small amount of β-myrcene. On the other hand, the β-myrcene content of susceptible trees was comparatively high. This relationship was observed in both inter- and intraspecies and also between young and older trees of the same species,
i.e.,
Pinus densiflora or
P. thunbergii. The β-myrcene content can feasibly be used as an indicator for screening resistant Japanese red and black pine trees.

Acrylonitrile-hydrating activity was found in various bacteria belonging to the genera
Arthrobacter,
Corynebacterium,
Microbacterium,
Nocardia,
Pseudomonas and
Rhodococcus. A strain, N-774, isolated by acetonitrile enrichment culture from soil showed the highest activity. Taxonomic studies indicated that strain N-774 belonged to the genus
Rhodococcus. The acrylonitrile-hydrating enzyme of strain N-774 was constitutively formed in the cells. Besides acrylonitrile, many nitriles were hydrated to the corresponding amides.
n-Butyronitrile, succinonitrile and chloroacetonitrille served as suitable substrates. This bacterium could utilize many aliphatic nitriles and amides as a sole source of nitrogen but hardly utilized malononitrile, acrylonitrile or acrylamide. Cells having high nitrile hydratase activity (about 50units/mg of dry cells) could be easily prepared by cultivation at 30°C for 40 hr in a malt extract and yeast extract medium.

The culture conditions for
Rhodococcus sp. N-774 cells showing high nitrile hydratase activity and the reaction conditions for acrylamide production by the resting cells were optimized. Thiamine was essential for the growth of the strain. Yeast extract and Fe
2+ or Fe
3+ remarkably promoted the formation of nitrile hydratase of the cells. The reaction proceeded optimally at temperatures below 30°C. Incubation for 1 hr at above 40°C resulted in inactivation of the enzyme. Through reaction at a temperature as low as 0°C, the inhibition and inactivation of the enzyme activity by the substrate, acrylonitrile, and the product, acrylamide, were remarkably reduced, and higher accumulation of acrylamide could be attained. Under the optimal conditions, a more than 20% (w/v) acrylamide solution was obtained with a conversion yield of nearly 100%. Thus, the aqueous acrylamide solution obtained showed a high enough quality for use for the commercial preparation of polyacrylamide.

A mesophilic fungal strain Y-94 produced three types of thermostable endo-xylanases accompanied by the formation of a large amount of cellulase. These xylanases were separated from the cellulase by heat treatment at 65°C for 2.5 hr and purified by DEAE-Toyopearl chromatography, chromatography on an anion exchanger (PBE 94), and Bio-Gel A 0.5m gel filtration. The molecular weights of the three types of xylanase, designated as xylanase A, B and C, were 51, 000, 48, 000, and 35, 000, respectively. All three enzymes showed highest activity at pH 4.9 and 80°C in 10min of incubation, and had the same hydrolysis pattern of larch wood xylan with the endproducts of xylobiose and xylose. Thus their activities appear essentially the same but not their stabilities. Xylanase A and B were stable from pH 2.5 to 9.0 but xylanase C was unstable above pH 5.5. Xylanase C was unstable at 70°C where other two were stable.

The membrane lipids of six higher plants that differ in salt tolerance were analyzed and compared. The root lipids increased in a ratio of glycolipid/phospholipid with increasing salttolerance. A similar increase in the ratio was observed with increasing external salinity when halophytic orach and salt-sensitive cucumber were exposed to varying salinity, although the latter plant was limited to only a little increase. Measurements of ion-transport rates with artificial lipid membranes revealed that the root lipids from a salt-resistant plant formed a more permeable membrane than those from a salt-sensitive species. It was found that the membrane permeability was related to the glycolipid/phospholipid ratio in the membrane lipids, where the glycolipids were stimulative and the phospholipids were repressive for ion-flow. These different effects of the two lipid classes may be attributed to their molecular species and head groups.

Bacillus cereus, in addition to soybean β-amylase [

99, 1631 (1986)]. However, none of the compounds used inactivated any α-amylases [EC 3.2.1.1] of porcine pancreas,
Aspergillus oryzae, or
Bacillus amyloliquefaciens. Irreversible incorporation of
14C-labeled α-EPG into β-amylases was stoichiometric,
i.e., one α-EPG per active site of the enzyme was bound, and the inactivations were almost complete. The results suggest that α-EPG is an affinity labeling reagent selective for β-amylase. Slow inactivations by the other compounds were also observed, depending on the difference of source of β-amylase.

