Histochemical studies of the serial sections of the anal canal from 35 human subjects consisting of 14 fetuses, 3 newborn infants, 2 children, and 16 adults, embedding the speci-mens from 32 cases in paraffin and from 3 in carbowax, resulted in the following findings.
1) The epithelial tissues of the anal canal can generally be classified into the following four : (a) single-layered columnar epithelial cells which have a rectal glandular structure and which are present from the Herrmann's line to the anal column, exhibiting great num-bers of goblet cells ; (b) cuboidal epithelial cell tissues in 2 to 4 layers which are present from the vicinity of the pectinate line ; (c) stratified squamous epithelial cell tissues in 10 to 15 layers, , which are present from the vicinity of the base of the crypts ; and (d) ordinary skin tissues which are present outward from the anal groove. Occasionally one or two layers of small cuboidal cells are observed between (a) and (b), and also, in the vicinity of the anal crypts, pseudostratified epithelium whose columnar epithelial cells secreting mucus. The epithelial tissues of the anal canal cannot be indicated by only a single standard type.
2) In the anal crypts, on the upper part, crypt glands are observed which are continuous to that part of the tissues which often has the above mentioned exceptions. In children and adults, many small rectal glands open on the upper portion of the crypts. The central part consists of cuboidal epithelial cells in 2 to 4 layers, and the lower part is covered by stratified squamous epithelial cells.
3) The anal glands generally are in the form of a simple branched tubular gland. Their tegmental epithelium consists of 2 to 6 layers of cuboidal or columnar epithelium.
The tip of the duct has one to 6 acini. All the glands open on the anal crypts.
4) The number of the anal glands in each subject is from one to ten, varying much, with the individual.
The anal glands run underneath the epithelium of the anal-canal, within or through. the sphincter ani internus, and reach the conjoined longitudinal muscle. No anal gland goes outside of the conjoined longitudinal muscle.
Examining 29 subjects whose anal glands were in a relatively good continuity, I found 107 anal glands in all, averaging 3.7 per subject. Of the anal glands, 28.0% was in the anterior quadrant, 22.4% in the left quadrant, 23.4% in the right quadrant, and 26.1% in the posterior quadrant. No erratic distribution of the anal glands to any specific area was observed.
5) The anal glands are capable of secretion. The anal glands are merocrine in 4 to 6 months old fetuses, and have holocrine secretory stage in newborn infants, children, and adults.
In two adults, the columnar epithelial cells of the most superficial layer of the anal glands were observed disintegrated and liberated from the glandular tissues. The cells which were thus disintegrated and liberated in the acini had a nucleus deeply stained by hematoxy-lin ; and the protoplasm was positive for mucicarmine staining, alcian blue staining, and PAS staining. Within the glands were observed secreation positive for myxo-staining and a nucleus in the incipient stage of degeneration.
In four other adults, the secretion within the duct was stained as two different substances, one with a different staining property from the other. In other words, one was positive for nuclear staining, and the other was positive for myxo-staining. The former was stained red by nuclear fast red, red by Masson staining, and red by azan staining. The latter was stained blue by alcian blue, red by mucicarmine, green by Masson staining, and blue by azan staining. PAS staining did not distinguish the two from each other, both being uniformly stained red-violet.

Rectal administration of Indomethacine suppository was employed to treat hemorrhoid concervatively and to remove pain after anal surgery.
Results;
(1) pain ; 1 week after effective in 52% cases and 2 weeks after 80% effective in total.
(2) hemorrhage ; 1 week after effective in 44.4% and 2 weeks after in 76% cases.
(3) effect for prolaps of pile; grade 1-not examined. grade 2-effective in 44.4% cases (1 week after). grade 3-effective in 50% cases (1 week after).
This agent was useful for hemorrhoid.
Immediately after surgery of anal region this suppository was administrated to remove pain and edema. Effectiveness was remarkable, so no narcotics required.

A woman aged 76 years underwent. an operation for hemorrhoids. After a month of the operation I observed malignant tumors in both inguinal regions. The regions had been badly swollen. As a result of pathrogic diagnosis of the extirpative matter, I ascertained it Melanoma. The therapy with 60Co 4600 Rad, carcinostatic drug (Mytomycin 24 mg, 5 FU 5000 mg, Endoxan 1200 mg, Toyomycin 12 mg) did not work well. The Melanoma metasta-sised on the lungs and other organs and it caused her death. I made the report of this case for the rarity of the Melanoma in the digestive system.

It is important to evaluate the clinical response of medical treatment by means of standardized method in comparison between each different kind of drugs. An attempt to standardize the clinical evaluation of treatment of hemorrhoids was made. The principle of method was to give marks to each symptom of hemorrhoids according to their severity and final evaluation was made by means of percentage of demerit marks between pre-and post-treatment.
Results of oral administration of Fragivix, which is a new oral drug for hemorrhoids, with its double blind test are reported.
