  • Studies on the blood pH estimated by the glass electrode method. V

    1. A new micro-lass electrode for 0.07cc. material was designed. Method of the pH determination with it was described in details.
    2. This micro-electrode was applied to the measurement of the blood pH, and it was found that it gave a correct value within 0.02 pH.
    3. In experiments on rabbits loosely fixed on a board and on resting human adults, it was confirmed by direct measurement that the pH of arterial blood is more alkaline by 0.03 pH on an average than that of venous blood, while the former coincides with the pH of cutaneous capillary blood within the scope of experimental error.

    Each phase of the reactions involved in the Embden-Meyerhof theory on the production of lactic acid from hexosedi-phosphoric acid by muscle tissue, was investigated with the employ-ment of Kato spindle cell sarcoma of rabbit instead of muscle. The results obtained are as follows.
    1. Triosephosphoric acid was produced from hexosediphos-phoric acid by tumour tissue. The velocity of the reaction was. however, slower than that of muscle tissue.
    2. Tumour tissue was unable to dismutate two molecules of triosephosphoric acid into phosphoglyceric and glyceroiphosphorie acids.
    3. The decomposition of phosphoglyceric acid into pyruvic acid was not carried out by tumour tissue, the reaction being stopped at the stage of phosphopyruvic acid. However, pyruvic acid was easily produced from phosphopyruvic acid in the presence of adenosinetriphosphoric acid and adenylicc acid, or phosphatase.
    4. The dismutation of pyruvic acid into lactic acid in the presence of α-glycerolphosphoric acid was not observable in tumour tissue.
    5. In spite of the facts mentioned above that the process of the glycolysis of the Embden-Meyerhof hypothesis was stopped at several phases, a small amount of lactic acid was always produced from hexosediphosphoric acid by the tumour slices. The mechanism of this reaction is quite obscure at present. This reaction was inhibited only to a relatively slight degree by the quantities of fluoride or monoiodoacetic acid which completely stopped the production of lactic acid from glucose.

    1. In this paper the comparison was made of the action of tyrosinase on various phenolic substances alone or in the presence of a number of amino acids and other amino compounds.
    2. Only those phenols, which are likely to produce ortho-quinones on oxydation gave coloured substances and the presence of amino acids markedly accelerated the production of pigments.
    3. The oxygen up-take per molecule of p-cresol or catechol by the action of tyrosinase was found to be one atom more than that corresponding to the formation of ortho-quinone.
    4. The presence of amino acids and a series of aromatic amino compounds remarkably accelerated the rate of oxygen consumption. The total amount of oxygen absorbed however was not in any way affected by the addition of amino compounds.
    5. The suggestion was made that amino acids combine with orthoquinones produced by the tyrosinase action.
    6. Hydroxylamine accelerated the colour development of the tyrosinase system. The oxygen consumption was on the contrary depressed. The discussion was made on the probable cause of this phenomena.