The effects of glycerin and ethylene glycol on the elastic modulus and DSC thermograms of agarose and kappa-carrageenan gels were examined to clarify the relation between structure and properties. The elastic modulus of these gels as a function of the concentration of polyols increased up to a certain concentration and then decreased with increasing concentration of polyols. These polyols shifted the melting temperature of the gel to higher temperatures in kappa-carrageenan gels but to lower temperatures in agarose gels. The temperature dependence of elastic modulus was changed in opposite directions in agarose and kappa-carrageenan gels by the addition of polyols, and this is discussed on the basis of model consisting of junction zones which are connected by Langevin chains. It was suggested that the mean distance between junction zones became shorter in the presence of a small amount of polyols.

Wall-bound α-glucosidase (EC 3.2.1.20) has been solubilized from suspension-cultured rice cells with Sumyzyme C and Pectolyase Y-23 and isolated by a procedure including fractionation with ammonium sulfate, Sephadex G-100 column chromatography, CM-cellulose column chroraatography, Sephadex G-200 column chromatography, and preparative disc gel electrophoresis. The molecular weight of the enzyme was 64, 000. The enzyme readily hydrolyzed maltose, maltotriose, and amylose, but hydrolyzed isomaltose and soluble starch more slowly. The Michaelis constant for maltose of the enzyme was estimated to be 0.272 mM. The enzyme produced panose as the main α-glucosyltransferred product from maltose.

The L-leucine productivity of an L-leucine producing strain, H-1204, of
Corynebacterium glutamicum substantially decreased during a large-scale culture or repetitive subculturing. This instability was found to be due to the appearance of revertants with lower or no L-leucine productivity. Chlorophyll- and β-carotene-egg albumin complexes were prepared. As these pigment-protein complexes were soluble in water, the chlorophylls and β-carotene were endowed with a water-soluble character. The molecular numbers of pigments incorporated into the complexes were increased by using proteins treated with charcoal, and additionally by using chemically modified proteins, acetylated or carbamoylated egg albumin. When compared with the stabilities of these pigments in detergent or in an organic solvent, the chlorophyll in the complexes was stable against photobleaching and for a wide range of pH values (pH 5-10). β-Carotene in the egg albumin complexes was relatively stable under UV irradiation.

The nucleotide sequence of cDNA encoding the glycinin A

1a subunit from var. Shirotsurunoko was determined and compared with that in the case of var. Bonminori. The comparison showed six nucleotide substitutions in the coding sequence, one of which results in one amino acid replacement, and three in the 3'-noncoding region. These differences indicate the occurrence of polymorphism of the glycinin A

Bacillus circulans F-2 requires a special carbon source or cultural conditions for amylase production. The α-glucosidase production of this bacterium was studied in various cultural conditions with measured glucose concentrations. High amylase production was always accompanied by low α-glucosidase production and the absence of glucose in culture broth. Usually higher α-glucosidase production was observed in cultural conditions where little amylase was produced. In the presence of 1-deoxynojirimycin, an inhibitor of α-glucosidase activity, the bacterium produced significant amounts of amylase even in conditions giving high α-glucosidase production. It was concluded that the special requirement of this bacterium to produce amylase is effected by its high sensitivity to glucose repression and by the production of α-glucosidase which leads to the formation of glucose. Production of α-glucosidase was, like that of amylase, induced by maltooligosaccharides and repressed by glucose, but both its induction and repression are less sensitive to glucose than those of amylase.

Caffeine and caffeine-containing beverages (instant coffee, black tea, green tea, or oolong tea) caused a significant decrease in serum tryptophan, and significant increases in brain tryptophan, serotonin, and 5-hydroxyindole acetic acid over those in rats fed a control diet. Adenosine supplementation partially counteracted the increase of brain serotonin caused by caffeine. These results are interpreted as indicating that caffeine-containing beverages may have some nutritional and behavioral effects.

A phenylcarbamate degrading enzyme was isolated from
Pseudomonas alcaligenes. The enzyme was purified to a specific activity of 119U/mg by ammonium sulphate precipitation, gel filtration, DEAE and hydroxy-apatite chromatographies. The purified enzyme was found to be homogeneous on SDS polyacrylamide gel electrophoresis. The molecular weight was estimated to 68, 000. The pH optimum was around 9.5 and the temperature optimum was 28°C. The
Km for CIPC was 1.19 × 10
-5M. Hg
2+, PMSF inhibited the enzyme, but thiol reagents and EDTA had no effect. The enzyme degraded a number of phenylcarbamate herbicides (CIPC, BIPC, IPC and swep) and propanil but did not hydrolyse barban and carbetamide, which are phenylcarbamates, or monuron and linuron, which are phenylureas. The enzyme is probably an amidase.

