  • Second report: Relationship between accelerating function to produce immunbodies and changing blood picture caused by skin irritation

    In order to experiment the function to produce immunbodies by skin irritation, I applied various sorts of irritations such as chemical, physical and mechanical irritation of many degrees on certain part of rabbits' abdomen-skin.
    My first experiment revealed, as I said in first report, that the skin irritation of certain degree accele rates the function to produce the immunbooies, however, on the other hand, too strong an irritation, above certain degree, obstructs the function.
    Now, by the second experiment I got the following results:
    1. Blood platelets count and white blood cells count are increased by skin irritation.
    2. The thrombocytele and leucocytele skin irritation have no relation to the sort of irritation I applied.
    3. The thrombocytele skin irritation could be caused if the irritation I applied comes up to certain degree, while the leucocytele has nothing to do with it.
    4. The trombocytele irritation has close connection with the accelerating function to produce immunbodies, though the leucocytele does not show any relation.

    For the purpose of clarifying the mechanism of the accelerating function to produce immunbodies by skin irritation, I have made experiments to reveal the relationship between the above mentioned accelerating function and the thyroid gland and spleen. I have removed the thyroid gland of several rabbits at first, and spleen next. Then I injected a definite dose of typhus bacillus in the vein and I applied the definite irritation on their abdomenskin, which will accelerate the function to produce immunbodies. As the result I have observed the increase of immunbody in the number worth against the typhus bacillus day by day.
    The result was as. follows :
    For the accelerating function to produce immunbodies by skin irritation the existence of thyroid gland and spleen is absolutely necessary. This relation is completely the same to that of reaction of our skin hormone so-called Esophylaxin, that we have experimentally and clinically studied about 10 years.

  • ESPECIALLY THE FUNCTIONAL ALTERATION OF TESTIS CAUSED BY THE REPEATED INJECTION OF TESTICULAR EMULSIONS

    The writer has studied the change of testicular function influenced by repeated irritations and has obtained the following results:
    1) The rabbit shows a decrease of serum calcium when 5% testicle-emulsion of the rabbit is injected, but when it is repeatedly injected over 15 times, it begins to show no reaction and during this time an active substance appears in the ear vein.
    The function of the active substance brings on the atrophy of the testicle of the mice and stops development of the testicle and the cockscomb of the cocks. The active substance appears earlier, and in/greater quantity in the serum of Vena spermatica interna and in the testicle than in the ear vein of the rabbit. And if the testicle is removed, this substance is vanished from the blood.
    2) The character of this active substance is a protein of globulin fraction and when the above so altered terticle is retransplanted to another body, the same substance appears in the blood.
    Therefore it is believed that this sunstance is a product produced by the qualitative alteration of the testicular function.
    From the above experiment is believed that the repeated injections of the organ-emulsion always makes the function of the concerned organ alter qualitatively.

  • ESPECIALLY THE RELATION BETWEEN THE PITUITARY GLAND AND SPLEEN INDICATED BY THE ABNORMAL SALT-FEVER

    Dr. N. NISHIO has formerly found that hypophysectomized rabbit is not fevered by injection of central pyretics (vaccine, milk, pepton, etc.), but invites hyperthermia by injection of peripheral pyretics (adrenalin, etc.).
    Thereupon I studied the change of salt-fever after removal of pituitary gland and its pathogenesis according to prof. Y. TOKUMITSU's emphasis that the deficiency states of the endocrine organs are not merely negative phenomena resulting from lack of these hormones but positive phenomena caused by the change of function of other endocrine organs. The results are as follows:
    Hypophyscctomized rabbit occurs no change of temperature by injection of NaCl or CaCl2 solution as the result of becoming dull to salt-fever.
    The salt-fever of the rabbit which was injected the serum of hypophysectomized rabbit becomes dull like the hypophysectomized rabbit. Thus the appearance of some active substance in the blood of hypophysectomized rabbit will be clear. This active substance vanishes after splenectomy or transplantation of normal spleen to hypophysectomized rabbit.
    Therefore this dullness to salt-fever is caused not only by negative phenomenon resulting from lack of hypophyseal hormones but also by active substance in the blood which is produced in the aberrated spleen as the result of hypophysectomy.
