    We investigated changes in the glycosaminoglycans (GAGs) during progression of a human gingival carcinoma xenograft line, GK-1, in nude mice. The GAGs extracted from cancers 3, 5, 7, 10 and 15 weeks after transplantation consisted of hyaluronic acid (HA), chondroitin sulfate (CS) and heparan sulfate (HS) as major components, and dermatan sulfate (DS) as a trace component for all cancers. HPLC analysis revealed that the HA content per defatted tissue dry weight increased in the cancers 5 weeks after transplantation compared to those of 3 weeks (p<0.05), while CS for cancers at 10 weeks decreased compared with 7 weeks (p<0.05). However, HS showed no significant change. Both the CS and DS contained primarily 4-sulfated disaccharide units. Immunohistochemical staining with antibody 2-B-6 for the PGs having ΔDi-4S produced by chondroitinase ABC digestion showed that CS is located in the tissue surrounding the cancer nests and mass.
    These results indicate that the location of accumulation of CS, which primarily contains 4-sulfated disaccharide units, plays an important role in cancer progression.

    Treatment of human leukemia HL-60 cells with ceramide, a breakdown product of sphingomyelin, induced both programed cell death (“apoptosis”), and cellular differentiation. Apoptosis in response to ceramide occurred in a concentration-dependent manner. Apoptosis induced by ceramide in HL-60 cells requires the presence of c-jun protooncogene. However apoptosis is inhibited by curcumin, a specific inhibitor of c-Jun/AP-1. Whereas curcumin restores ability of inhibited cells to grow, it does not affect ceramide-induced differentiation. These results indicate that ceramide controls cell differentiation and proliferation through apoptosis by activating the nuclear transcriptional factor AP-1. Further, AP-1 is apparently more closely related to apoptosis-inducing signal transduction pathway than to the pathway leading to cellular differentiation.

    We observed vertical motion of the tongue during mastication in ultrasonographic images of 6 males with an average age of 28.7 years. All subjects had complete dentition and no evidence of neurological disorders. We used a 7MHz mechanical convex scanning transducer to obtain a continuous, real-time ultrasonic image of the tongue, and carried out real-time B-mode ultrasonic observation at the median sagittal plane of the mandible. We recorded vertical motion of the tongue in the M-mode on a line connecting the mandibular first molars. The transducer produced good quality images of the dorsal surface during mastication of peanuts, rice crackers, boiled fish paste, pickled radishes, pudding and bananas. The transducer, which was small and light, permitted capture of the images on videotape without manual placement of the scanner. Continuous images showed that the tongue turned the food, mixed it with saliva, sorted out unsuitable particles and aided in bolus formation. Vertical motion of the tongue had two phases, sorting and bolus formation.
