    Paternal transmission of mammary tumor virus (MTV) was revealed by the crosses between high mammary-cancer DD/Tbr and low mammary-cancer BALB/c or C57BL/6J strains of mice. The DD/Tbr strain of mice developed mammary tumors with 77.7% incidence at an average tumor age of 9.3 months in F44 to F56 generations. The hybrid mice between DD/Tbr (D) and BALB/c (C) developed mammary tumors with identical incidence in both reciprocal crosses; 66.7% incidence at an average age of 16.4 months in DCF1 and CDF1. In the hybrids between DD/Tbr (D) and C57BL/6J (B), an incidence of mammary tumor was 90.8% in DBF1 and 84.2% in BDF1 at an average tumor age of 8.7 months and 8.9 months, respectively. The DD/Tbr-MTV was more susceptible to the hybrid mice between D and B than to those between D and C. Mammary tumors from these hybrid mice demonstrated type B particles, suggesting that the DD/Tbr-MTV was transmitted by either maternal or paternal route. Plaque-inducing ability and paternal transmission are specific for DD/Tbr and GR strains, but the mammary tumor age in DD/Tbr strain is not so young as in the GR strain which carries MTV-P. RIII strain has a plaque-inducing ability, but fails to transmit its MTV (MTV-PS) by paternal route. Therefore, DD/Tbr-MTV has intermediate characteristics between MTV-P and MTV-PS.

    Histopathological study was made on 473 polypoid lesions in the stomach obtained surgically from 297 patients. The maximum diameter of each polypoid lesion was measured. Histological gradation based on the extent of the epithelial pseudostratification was used as the indicator of the process in the development of a polypoid lesion. Polypoid lesions were also classified histogenetically into the metaplastic and gastric types. The latter type was subdivided into the pure form and the mixed form with secondary metaplasia. The metaplastic type of polypoid lesions revealed a clear correlation between their size and histological grade, and showed a high incidence of cancerous lesions. Almost all polypoid lesions of this group had a diameter less than 20mm in the grades below V. Of the gastric type polypoids, on the other hand, the pure form not associated with intestinal metaplasia rarely underwent malignant change; and the average size of the lesions in each grade was larger than that of the metaplastic type. Approximately 10% of the pure form lesions in grades I, II, and III were found to have diameters larger than 20mm. The gastric type with secondary metaplasia revealed a combined character of both the metaplastic type and the pure gastric type in terms of the size as well as the malignant potential.

    Since it was demonstrated that 60-F product, which was precipitated by 0.6-saturation of ammonium sulfate from streptomycin-pretreated cell-free extract of hemolytic streptococci Su strain (Su cocci), possessed a potent anticancer activity, isolation of 60-F product was carried out by the same method using other streptococci and microörganisms. Results obtained were as follows: (1) The 60-F product, which was effective in depriving the invasiveness of Ehrlich carcinoma cells, was obtained from Blackmore and C203S of group A streptococci, same as Su cocci, all of which possessed the streptolysin-S forming ability and an anticancer ability. (2) The 60-F product obtained from six other streptococci and microörganisms, all of which did not possess the streptolysin S-forming ability and the anticancer ability, was entirely ineffective. (3) All of the 60-F products with cytotoxic activity possessed a hemolytic activity, and hemolysis by these products was inhibited by Trypan Blue, indicating that the hemolysis is due to streptolysin-S contained in the 60-F product. From these results, it was deduced that the cytotoxic 60-F product against tumor cells is obtained from group A streptococci possessing the streptolysin-S forming ability and anticancer activity.

    The effect of leupeptin on the induction of lymphoblastic leukemia in ICR/JCL female mice by N-nitrosobutylurea (NBU) was investigated. NBU was given as a 0.02% solution in drinking water for 10 weeks. A 0.1% concentration of leupeptin was given in the diet. Group A was fed on the leupeptin diet from the beginning, Group B received it after the end of NBU treatment, and Group C was fed on a leupeptin-free control diet throughout. The average periods in the appearance of leukemia in groups A, B, and C were 115±50, 112±43, and 100±17 days (mean ± SD), respectively, and there was a significant difference between groups A and B and Group C at P<0.001. In regard to this point, leupeptin might have the effect of retarding the process of leukemogenesis. However, leupeptin showed no effect on the incidence and histopathological finding of leukemia.