Eighty-one strains of bacteria were tested for their ability to catalyze the release of chloride ion from DL-2-amino-4-chloro-4-pentenoic acid. A dehalogenating enzyme was obtained from the cells of
Proteus mirabilis IFO 3849, which can use the L-isomer. The enzyme was constitutively produced. The conversion of L-2-amino-4-chloro-4-pentenoic acid to 2-keto-4-pentenoic acid, ammonia, and chloride ion was demonstrated. The reaction product, 2-keto-4-pentenoic acid, was isolated as its 2, 4-dinitrophenylhydrazone and identified by catalytic hydrogenolysis of the hydrazone to the corresponding amino acid, norvaline.

An aqueous-alcohol solution containing gliadin (or zein) and linoleic acid was pulverized by spray-drying and the powders were examined for their stability, digestibility and application. Linoleic acid in the powders was kept stable during three months' storage in each case of low, moderate and high water activity. When the spray-dried powders were treated with such digestive enzymes as pepsin and pancreatin, linoleic acid was effectively changed into the hexane-extractable form concomitantly with the digestion of gliadin. The powders were administered to rats, instead of gluten and essential fatty acid in a semi-purified diet, and there was no abnormal excretion of fatty acid into the faeces. These facts indicate that two components of the powders could be digested and absorbed in the gut as usual. On the other hand, supplementation of bread with gliadin and linoleic acid in the form of spray-dried powders led to a considerable increase in bulk after baking, without affecting the taste and flavor. Linoleic acid in a loaf of bread seemed to be fairly resistant to peroxidation during baking in an oven and during storage under aerobic conditions for a week.

In order to investigate the role of phenylalanine and tyrosine residues in the bitter taste of peptides, some oligopeptides containing phenylalanine or tyrosine were synthesized and their taste was evaluated. The hydrophobicity of the phenylalanine or tyrosine molecule markedly caused the bitter taste in peptide. The bitterness was more intense when phenylalanine was located at the C-terminus and when the content of phenylalanine or tyrosine was increased in peptides. The hydrophobic residue in peptides functioned as a bitter taste determinant site. The experimental results suggest the existence of an additional site for the bitter taste of peptides.

An endo-β-1, 6-glucanase (E.C. 3.2.1.75) was purified from the culture filtrate of
Neurospora crassa IFO-6068 by chromatographies on CM-cellulofine, Con-A Sepharose 4B, and Sepharose CL-6B followed by preparative affinity gel electrophoresis. The purified enzyme had an apparent molecular weight of 47, 000. The pH and temperature optima for the activity were 5.0 and 50°C. The enzyme acted on β-1, 6-glucan (Pustulan) and yielded a series of gentio-oligosaccharides with endotype action, and finally, glucose and gentiobiose were produced. The enzyme was also able to act on
N. crassa cell wall β-glucan, and a small amount of hydrolysis fragments were liberated without apparent change of the cell wall glucan molecules.

Among 21 strains of
Gluconobacter,
Gluconobacter cerinus (IFO 3268) had a thermostable superoxide dismutase activity. The enzyme was purified about 110-fold to homogeneity from a cell-free extract by ammonium sulfate fractionation and DEAE-Toyopearl and Sephadex G-100 chromatography. The enzyme has a molecular weight of 47, 000 and consists of two identical subunits, and contains 1.22 g atoms of Mn per mole of enzyme as the catalytically active metal. The enzyme disappeared from circulation in the guinea pig with a half-life of 45min, but enzyme modified with polyethylene glycol 5, 000 had a half-life of 330 min.

When
Neocosmospora tenuicristata was cultured on shredded wheat medium, monorden was produced in relatively large quantities. Monorden significantly (
p<0.01) inhibited the growth of etiolated wheat coleoptiles at 10
-3 and 10
-4 M, by 100 and 44% respectively, relative to controls. Neither 6-week-old tobacco plants nor 10-day-old bean plants were affected by monorden sprays, but 10-day-old corn plants had slight necrosis when treated with 10
-2 M solutions.
Bacillus subtilis and
B. cereus were moderately inhibited by monorden. Single crystal X-ray diffraction studies defined the entire stereochemistry for the first time and the three asymmetric carbon atoms had R configurations. Thus, the configuration of the CH
3 at C17 has now been established.

