This study has been done in order to clarify the effect of Euvestin (diethylstilbestrol) sol instead of Euvestin oil for the convenience of practical application, in the case of induction of lactation in goats, which has been studied for the test of the possibility of prediction of dairy performance in dairy animals.
The object of this report is to present one modification of biological assay method of small amount of estrogen in the blood, and the changes of estrogen level in the blood of goatlings treated with Euvestin sol or oil, measured by this method.
Three Saanen and one Alpine goatlings were divided into two groups. The treatments are as follows; animals of group I were injected intramuscularly with 2.5 mg Euvestin sol twice every 10 day. After 13 days from the 2 nd injection, 20 mg Euvestin sol was added two times with 10 days interval. Those of group II were injected subcutaneously with 0.25 mg Euvestin oil daily for 20 days.
The results obtained are as follows:
(1) Our assay method was almost similar to that of FUJII
et al. (1955), which was one of localvaginal response methods. But in considering the persistency of the effect of a test substance and the simplicity of judging so on, a little modification was done ; 48, 60 and 72 hours after the 1st insertion of the test substance to the vagina, smears were tested and the points of 3, 2, 1 and 0 were given to the signs of ( ?? ), (+), (±) and (-)respectively. Then, the index of the response was determined as the average of the scores of mice for three periods calculated.
According to this method, 2.44 was equal to 1 Vaginal Unit of ALLEN-DOISY Method.
(2) The changes of the level of estrogen in the blood of Euvestin treated goatlings were very fast. The estrogen level of the animals of group I was maximum on the 6 th day after the 1st injection and falled down rapidly even after the 2 nd injection. Those of group II was maximum on the 15 th day after the 1st injection and afterwards failed down in spite of the continuous injection for 20 days.
Further studies may be necessary for clarifying these phenomena mentioned above.