    Incorporation of uracil and uridine into ribonucleic acid (RNA) was compared among the ascitic and solid forms of Ehrlich mouse tumor, Morris hepatoma, Rhodamine sarcoma, gastric cancer and ulcer from human patients, and several normal rat tissues.
    Of these cells tested, the cells of Ehrlich ascites and solid tumors, human gastric cancer and ulcer, and certain tissues of a normal rat showed a considerably high activity. Furthermore, Ehrlich ascites tumor cells indicating a high incorporation activity was also high in activities of both phosphorylase and kinase for uridine, while Rhodamine sarcoma as a representative having a low incorporation activity was considerably low in these two enzymic activities. RNA synthesis from uridine phosphates by Rhodamine sarcoma was maintained to a fairly high extent contrary to its low activities of the phosphorylase and the kinase.
    Consequently, the low utilization of uracil and uridine by certain tumors was suggested to be due to the extremely low activities of both enzymes.

    Ten out of 31 male Buffalo rats developed thymomas, 18 months after birth, from the subcutaneous thymuses grafted neonatally either from female or male litter mates. Most of them were of lymphoid cell type and contained numerous lymphoid cells within the network composed of neoplastic epithelial cells, as mediastinal thymomas. Lymphoid cells in the subcutaneous thymomas had normal diploid karyotype and were of host type with the Y chromosomes. These facts may indicate that lymphoid cells intermingled in the network of neoplastic epithelial cells are non-neoplastic in nature.

    Time course changes in the concentration of the plasma proteins, especially α1-acid glycoprotein and α1-antitrypsin, after partial hepatectomy were investigated. The concentration of α1-acid glycoprotein rose steadily to the highest value, about four-fold of that of normal non-hepatectomized rats, at 27.5hr after partial hepatectomy, indicating excretion of the protein from hepatocytes in the residual liver during a very early period after partial hepatectomy, before hepatocytic mitosis, and thereafter fell slowly. The concentration of α1-antitrypsin decreased sharply to one-sixth of that of the non-operated control rats, at 18hr after partial hepatectomy, 10hr before the peak of mitosis, and thereafter rose steadily to the highest value above the control at 168hr, when the mitosis almost ended, suggesting a close correlation of the protein to the occurrence and suppression of mitosis of the hepatocyte.
    These results suggest the existence of a double-layered pattern of humoral factors involved in cell division; α1-acid glycoprotein may be a primary mitotic inhibitor, whose intracellular concentration over a critical level inhibits cell division and, in contrast, α1-antitrypsin may be one of secondary mitotic inhibitors, whose extracellular concentration below a critical level facilitates the excretion of the primary mitotic inhibitor from the cell in question, allowing cell division, and that over a critical level impedes the excretion of the primary one from the cell in question, inhibiting cell division.
    Based on these findings, the essential part of the regulatory mechanism of cell division in the regenerating rat liver is concretely discussed.

    Autoradiographic study was made on 24 cases of uterine cervical tumor, including 22 cases of squamous cell carcinoma, using a technique of in vitro labeling of surgical specimens with tritiated thymidine (3H-TdR) under hyperbaric oxygen. Labeling index of the cervical cancer ranged from 8.7 to 30.4% and was well correlated with the histological subtypes (spinal, transitional, and basal cell types). The upper limit of the growth fraction was also estimated from the general relationship between phase lengths. The necessity of sufficient oxygen tension for the S phase cells to effect synthesis of DNA was stressed.

    A response to diethylstilbestrol (DES) of DES-induced renal carcinoma in Syrian hamster was studied by comparing subcutaneous growth of this tumor in DES-treated and untreated animals with respect to the growth curve, cell cycle time, Tc, growth fraction, GF, and cell loss factor, φ. In both conditions the tumor growth followed the Gompertz curve, but the latent period was 14 days longer without DES than with DES. In the untreated animals Tc was 80% longer due to increases of TGl and TS. However, φ was not changed significantly and GF rather increased without DES. It was concluded that DES-dependency of this tumor growth was manifested in the prolongation of Tc.

    In order to clarify the relationship between histological subtypes or small cell carcinoma and the degree of their response to chemotherapy, 58 cases of small cell carcinoma of the lung were studied. Cyclophosphamide, vincristine, and 5-fluorouracil were administered intravenously. Fifty percent survival of small cell carcinoma patients was the longest in the stage I group and shortest in the stage III group, regardless of the secondary treatment. Fifty percent survival was the longest in the group responding markedly to the initial chemotherapy and shortest in the non-responding group, regardless of the clinical stage. The chemotherapy was found effective on the lymphocyte-like subtype according to the WHO histological classification, the fusiform cell type and the polygonal cell type, in that decreasing order. Different ultrastructural findings were observed in cases in the responding group and in the non-responding group. Noteworthy findings were (a) a marked irregularity in the nuclear shape, (b) irregular distribution of abundant condensed chromatins, (c) many perichromatin granules, and (d) abundant nucleolus-associated chromatins in the non-responding group. Although almost all small cell carcinoma contained neurosecretory granules, granules were few in the non-responding group.

