    Oxygen uptake of the microorganisms cultured from river water were examined to find the effect of heavy metals with or without a substrate. Without the addition of a substrate, effect of a metal was examined in median toxic concentrations of oxygen uptake (TC50), and the metal toxicity decreased in the order of Hg2+, Cu2+, Ni2+, Cd2+, Zn2+, Al3+, Pb2+, Fe3+, Mn2+. When a substrate was added, the effect of metals appeared at a higher concentration. The modified Wiegand equation was applied to analyze the form of oxygen uptake inhibition caused by cadmium. The decrease of L and y0 values was indicated without substrate added, and when the substrate was added, to the effect of cadmium was different from the former. Dehydrogenase activity was related the decrease of oxygen uptake in the absence of a substrate.

    Gas-liquid chromatographic determination of nanogram quantities of chlorpheniramine maleate (CP-M) and dl-methylephedrine hydrochloride (ME-HCl) in antipyretic and analgesic preparations was examined. Prescribed amounts of CP-M and ME-HCl in the preparations are 0.4-1.5 mg and 2-5 mg, respectively. Rubidium chloride alkali flame ionization detector was more sensitive for nitrogencontaining compounds than normal flame ionization detector and could easily detect 40 ng of CP-M which showed 30% of the peak height of full scale without base line drift or noise. Each base was extracted from alkaline aqueous solution with benzene containing internal standard. It Noscapin was prescribed for the preparation, it was necessary to decompose it in sodium hydroxide solution by warming at 50°for 30 min and extraction with benzene because Noscapin has a long retention time and interferes the routine analysis of CP-M and ME-HCl. Benzene layer (1.0 μl) was injected into the gas chromatograph. Internal standards used were N-butyryl-o-toluamide for ME-HCl and diphenylpyraline for CP-M. Gas chromatographic conditions were : 3.5% JXR on Chromosorb G (80-100 mesh, acid-washed and silane-treated), glass column (2.5 mm×2 m) at a temperature of 180°, nitrogen as a carrier gas at the flow rate of 30 ml/min, and using RbCl alkali flame ionization detector.

    Gas-liquid chromatographic determination of nanogram quantities of p-chlorophenol (PCP) in Clofibrate capsule was examined. Clofibrate in capsule preparations contains parabens which are added as a preservative to the capsules made of gelatin. J. P. IX method is necessary to separate PCP and Clofibrate-parabens before color reaction and is very time consuming. This proposed method utilizes derivatization of PCP and parabens to their diethylphosphate esters with diethyl chlorophosphate and sodium methoxide in hexane at 50°for 30 min in the presence of Clofibrate. The diethylphosphate esters are determined by flame photometric detector (FPD) and flame thermionic detector (FTD) on a column (2.5mm×2m) of 3.5% silicone JXR on Chromosorb G. HP (80-100 mesh), at 190°, nitrogen as a carrier gas at the flow rate of 30 ml/min. Clofibrate, PCP-diethylphosphate ester, and paraben-diethylphosphate esters were separated under this condition. PCP was determined by the internal standard method using p-bromophenol diethylphosphate ester. Coefficient of variation of peak height ratio for 3.87, 5.80, and 7.74 μg of PCP in 10 ml were 0.87%, 0.56%, and 0.56% with FTD, and 1.50%, 0.57%, and 0.56% with FPD, respectively. PCP in commercial preparations was 1.6 to 5.1 ppm.

    The changes of lipid metabolism in carp caused by feeding a PCB diet were as follows. 1) In in vivo experiments, total lipid and triglyceride in the carp hepatopancreas increased slightly, and glycogen decreased. 2) In in vivo experiment, the rate of incorporation of 1-14C-palmitate into the hepatopancreatic lipid fractions did not change, but the incorporation of radioactivity into blood-plasma lipid increased in the triglyceride and free fatty acid fractions, and decreased in the phospholipid and free cholesterol fractions. 3) In in vitro experiments, the incorporation of 1-14C-palmitate into lipids of the hepatopancreas increased slightly in the triglyceride fraction, especially in the α, α'-position of the triglyceride. 4) The triglyceride synthetase and α-glycerophosphate dehydrogenase activities in hepatopancreas were slightly increased by PCB administration. The changes of lipid metabolism in the carp hepatopancreas on PCB administration are discussed in comparison with those in mammals.

