In order to make
mirin containing glycerol, suitable
koji molds were screened in
Mucor and
Rhizopus stock cultures, which have been practically used in the production of Chinese beverages and Indonesian Tempeh.
R. oryzae IFO 4716 was selected as a favorable strain for accumulating glycerol in rice
koji. The fermentation conditions for glycerol production on
koji were examined with this strain. The optimal conditions for the glycerol production in
koji with crushed rice of polishing ratio 90% was determined to be 33-35°C, 10-45%(v/v) of oxygen concentration, pH and 90-95% of relative humidity.
Under these fermentation conditions, this strain accumulated in 7 days more than 6% glycerol based on the koji wet weight. The content of carbamide in
koji prepared with
R.oryzae IFO 4716 was more than 8 times that of
koji prepared with
Aspergillus oryzae.
