    In combined therapy, time sequence and time interval of the two kinds of treatment must be in mind. Mechanism of these action is not similar, accordingly cell cycle dependent sensitivity and cell group affected should be different. MTX (0.04mg) and irradiation (200R) were administered to solid tumor growing in Donryu rat by inoculation of Yoshida ascites tumor cells. Various combination of the two treatments were administered in respect to their order and interval. DNA synthesis rate and change of radioactivity in both periphery and center of solid tumor treated by MTX administration or irradiation were studied by 125I-UdR labelling.
    As the results, combined effects were in the order 1) administration of MTX on 3 days after irradiation, 2) simultaneous administration, 3) irradiation on 3 days after administration of MTX. On the other hand, the radioactivity of treated tumors in periphery or center were different in comparison with nontreated tumor. From these points, tumor cells might be not homogenous in solid tumor.
    These suggested that in fractional course of administration in combined therapy, simultaneous use was practicable, because daily irradiation and 2 times a week of the drug in clinical use could not demand the time sequence of the two treatments.

    The incorporation of labeled precursors into the DNA is often believed to be the indicator of the DNA synthetic rate. However, the detailed kinetic changes in incorporation immediately following irradiation has been not observed.
    In this experiments, the radiation-induced changes of DNA precursor incorporation (3H-TdR) was pursued concerning the labeled index and the grain count in labeled tumor cells (Yoshida ascites tumor) .
    As the result,
    1) Incorporation immediately after irradiation (500-1000R) was a dose-independent phenomenon, namely irradiation has no influence upon the thymidine incorporation into DNA injected immediately following irradiation.
    2) In irradiated early S-phase cell (or late G1-phase cell), it was likely that TMP de novo synthesis was inhibited, and salvage was increased.

    It was expected that radiation damages might be protected by THFA which was a precousor of folic acid and a easy oxydizable substance.
    These experiments were carried out to find the protective effect of THFA on both yeast cells and mice.
    As a results, it was found in yeast cells that THFA (10-5M) was effective in pretreatment but ineffective in post-treatment. There was no correlation between the protective effect and a labelled THFA concentration per cell.
    On the other hand, the number of mice surviving after 30 days increased significantly even in past-treatment, in which THFA (0.5mg) was injected intraperitoneally at 6 h. or 24 h. after irradiation. (500R or 750R)
    From these findings, it was supposed that THFA would effect as a reductive agents in yeast cells and as a promoter to the recovery of bone marrow cells resulting the decrease of bone marrow death in mice.

    That the bone marrow gets purine from liver has been claimed by Lajtha. 2-mercap-topropionyl-glycine (MPG), containing SH in the molecule is expected to protect the liver from radiation damage given prior to liver irradiation. However, that liver disorders of various kinds have been ameliorated by MPG has a good reason the drug to ameliorate radiation-induced liver disorder though given after irradiation.
    Some experiments were carried out and following results were obtained. Decreased DNA synthetic rate in bone marrow and prolonged BSP clearance rate after liver irradiation were improved by MPG administration.

    Xe clearance measurement is available in clinical practice to know the blood flow of tumor to some extent. Yoshida ascite tumor cells were trnsplanted in the subcutane of Donryu rat. Correlation between tumor size, Xe cleance, and tumor capillary pattern was investigated with regard to irradiation. Tumor smaller than 1 cm in diameter revealed good blood flow as well as. homogenously distributed capillary. However that of more than 2 cm contained necrotic lesion in its center without capillary. In accordance with this clearance from the center was badd contrary to that from the periphery which was good.
    Tumors of 2 cm in size irradiated with various single dose or f ractionted dose. The time required to reduce the tumor size to 1 cm was dependent to dose or fractionation. However these 1 cm tumors were worse than non-irradiated 1 cm tumor in blood flow as well as in capillary distribution in the center. These findings strongly suggested the parallelity of blood flow and capillary distribution.