    The estrogen receptor was measured in breast cancer of two male patients. Both patients had a positive receptor content in their tumors. The dissociation constant and number of binding sites were both very similar to those in the female counterpart. In one of the patients, the number of binding sites in the metastatic lesions of the liver was similar to that in the primary site. The patient responded well to the endocrine ablation therapy with a remission period of 8 months.
    The estrogen receptor was found to be positive in 12 out of 14 cases of male breast cancer reported previously including the present 2 patients. Three out of 4 cases with positive receptor content, including the present case, responded to orchiectomy.

    Mepitiostane (2α, 3α-epithio-5α-androstan-17β-yl 1-methoxycyclopentyl ether) is an orally active anti-estrogenic and anabolic-androgenic steroid compound. Mepitiostane administered orally suppressed the growth of transplanted estrogen-dependent mammary tumor in rats. This result was compared with the oral administration of fluoxymesterone, which is widely used orally for treatment of advanced breast cancer. Experimental result showed that Mepitiostane has a more dominant antitumor activity than fluoxymesterone. Therefore, this new compound may be useful for clinical treatment of breast cancer.

    Squamous cell carcinomas were readily induced in the lung of C57BL/6 mice by repeated intratracheal instillations of benzo[a]pyrene with charcoal powder suspended in saline solution. Intratracheal instillations were performed once a week for 8 weeks, and the observation period of the experiment was 40 weeks. Some of the tumors were highly keratinized and others were moderately keratinized squamous cell carcinomas. At a later period of observation, poorly differentiated squamous cell carcinomas were found. Invasion into blood vessels was occasionally seen but metastasis to regional lymph nodes or distant metastasis was not found. One tumor from the mouse killed on the 280th day was successfully transplanted subcutaneously into C57BL/6 mice. This mouse, killed on the 84th day after the first transplantation, had a pulmonary metastasis.

    Carboquone was found to greatly stimulate the aerobic NADH oxidation in the presence of both the partially purified NADH-cytochrome b5 reductase and cytochrome b5 prepared from hepatic microsomes by acting as an electron carrier from cytochrome b5 to molecular oxygen. Other quinoid anticancer chemicals, mitomycin-C, adriamycin, and daunomycin practically failed to do so.

    Three human cell lines, Burkitt lymphoma cells (P3HR-1), epidermoid carcinoma cells (HeLa S3-1), and melanoma cells (HMV) were irradiated with 200kV X-rays under three different oxygen conditions. The values of D0 and D10-2 were estimated for survival curves, and then the dose-modifying factors (DMF) were calculated, for the oxygenated and hypoxic irradiations. These DMF values for oxygenation were not different for each cell line, but those for hypoxia revealed considerable difference for cell lines. From the comparison of modifications due to variable oxygen concentrations on the survival curves of Burkitt lymphoma cells with those of other resistant cells, it is concluded that the shoulder of the survival curves becomes larger primarily with lowering of the oxygen tension, and then D0 value increases followed by a decrease in n value, while in oxygenation, the decrease in D0 value takes place after n value becomes close to 1.

    Three ascites hepatoma cells of the rat, AH-41B, AH-34, and AH-64B, which had been determined in vivo as α-fetoprotein-negative, were cultivated in vitro in a medium containing adenosine 3', 5'-cyclic monophosphate (cyclic AMP), cyclic dibutyryl-AMP, or theophylline. The concentration of α-fetoprotein in culture media was measured by the 125I-radioimmunoassay. Results demonstrated that the AH-41B cells produced α-fetoprotein in vitro, the concentration of which being elevated in the media with three substances, while the remaining AH-34 and AH-64B cells did not. A comment was made on the producibility of α-fetoprotein in the so-called α-fetoprotein-negative hepatoma cells.

    In the dietary administration of alkylbenzenesulfonate to Wistar rats for their lifespan, the preputial gland tumors were observed in both sexes including the control. This tumor was found in 12 out of 104 rats. As histological diagnosis of these 12 tumors, 7 were adenoma and 5 were adenoacanthoma.

    Carcinogenic activity of Panfuran-S in male rats was studied. Adenocarcinoma of duodenum and jejunum, and squamous cell papillomas of the forestomach developed in rats receiving 1750ppm or 3500ppm of Panfuran-S for 35 weeks. These results demonstrated the carcinogenic activity of Panfuran-S in rats.

    Direct mutagenicity of N-hydroxy-4-aminoazobenzene and 4-nitroazobenzene, and their 2-and 3-methoxyl derivatives to Salmonella typhimurium TA-100 and TA-98 was examined. Mutagenic activity of these dyes was directly related to carcinogenic activity of the corresponding 4-aminoazobenzene dyes.
