    In amyloidosis of various animal species, the changes in affinity for congo red after the pottasium permanganate treatment were compared with the amyloid protein determined by the immunohistochemical technique. Many cases examined showed sensitivity regarding the potassium permanganate treatment losing affinity for congo red (cows: 21/27; cats: 1/6; pigs: 2/2;dogs: 1/1; rabbits: 1/1; mice: 0/1; birds: 6/8) and reacted positively to the anti-AA antibody. Nearly all of the other cases showing some resistance to the potassium permanganatetreatment and retaining affinity for congo red (cows: 6/27; cats: 5/6; mice: 1/1; birds: 2/8) also reacted positive for the anti-AA antibody (cows: 6/6; cats: 5/5; mice: 0/1; birds: 1/2). “Ham spleen” in rabbits and cats showed an AA-protein deposit in contrastto the AL-protein in human beings. From these facts, it seems to be difficult to determine amyloid protein in animals using the pottasium permanganate treatment and the pattern of amyloid deposit.

    An attenuated Aujeszky's disease virus (ADV) vaccine with deletions in thymidine kinase andglycoproteing III genes was evaluated for an experimental dual infection with ADV and Actinobacillus pleuropneumoniae in specificpathogen-free pigs. Pigs were assigned to vaccinated and unvaccinated control groups. Four of the 8 pigs (4 weeks old) were vaccinated with the attenuated ADV vaccine, and the remaining pigs were not vaccinated. All pigs wereinoculated intranasally with ADV (106.0TCID50/ head) and serotype 1ofA. pleuropneumoniae (106.2 CFU/ head) at 7 and 8 weeks respectively.The duration and severity of clinical signs ofvaccinated pigs after challenge with ADV and A. pleuropneumoniae were obviously reduced, compared with those not vaccinated. Two out of 4 unvaccinated control pigs died during the experiment, but none of the vaccinated pigs did. These results indicate the efficacy of the vaccine on the dual infection with ADV and A. pleuropneumoniae.

    Four hundred three (20%) out of 2012 piglets died during the January-March period of 1990 on a pig farm in Chiba Prefecture. The disease almost exclusively affected newborn pigletswithin the 1st week of life, especially in the first 3 days. Main clinical signs were mucohemorrhagic diarrhea and dehydration. Autopsy revealed hemorrhagic enteritis in the small intestine, particulary in the jejunum, showing necrosis and hemorrhage. Clostridium perfringens was isolated from all the intestines tested on the order of 107 to108 CFU/g. Toxinsdetected in the intestinal contents of three piglets and in bacterial cultures were of C. perfringens type C. The isolates were highly sensitive to penicillin and ampicillin.

    Twelve cases of feline thymic lymphoma were studied clinicopathologically. The cats in this study ranged from 8 months to 8 years in age (mean: 2.3). Hematological abnormalities included anemia (2 cats, 16.7%), leukopenia (1 cat, 8.3%) and leukocytosis (4 cats, 33.3%). Neoplastic cells in the peripheral blood were observed in 6 cats (50%). During the serum investigation, hypogammaglobulinemia (5 cats, 50%) and elevated lactate dehydrogenase (LDH)(6 cats, 67%) were observed. Since all 6 cats with elevated serum LDH levels had neoplastic cells in their peripheral blood, there seemed to be a correlation between the elevation of the serum LDH level and blood involvement. According to the WHO clinical staging, these coses were categorized into stage I or II (3 cats), stageIV (3 cats) and stage V (6 cats). Nineof the 10 cats treated with induction chemotherapy experienced a remission. Survival time ranged from 10 to 330 days (median: 69 days).     A total of 58 domestic bitchs were subcutaneously injected with proligestone (S-C-39) fromone to seven times. To investigate the effects of estrous control, harmful aftereffects, changes in clinical findings and blood characteristics, and return to estrus after the final injection were observed once a month during the study. Plasma levels of progesterone were measured by radioimmunoassay once a month. Due to the results obtained from clinical findings and plasma progesterone levels, it could be seen that estrus and ovulation were suppressed in 35 bitchs out of 38 (92.1%) injected four times or more with proligestone. Amongthe remaining three bitchs (7.9%), estrus was not fully suppressed, namely two showed heatjust before the 4th injection. In addition, in one, it seemed that estrus would return dueto the temporary high levels of plasma progesterone, although no visible signs of estrous behavior could be observed. Abnormal findings were noted in three cases: the first case with maceration of fetus, the second case with pyometra-like syndrome and the final case with abortion detected during injections. However, the former two cases may have been affected before the first injection. Estrus did not return in 30 bitchs out of 40 during more than 5 months after the final injection. In the remaining 10 bitchs, estrus returned after 7.6 months on the average. The present results demonstrate that proligestone is sufficient for estrous control without any harmful aftereffects in bitchs.

