Garcinia extract, which is extracted from the rind of Garcinia cambogia, is available as an herbal supplement and is used to reduce weight. Ca-type Garcinia extract (GPCR) was reported to harm testis of rats fed at a dose of 5.0% as a dietary administration. This study was investigated effects of zinc supplementation on improvement of testicular toxicity in F344/DuCrj male rats fed 5% GPCR containing diet. In 5% of GPCR given group, slight atrophy of spermatids were observed. On the contrary, no testicular toxicological changes were observed in zinc supplemented groups. Serum calcium levels were significantly increased in GPCR given groups than that in control. Zinc level was reduced in 5% GPCR given group compared to control, but not in 0.01 % zinc supplemented group. Mg, Fe, Na and K levels in serum were not changed. The level of inhibin B in serum was reduced in GPCR given group compared to control, but not in zinc supplemented group. As testicular toxicity of GPCR in F344 male rats was prevented by zinc supplementation of diet, zinc deficiency in diet may induce the testicular toxicity of GPCR. The present study suggested inhibin B measurements may provide a useful tool to investigate the testicular toxicity by GPCR.

F344/DuCrj(Fischer)系ラットを用いて、マクロホモプシスガムの28日間反復投与毒性試験を飼料中濃度0, 1.25, 2.5および5.0%で実施した。マクロホモプシスガム投与の影響として以下の変化が観察された。食餌効率では、雌雄の5.0%投与群で低値傾向を認めた。血液学的検査では、白血球百分率において分節好中球の低値を雄の全投与群で認めた。また、プロトロンビン時間の延長を雄の2.5%および5.0%群で認めた。血液生化学検査では、総ビリルビンの軽度な高値を雌の2.5%および5.0%投与群で認めた。総コレステロールの低値を雄の5.0%投与群および雌の2.5%投与群で、リン脂質の低位を雄の5.0%投与群で、総蛋白の低値を雄の2.5%および5.0%投与群で、アルブミンの低値を雄の2.5%投与群でそれぞれ軽度に認めた。しかし、これらは毒性学的影響のある変動とは考えられなかった。臨床症状、生存率、体重、摂餌量、摂水量、尿検査、眼科学的検査、肉眼的病理学検査、器官重量および病理組織学的検査ではマクロホモプシスガム投与の影響を認めなかった。以上の結果より、無毒性量(NOAEL)は、5.0% (雄で3956mg/kg/day、雌で4257mg/kg/day)であった。

"Eucalyptus leaf extract", a natural food additive, is commercially available as steam distillate, or an ethanol extract of eucalyptus (Eucalyptus globulus LABILL) leaf and is officially approved in the "Lists of Existing Food Additives in Japan". In our ongoing study to evaluate its quality and safety as a food additive, major components in "Eucalyptus leaf extract" were purified and characterized. Five major compounds were purified from "Eucalyptus leaf extract" by the preparative HPLC. Each component was identified as gallic acid, quercetin 3-O-β-D-glucuronide, kaempferol 3-0-β-D-glucuronide, 2, 4-dihydroxy-3-(3-methyl-l-butenyl)-5-methylbenzaldehyde 6-O-β-D-glucopyranoside, named globuluside and macrocarpal I by NMR and FAB-MS. A novel compound, globuluside, was newly purified and characterized from naturally occurring materials on the basis of the spectral evidence.

ネオマイシンホスホトランスフェラーゼII(NPTII)タンパク質を特異的に検出するELISAシステムを組換え大腸菌で大量発現したNPTIIタンパク質に対する抗体を使用して構築した。このシステムを用いることで、遺伝子組換えタバコで発現しているNPTIIタンパク質を定性的に検出することができた。一方、遺伝子組換えしていないタバコ、ダイズ、トウモロコシ、ジャガイモでは非特異的な検出は認められなかった。これらの結果から、本研究で構築したELISAシステムは、遺伝子組換え食品中のNPTIIタンパク質のモニタリングに適用可能であることが示唆される。

Genetic and epidemiological studies on alcohol tolerance frequencies were conducted by analyzing genetic materials from 149 healthy female students. Aldehyde dehydrogenase 2 (ALDH2) gene was characterized by the combination of polymerase chain reaction and agarose gel electrophoresis of respective DNA specimen. Nail clippings were found to be relatively easy to collect and extract DNA from volunteers especially when they were restricted to female students. Plucking hair roots was not successful, since it caused sometimes pains on the volunteers. Among 149 individuals tested, 65.6% were the typical or normal homozygote, 32.2% were the heterozygote, and 2.0% were the atypical mutant homozygote. The reliability of the scale ratings using ALDH2 genotype analysis was assessed and confirmed by ethanol patch test and other psychiatrist's evaluation methods, such as TAST (Tokyo-University ALDH2-Phenotype Screening Test). However, the genotype analysis is not directly associated with AAIS (Adolescent Alcohol Involvement Scale). Accordingly, careful case-control study must be undertaken on genotype analysis and it must be accompanied by several combination of epidemiological studies.

酵素処理イソクエルシトリン(EMI)の経口摂取後の代謝挙動を明らかにする目的で、主8成分(IQC, IG₁〜IG₇)各々について、唾液α-アミラーゼ、膵液α-アミラーゼ、人工胃液存在下での反応生成物をHPLCを用いて定量解析した。主8成分のうち、イソクエルシトリン(IQC)、IG₁およびIG₂は、いずれのα-アミラーゼによる加水分解をほとんど受けないことが判明した。一方、IG₃からIG₇については、生体内での滞留時間を考慮した場合、ほぼ完全にIQC,IG₁,IG₂へと代謝されることが明らかとなった。また、EMIのいずれの成分も、人工胃液および唾液アミラーゼ含有人工胃液処理においては、まったく代謝を受けないことが判明した。これら一連の結果から、経口摂取されたEMIは、唾液、膵液アミラーゼの作用で小腸に移行される前の段階で、IQC, IG₁, IG₂に集約されることが明らかとなった。

We developed a rapid method to determine the concentrations of formaldehyde (FA) and acetaldehyde (AA) in polyethylene terephthalate (PET) resin of PET bottle. PET samples were ground with liquid nitrogen and heated with water at 120℃ for 60minutes. FA and AA in the extract solution was derivatized with 2, 4-Dinitrophenylhydrazine (DNPH) and quantified by High Performance Liquid Chromatography (HPLC) with good accuracy (accuracy for 5 repetitions ; CV (%) : FA, 2.0 ; AA, 1.4). When we compare the data of AA, we had to separate them into the bottle types, thermostable or non-thermostable. Because the bottle types influenced the data of AA. But they didn't influence the data of FA.

アントシアニン高含有アカダイコン(Raphanus sativus L.)の育種研究の一環として、種子にγ線を各々200Gy、400Gy、600Gyの線量で照射した3集団とγ線無照射の集団を秋期栽培し、収穫個体のアントシアニン含量をHPLCにより定量し、放射線照射の生育およびアントシアニン産生能への影響を検討した。この結果、200Gy照射集団は、γ線無照射集団と比較して、アントシアニン含量が単位体積あたりと個体あたりどちらについても有意に増加し、個体体積もほとんど変動しなかった。さらに、種子に対するγ線照射は、いずれの強度においてもアカダイコン中のアントシアニン組成にほとんど影響を与えなかった。したがって、200Gy照射はアントシアニン高含有アカダイコンの育種に有効な手法となる可能性のあることが示された。
