The following are the results obtained from fundamental and clinical study with fosfomycin-Ca in surgical field.
The antibiotic activity of fosfomycin-Ca was determined against
Staphylococcus aureus and gramnagative rods isolated from surgical lesions.
The peak of MIC obtained was 3.2 μg/ml for
Staphylococcus aureus, 6.3 g/ml for
E.coli, 12.5 pg/ml for
Klebsiella, 6.3 μg/ml for
Proteus vulgaris and 6.3-12.5, μg/ml for
Pseudomonas aeruginosa respectively, suggesting a broad antibiotic spectrum.
Three healthy sdults were given 1g of fosfomycin-Ca at fasting, and blood level and urinary excretion were examined. The peak of blood level was attained 3.2 μg/ml on average 2 hours after administration.
The amount excreted in urine was highest during 2-4 hours after administration, indicating total excretion rate of 16.6% for 12 hours.
Fosfomycin-Ca in oral form was administered to 20 cases of acute purulent infections of soft tissues in surgical field. Out of 20 cases treated, the results obtained were excellent in 4 cases, good in 9 cases, fair in 6 cases and poor in 1 case, efficacy rate being 95% including fair cases.
As a side effect with fosfomycin-Ca, one patient complained of a diarrhea, and yet the drug interruption was not necessary.