We screened for fungi that can decolorize molasses melanoidin in the tropical zone and isolated some strains, mainly in the genus
Aspergillus. Of these, strain No. G-2-6 was most active and was identical with
Aspergillus fumigatus based on detailed morphological studies.
This strain decolorized about 75% of a molasses melanoidin solution when the strain was cultivated on a glycerol-peptone medium at 45°C for 3 days with shaking. In successive decolorization reusing the mycelia, this strain had more than 60% of the melanoidin-decolorizing activity at the eighth replacement in the presence of 4% glycerol.
Continuous decolorization of molasses melanoidin solution in a jar fermentor had an almost constant decolorization yield of about 70% at a dilution rate of 0.014hr
-1. At the same time, about 51% of the chemical oxygen demand and 56% of the total organic carbon in the initial solution were removed. In contrast, continuous decolorization of non-dialyzed molasses melanoidin solution removed a little more chemical oxygen demand and total organic carbon than those of dialyzed molasses melanoidin solution, but had a lower level of melanoidin-decolorizing activity (about 40%).

The addition of an organic solvent to the reaction mixture led to a change in the activity of glycerol dehydrogenase. The activity on glycerol decreased to lower than that on 1, 2-propanediol. This was due to the apparent increase in the
Km value for each substrate. The system was applied to determination of 1, 2-propanediol. The standard curve for 1, 2-propanediol with a rate assay method was not affected by a 10-fold amount of glycerol in the presence of
n-butanol. Chemical modification, except for succinylation of the NH
2-residue, did not cause any change in substrate specificity. The participation of a His-residue in the active site was suggested by the results of chemical modification.

The characteristics of dopamine- and norepinephrine-stimulation of respiration in isolated rat brown adipocytes were compared. Although both dopamine and norepinephrine rapidly stimulated the respiration and lipolysis of isolated cells, much higher concentrations of dopamine compared to norepinephrine were required to cause the same extent of response. Antagonists of the dopamine receptor (haloperidol) and of the β-adrenoreceptor (propranolol) suppressed respiration that was stimulated by either dopamine (50 μM) or norepinephrine (0.2 μM). Unexpectedly, dopamine-stimulated respiration was completely inhibited by the β-adrenergic blocker at a concentration far lower than that required to inhibit norepinephrine-stimulated respiration. Both catecholamines added at concentrations of a large excess to stimulate respiration overcame the inhibitory action of propranolol but not of haloperidol. An agonist of the dopamine receptor, apomorphine (50 μM), failed to stimulate the respiration of brown adipocytes. These results indicate that dopamine at a high concentration stimulates the respiration of brown adipocytes primarily through interaction with the β-adrenoreceptor, but not with the dopamine receptor.

We fractionated the homogenate of Antarctic krill,
Euphausia superba, with a Sephacryl S-400 column and obtained two protease fractions, A (M.W. 50 K) and B (M.W. 30 K). Both fractions digested preferentially the peptide bond adjacent to arginine, and also digested peptide bonds adjacent to lysine and leucine. Their optimum pH was 9.0 and optimum temperature was 37°C in the absence of 2-mercaptoethanol. In the presence of 4.4% 2-mercaptoethanol, the activity of fraction A was stimulated below 30°C, but inhibited at higher temperatures. This led to the shift of optimum temperature from 37°C to 20°C. Some of alcohols also strongly stimulated the krill protease at 20°C.

The leucine dehydrogenase (L-leucine: NAD
+ oxidoreductase, deaminating, EC 1.4.1.9) gene of
Clostridium thermoaceticum was cloned and expressed in
Escherichia coli C600 with a vector plasmid, pICD242, which was constructed from pBR322 and the leucine dehydrogenase gene derived from
C. thermoaceticum. The enzyme overproduced in the clone was purified about 12 fold to homogeneity by heat treatment and another two steps with a yield of 46%. The enzyme of
E. coli-pICD242 was immunochemically identical with that of
C. thermoaceticum. The enzyme has a molecular weight of about 350, 000 and consists of six subunits identical in molecular weight (56, 000). The enzyme is not inactivated by heat treatment: at pH 7.2 and 75°C for 15min; at 55°C and various pH's between 6.0 and 10.0 for 10min. The enzyme catalyzes the oxidative deamination of branched-chain L-amino acids and the reductive amination of their 2-oxo analogues in the presence of NAD
+ and NADH, respectively. The
pro-S hydrogen at C-4 of the dihydronicotinamide ring of NADH is exclusively transferred to the substrate; the enzyme is
B stereospecific. The enzymological properties are very similar to those of the
Bacillus stearothermophilus enzyme [T. Ohshima, S. Nagata and K. Soda,
Arch. Microbiol.,
