    Numbers of studies on the morphology and the mode of growth of TB bacilli have been reported. For the cytological investigations of human type TB bacilli by means of electronmicroscope, the present authors designed a slide-collcdion-culture method, resulted from the investigation on the technique of preparing samples by slide-culture method. By this method, the process is relatively simple, the influences from outside is restricted and the changes of cell structures and the modes of multiplication can be successively observed under electron micrescope as well as through ordinary microscopes. Samples of TB bacilli were treated with Anderson's extraction method or with the method of removing the nucleic acid.
    The results obtained were as follows:
    1) Through alcohol-ether treatment, non-transparent bodies (body-A) increased slightly their transparency and the RNA of body-A was depolimerized. The cells were condensed, still the distribution of the RNA could be observed.
    2) Through chloroform treatment, preceded by alcohol-ether extraction treatment the contour of body-A became indistinct and the cells increased their density.
    3) Through chloric acid-alcohol-ether treatment, preceded by the successive employment of alcohol ether and chloroform extraction treatment, the body A could not generally be observed and the Feulgen-positive granules disappeared. The contour of cells body as not clear-cut and the cells had increased density.
    4) Through any method of trichloric acid treatment, body-A disappeared and almost all the granules became undetectable by means of whatever staining methods employed. The Feulgen's reaction turned to negative, and the cells lost their nature of affinity to pyronin.

    Since 1950, we have been conducting studies on the relation between clinical findings and Streptomycin resistance of TB bacilli in the treatment of 157 pulmonary TB cases. The results were as follows:
    1) Strains, which were resistant to 10y per ml or more of Streptomycin, were found in 2.6% of the 154 cases, before Streptomycin treatment.
    2) After the Streptomycin treatment, the ratio became higher in proportion to the overall doses of Streptomycin administered.
    3) No consistent relation was found between appetite, body weight, temperature, erythrocyte sedimentation rate, cough, sputum and the creation of Streptomycin resistant bacilli.
    4) The emergence ratio of the Streptomycin resistant bacilli was gererally high in the cases of which pulmonary tubercular cavities were distinctly recognized, and the ratio was small in the cases where the pulmonary tuberculous cavities disappeared during the period of Streptomycin treatment.
    5) In accordance with the extention dimensions of the pulmonary lesions, the emergence of the Streptomycin resistant bacilli had increased.
    6) In case of patients, who discharged TB bacilli in large quantity and whose discharge quantity was not affected by the Streptomycin treatment, the emergence ratio of Streptomycin resistant bacilli was high.
    7) According to our clinical experiments, the patients, who had Streptomycin resistant TB bacilli through Streptomycin administrations, could not receive the expected effects of Streptomycin and the larger the degree of Streptomycin resistance was, the less was the effects.

    The determination of the resistance of Mycobacteria Tuberculosis against Streptomycin, PAS and Isoniazid by direct, indirect and rapid methods is discussed in this report.
    The materials used were 46 sputa and one pleural exsudate, in which TB bacilli were proved by smear test.
    Methods: The Kosakai's modification of Urabe-Yamada's medium, containing 10 and 100γ/cc of Streptomycin, was employed in order to evaluate the resistance against _Streptomycin. For the determination of PAS's and Isoniazid's resistance, 3% of KH2PO4 media containing 1, 10 and 100γ/cc of PAS and 0.1, 1, 10 and 100γ/cc of Isoniazid were prepared for the direct method and 1% of KH2PO4 media containing the same proportions of PAS and Isoniazid were prepared for the indirect method. As for the rapid method, the Tatsuji-Ogawa's method was employed.
    The recordings were taken on the 30th and the 40th day in case of direct method and at the end of the 2nd and 3rd week in case of indirect method, after cultivation. For the rapid method, the readings were taken when the growth of TB bacilli was obtained in the control media which were carefully observed every other day, from the 7th day of cultivation.
    Results and Conclusion: The resistance was determined as the highest concentration in which the growth of TB bacilli was same as in the control media.
    1) Through the three methods which were carried out concomitantly, the same results were obtained in 23 cases out of 42 cases (55%).
    2) 28 cases out of 42 cases (67%) gave the same results in rapid method group and direct method group. Out of 14 cases, which gave different results by each method, 7 cases gave higher values and the remaining 7 cases gave lower values of resistance. by rapid method compared to that of the direct method.
    Acording to the above results, Ogawa's rapid method may still be inadequate as the routine test.
    3) 36 cases out of 47 cases (77.0) gave same values of resistance by indirect and direct methods. All cases which gave different values by each method gave lower values by indirect method than the values obtained by direct method.
    4) The results obtained by direct method on the 40th day were same values as that of the 30th day. The results obtained by indirect method at the end of the 2nd week were same values as that ones taken at the end of the 3rd week.
    Therefore, the readings should be taken on the 30th day of experiment in case of direct method and at the end of the 2nd week in case of indirect method.

