ジャーナル フリーCannabis sativa produces novel secondary metabolites called cannabinoids. Among them, Δ1-tetrahydrocannabinol (THC), the psychoactive component of this plant, has attracted a great deal of attention, because this cannabinoid is shown to exert a variety of therapeutic activities such as the relief of nausea caused by cancer chemotherapy. To reveal the mechanism of THC formation in Cannabis sativa, we attempted biosynthetic studies. Consequently, we established that Δ1-tetrahydrocannabinolic acid (THCA), the precursor of THC, is derived from cannabigerolic acid and that this reaction is catalyzed by the novel oxidocyclase, THCA synthase. Here, we show the biochemical and structural properties of THCA synthase.

ジャーナル フリーAlthough photoperiodic response has been an object of study for a long time, little is known about molecular mechanisms underlying this phenomenon. Recently, we have identified the gene (Dio2) which regulates photoperiodic response in gonadal growth in birds and possibly in mammals. Prolactin secretion is also regulated by photoperiod. However, the mechanisms of this response are different from those of gonadal response. Here, we overview current knowledge about vertebrate photoperiodic response, providing our recent results on molecular studies.

ジャーナル フリーA hybrid approach using X-ray crystallography and cryo-electron microscopy provides opportunities to reveal the activation mechanism and pathway of biological macromolecules with large conformational changes. Structural analysis of the Ca2+ release channel, inositol 1,4,5-trisphosphate receptor (IP3R) is a good example to introduce this powerful approach. Docking of the atomic coordinate of the fragment including binding domain of inositol 1,4,5-trisphosphate (IP3) and Ca2+ into the three-dimensional density map of the type 1 IP3R reconstructed from cryo-electron micrographs leads to proposing a novel regulation mechanism of Ca2+ mobilization.

ジャーナル フリーWe examined dendritic activity in identified mechanosensory interneurons in the cricket cercal sensory system using Ca2+ imaging and intracellular recording techniques. Different dendritic branches of the interneurons displayed distinct directional sensitivity profiles to air-current stimuli. The amplitude of evoked spikes was larger at the dendritic branches in which the Ca2+ elevation was larger. Dual-site recordings of the dendritic spikes indicate that the spike-initiation site shifts depending on the location of the site of synaptic activation. We propose that a difference in spike-propagation direction could change the spike waveform at the distal dendrite and could produce synaptic activity-dependent Ca2+ dynamics.