    A sensitive method for the quantitative determination of volatile halogenated hydrocarbons in water is described. Water sample is purged with nitrogen gas, and the gas is passed through a column of an adsorbent material (Tenax GC). The column traps and concentrates the volatile halogenated hydrocarbons, which are then desorbed from the column by heating under a gas flow. The desorbed halogenated hydrocarbones are quantitatively determined by a gas chromatograph equipped with an electron capture detecter. By this method, 0.1 μg/l of volatile halogenated hydrocarbons, such as chloroform, carbon tetrachloride, 1, 1, 1-trichloroethane, monobromodichloromethane, trichloroethylene, and tetrachloroethylene, can be determined quantitatively.

    A simple colorimetric method for the determination of cannabinoids was developed, using Fast Blue BB salt. Fast Blue BB salt produced stable diazo compounds with cannabinoids in alkaline media. The absorption maxima of Δ8-tetrahydrocannabinol, Δ9-tetrahydrocannabinol, cannabidiol and cannabinol were at 488, 489, 470 and 490 nm, respectively. The molar extinction coefficients of the above cannabinoids were estimated to be 3.3, 2.9, 5.2 and 2.5×104, respectively, and the coefficients of variation (10 analyses) were 0.7, 0.4, 0.4 and 2.7 (%), respectively. The contents of these cannabinoids in extracts from six kinds of cannabis leaves were determined by the colorimetric method in combination with thin-layer chromatography. The results obtained were in good agreement with those obtained by the gas chromatographic method.

    Upon single oral administration of 2-isopropylnaphthalene (2-IPN) (100 mg/kg) to rats, more than 95% of the dose was absorbed from the gastrointestinal tract within 24 hr. The maximum levels of 2-IPN in the blood, liver, kidneys, brain, heart, spleen and muscle were observed at 2 hr after the administration and then the levels decreased with time. However, the times taken to reach the maximum levels of 2-IPN in the adipose tissue and skin were longer than those in other tissues. The content and disappearance of 2-IPN in the adipose tissue were higher and slower, respectively, than those in other tissues. In addition, an extremely small amount of unchanged 2-IPN was excreted in the urine. In rats fed on the experimental diet containing 0.1% 2-IPN for 14 or 28 days, the 2-IPN content in the adipose tissue was much greater than that in other tissues. In the case of administration of the control diet for 1-21 days after feeding the experimental diet for 28 days, the 2-IPN contents in the adipose tissue and skin decreased progressively, However, the content and disappearance of 2-IPN in the adipose tissue or skin after continuous administration of 2-IPN were much lower and more rapid, respectively, than those of 2, 6-diisopropylnaphthalene. This suggests that the tissue accumulation of 2-IPN is low.

    Polychlorinated naphthalenes in water samples collected at 21 points in Tokyo Bay, Sagami Bay, Nagoya harbour, Osaka Bay, and the Kii channel and its offing were determined by gas-liquid chromatography after concentration procedure using dodecylbenzenesulfonic acid-binding polymide column and the separation from polychlorinated biphenyls by polyamide thin-layer chromatography. The levels in most samples ranged from 2 to 20 ng/l, and the samples collected near land showed higher concentration suggesting that the contamination was associated with those in the river. The concentration in the sea water in Tokyo Bay was 1/100 to 1/600 of that in the fish taken from the Bay.

    Relationship between the pH and the rate of nitrification in aeration tank for activated sludge was investigated. 1) For the determination of pH and nitrogen compounds of mixed liquor in the aeration tank, samples must be filtered at each sampling time. 2) The pH of the mixed liquor in the aeration tank decreased with increasing rate of nitrification, and increased with decreasing rate of nitrification. 3) While the nitrification was in progress. pH decreased in the order of the mlet, middle, and outlet of the aeration tank. 4) There was a good negative correlation between the rate of nitrification and the pH of the mixed liquor in the aeration tank. It was suggested that the pH could be used as an index to estimate the rate of nitrification of the mixed liquor in the aeration tank.

    A sensitive method was developed for the determination of methyl chloride in ambiant air. Air was passed through the concentration tube packed with Silicone DC-200 (supporter : Chromosorb WAW, DMCS) and chilled at -72°, and the concentrate was introduced into a gas chromatograph by raising temperature of the tube. Two liters of ambient air can be analyzed by this method and detection limit of methyl chloride was 11.9 ppb.

    We analyzed Cd contents in more than 500 samples of foods purchased in Tokyo and the Osaka-Kobe area, and estimated the general contents in foods. The highest value was obtained in seaweeds (0.36 ppm) and high contents were found in shellfish, squid, "Tsukudani" of fish and wheat germ. Other foods contained less than 0.07 ppm. The average uptake of Cd from foods was calculated to be about 40 μg/day/man, by multiplying the values estimated by averaging the general contents in similar foods by the amounts of daily uptake of the various food groups.