    Generally, 0.5% aqueous solution or 1.5% ointment of cortisone are customally used for the local application of cortisone against eye diseases, We conducted clinical study whether weaker solution of cortisone has any effects or not, In our experiments, 0.025% or 0.0125% of cortisone's aqueous solution and 0.01% cortisone ointment were administered 6 times daily or every one hour into conjunctival sacks or upon eye lids. Other symptomatic treatments were also given.
    The prescriptions were as follows:
    1) As for the eye-lotion, saline suspension of cortisone acetate Merck (24mg per cc) —0.05 or 0.1cc, methylcellulose —0.1g, sodium chloride—0.04g, sodium borate—0.2g and destilled water —10.0g.
    2) As for the eye-ointment; Merck's ointment of cortisone acetate (1.5%)—0.1g, lanolin anhydrid—7.5g and vaselin alba—7.4g.
    Although dramatic results, i. e., disappearance of all the symptoms within one or two days of administration were not obtained, subjective and objective symptoms were mitigated quickly and the periods of illness were shortened. Remarkable effects were found in phlyctenular conjunctivitis, phlyctenular keratitis, superficial punctate keratitis (initial stage), nummular keratitis, disk-shaped keratitis, rosacea keratitis, trachomatic pannus, lid der matitis, and tennonitis cases. Slight effects wete found in scleritis and scierosing keratitis cases. No effects were found in dendritic keratitis and syphylitic interstitial keratitis (end stage) cases.

    The bacteriocidal activities of INAH, DSM, PAS, TB1, Cepharanthin, Vitamin K, Z-Furan, Monafuracin, Guanofuracin, Protomin, Protozol, Diazone and Neo-Minophagen A. T. against H37RV strain of M. Tuberculosis were investigated. Results obtained were as follows:
    1) H37RV were immersed into each drug at various degree of density for 1, 24 and 48 hours, and cultivated in the Oka-Katakura media, and washing off treatment was applied. Charts were prepared to record the densities which completely or imcompletely killed the bacilli. The result showed that INAH was the strongest one.
    2) H37RV were immersed into INAH, DSM, PAS, TB1 and Vitamin K respectively, for 1, 24 and 48 hours (washing off treatment according to Sasano's Method was applied), and planted upon Oka-Katakura media. Bacteriocidal density of drugs was generally higher than in case of the experiment No. 1. INAH, TB1 and Vitamin K gave bacteriocidal results, while DSM and PAS did not at all. By this experiment, using both Proskauer-Beck culture media and Oka-Katakura media, we found that the bacteriocidaj effects of each drug were stronger in the former culture media than in the latter media. DSM and PAS also showed bacteriocidal effects, though insignificant, in Proskauer-Beck culture media. INAH showed the strongest effect.
    3) If the bacilli were immersed into INAH, DSM, PAS and TB1, respectively, for 1 week, 2, 3 and 4 weeks, they were killed even in comparatively dilute density.
    4) According to our experiments, the results seemed to be depndent more or less, on the culture media employed. For example, the results obtained in Proskauer-Beck media were higher values than in case of Oka-Katakura media. DSM hardly showed bacteriocidal effect, when the quantity of bacilli were large.