    Twenty-two dogs with allergic dermatitis, which are evaluated from physical findings, and 30 normal dogs were tested for this allergy using the immediate type-intradermal test with25 allergens, including house dust, red pine pollen, Japanese ceder pollen, ragweed pollen, wheat flour, cow's milk, whole eggs, egg yolks, egg whites, beef, pork, chicken, yeast, dog hair, cat hair, chicken feathers, sheep hair, cigarette smoke, kapok, Alternaria, Cladosporium, Aspergillus, Candida, Penicillium, and fleas, with both of the histamine phosphates diluted 1: 10, 000 as the positive control and 0.5% phenol-saline as the negative control. The tests were read 15 min after injection. According to fundamental study for standardization of criterion of the positive response, a positive reaction was determined when the diameter of the wheal at the injection site was larger than the median value between the positive and negative controls. Eleven of the 22 dogs with dermatitis were positive for fleas and one for both fleas and house dust. In 4 of the 30 normal dogs, 2 dogs werepositive for fleas and one for ragweed and another one for Candida. The histological examination showed marked perivascular infiltrations of basophils, mast cells and eosinophils in the positive site for flea allergen.

    Blastogenesis of canine peripheral lymphocytes was measured by the MTT (3-(4, 5-dimethylthiazole-2-yl)-2, 5-diphenyl tetrazolium bromide) colorimetric assay. MTT is cleaved only by living cells and formazan is produced. This process needs active mitochondria, and even freshly dead cells do not cleave MTT. Canine peripheral lymphocytes prepared by Ficoll-Conray gradient were cultured with lectins (PHA and Con A) for 96 h at 37°C under 5% CO2 using the microplate system. At the end of the culture, the MTT reagent and HC1-isopropanol were added to the culture, and then, the absorbance of each culture was measured by a microplate reader using a test wavelength of 570 nm, and a reference wavelength of655 nm. The mean value of SI (stimulation index) of canine lymphocytes from 14 normal dogswas 183±130 in PHA stimulated culture and 131±82 in Con A stimulated culture. The administration of predonisolone (5 mg/kg) to 3 normal dogs at 6 times over 3 days depressed SI from 69±31 to15±9 in PHA, from 70±14 to 6±4 in Con A. In conclusion, the MTT assay provides an efficient evaluation method for lymphocyte blastogenesis in dog in vitro.

    Neutrophil adhesion in healthy dogs was evaluated using a coverslip method. Adhesion was revealed as the adhesive index calculated from the neutrophil numbers on a coverslip and those in the blood sample. The adhesive index decreased significantly after ethanol was added to the blood sample. The results suggested that the evaluation of canine neutrophil adhesion was possible using this method. The adhesive index in 46 healthy dogs was 38.3±11.6 (mean±S.D.). As this coverslip method is simple and rapid with no special equipment needed, it may be useful in assessing the functions of canine neutrophils.

    dead wild mallard was found on a bank in Ikoino-Mori located in Komatsu-shi in November 1991. At necropsy and X-ray examination, 144 spent lead shots were recovered from its gizzard. The lead levels of the liver, kidney and skeletal muscle of the bird exceeded the probable toxic level for a normal mallard. These findings indicated that the bird might have succumbed to lead poisoning. To confirm this, another mallard duck was forced to eat 100 lead shots. The bird showed nervous signs and green diarrhea, and died after 6 days. The necropsy findings and the lead levels of the tissues of the bird were compared with those of the wild mallard.
