    The post-transcriptional "maturation" process involved in the biosynthesis of tRNA is reviewed in the light of our recent finding on tRNA precursors and the mode of their enzymatic cleavage. A number of tRNA precursors were identified in a thermosensitive mutant of E. coli defective in RNase P and extensively characterized by fingerprinting and by in vitro cleaving reaction. These studies have revealed the following features of maturation process:
    (1) Many tRNA genes, if not all, are clustered in the E. coli genome and thus transcribed in multimeric precursor forms, although the tRNA gene clusters are rather scattered throughout the entire chromosome.
    (2) The multimeric precursors are cleaved by RNase O at the spacer regions into monomeric and/or dimeric forms, which are subsequently processed by RNase P at the 5' sides of the molecules into smaller monomeric forms. The monomeric intermediates are then trimmed at the 3' termini by RNase Q.
    (3) Modification of nucleotides seems to occur at distinct stages of maturation process, thereby indicating that modification and trimming reactions are somehow interrelated.

    Recent studies on self-organization mechanism of protein folding are reviewed. First, brief descriptions of the difference between thermodynamical and self-organizational mechanism of protein folding are made. Then it is stressed that self-oranization mechanism is favorable for understanding the quick folding of the tertiary structure of protein and the strong cooperativity of denaturation-renaturation phenomena. Some theoretical and experimental evidneces for self-organization mechanism are critically reviewed.

    This review contains the historical development on the insulin structure study which was begun in 1935 by D.C. Hodgkin, and the relationship between the structure and the biological activity. As 2 Zn insulin crystals belong to space group R3 and the positions of the main heavy atom in the most derivatives are very close to the three fold axis, many difficalties had to be overcome during the structure determination. We describe the three dimensional arrangement of atoms in the molecule in 2 Zn insulin crystal and particularly its states of aggregation. The binding site of insulin receptor is proposed on the basis of the three dimensional structure, biochemical studies, and the studies using modified insulin, and the structure of proinsulin is also mentioned.
