The author carried out analysis of free amino acids, obtaining samples from fifty six infertile patients. Analysis of free amino acids in normal human semen were also conducted, the samples of which were obtained from five healthy males before and after they were vasectomized.
1. The samples were divided into three groups in accordance with sperm counts in semen and compared with each other.
The first group : sperm counts-over 40×10
6/cc
The second group : sperm counts-below 40×10
6/cc
The third group : azoospermia
In regard to the total quantity of amino acids, the second (593.3 mg/dl) and the third (610.6 mg/dl) groups showed lower value than the first group (826.3 mg/dl) with significant difference. As to the quantity of each amino acid, there were significant differences among sixteen kinds of amino acids in the first and the second groups, excepting Cysteic acid and Methionine. In the third group, twelve kinds of amino acids showed significant difference compared with the first group, excepting Lysine, Histidine, Arginine, Serine, Isoleucine and Tyrosine.
2. The samples were divided into three groups according to percentage of motile sperm in seminal fluid.
The first group : percentage of motile sperm-over 60%
The second group : percentage of motile sperm-below 60%
The third group : azoospermia
As to the total quantity of amino acids, the second (548.8 mg/dl) and the third (610.6 mg/dl) groups held lower value with significant difference than the the first group (822.7 mg/dl) . As to the quantity of each amino acid, significant difference was recognized in sixteen kinds of amino acids of the second group except in Taurine and Methionine, compared with the first group. The third group also showed low value with significant difference in thirteen kinds, excepting Lysine, Histidine, Arginine, Cysteic acid and Methionine.
3. The samples were divided into five groups according to sperm counts and percentage of motile sperm.
The first group : sperm counts-over 40×10
6/cc, motility-over 60%
The second group : sperm counts-over 40×10
6/cc, motility-below 60%
The third group : sperm counts-below 40×10
6/cc, motility-over 60%
The fourth group : sperm counts-below 40×10
6/cc, motility-below 60%
The fifth group : azoospermia
As to the total quantity of amino acids, the last four groups (2 nd group : 512.1 mg/dl, 3 rd group : 646.9 mg/dl, 4 th group : 561.1 mg/dl, and 5 th group : 610.6 mg/dl) showed low value with significant difference compared with the first group (872.2 mg/dl) . As to the quantity of each amino acid, twelve kinds of amino acids of the second group showed lower value with significant difference than those of the first group, except Lysine, Histidine, Cysteic acid, Taurine, Proline and Methionine. Eight kinds of amino acids of the third group also showed lower value with significant difference than those of the first group, except Cysteic acid, Taurine, Threonine, Serine, Valine, Methionine, Isoleucine, Leucine, Tyrosine and Phenylalanine. In the fourth group, seventeen kinds except Methionine, and in the fifth group, fifteen kinds except Lysine, Histidine, Arginine showed low value with significant difference, compared with those of the first group.
4. Amino acids in normal human semen obtained before and after vasectomy were examined. The quantity of total amino acids before vasectomy was 667.6 mg/dl on the average, while it was 594.2 mg/dl after vasectomy. As to each amino acid, Methionine showed 5.1 mg/dl before vasectomy and 1.6 mg/dl after vasectomy, but others showed various values.
As the results, these analysis seem to suggest that not only the function of testicle but also the function of accessory sex organs are incomplete in the abnormal human semen as far as amino acids are concerned.
