Levels of serum immunoglobulins, complement and macrophage activity were compared between plaque-susceptible (SUS) and plaque-resistant (RES, control) rats. SUS rats with pregingivitis showed the significant lower levels in serum immunoglobulins than those in RES rats, especially at early months of age. C3 level and complement hemolytic activity in SUS rats were also lower than those of RES rats at the same age. IgA level and complement hemolytic activity in SUS rats remained at low levels throughout the observed period although the levels of the other immunoglobulins increased gradually thereafter. C3 levels in SUS rats aged 2 to 7 months were higher than that in RES control, while complement hemolytic activity was always lower than that in RES rats. Antibody response against sheep red blood cells (SRBC) was depressed in young SUS rats. The observations suggest that some abnormalities of immaturities in the humoral immune system are present in young SUS rats before occurrence of gingivitis. The susceptibility to periodontal lesions may be based on these abnormalities.

For establishment of early diagnosis of enzootic bovine leukosis (EBL), the complement-dependent antibody cytotoxicity (CDAC) test using monoclonal antibody against tumor-associated antigen (TAA) expressed on EBL tumor cells, lymphocyte counts, lactate dehydrogenase (LDH) activities and concentration of serum sialic acid (SA) were examined in clinical subclinical and EBL cases. In TAA-positive clinical and subclinical cows, lymphocyte counts, total LDH, LDH2 and LDH3 activities and concentration of SA increased, whereas LDH1 activity decreased from those of TAA-negative healthy cattle. The cytotoxic index (CI) values of the CDAC test from individual cows examined were compared with the results of other clinical examinations. Significant correlation coefficients were observed between CI values and the lymphocyte counts, total LDH, LDH1 and LDH2 activities and concentration of SA (p<0.05-0.005).

Using male Wistar rats, the bipolar limb lead electrocardiogram (ECG), the vectorcardiogram (VCG), the unipolar chest lead ECG and the unipolar lead electrogram (EG) from the cardiac ventricular surface were recorded before and after the procedure of a hypothermic (30°C-25°C) condition and/or the administration of isoproterenol (1.5 or 3.0μg/kg, i. v.). The hypothermia caused a sudden prolongation of QT interval with an increased heart rate which was followed by the bifid change of T wave in the record of limb lead ECG, but the vectorcardiographical changes of T loops occurred only in the frontal and horizontal planes, with no marked change in the figures of QRS loops. Bifid T waves were constantly observed either in the precordial lead ECG or the cardiac surface lead EG when the lead positions were on the sternal line or near the cardiac septal area. By the hypothermic or the isoproterenol treatment, bifid T waves could be recorded from much wider area. These results suggested that the bifidity of the T wave recorded in the limb lead ECG is due to locally limited electrical events which can be magnified or exaggerated by hypothermia or sympathetic tension.

-3 contents of digesta in the stomach, the small intestine, the caecum, the colon were determined in the rats to study the effects of pH on absorption of Mg, Ca and P in the gastro-intestinal tract. Acidification in the stomach (pH3.46), sudden pH rise in the upper segment of the small intestine (pH6.52), sudden alkalization from pH6.89 to pH7.41 in the defined site of the lower segment of the small intestine, acidification in the caecum (pH6.91) and sudden alkalization in the colon (pH7.97) were observed. The positive correlations among Cl
-, HCO
-3, Na and K contents suggested that these minerals neutralized the low pH by HCl secreted in the stomach and organic acids produced by fermentation in the caecum. The significant negative correlations between pH and the ratio of Mg, Ca or P contents (Sup %) in supernatant fraction to total contents showed the possibility that the insoluble salts of these minerals precipitated as pH rose. The sudden increase of P content in the caecum might be due to the accumulation of P into multiplied bacteria. These results suggested that the sudden alkalization of digesta might decrease the absorptions of Mg, Ca and P in the intestine.

Highly purified Sm small nuclear ribonucleoproteins (snRNPs) in the nuclear extract of embryoid bodies and F9 cell extract that contained small nuclear RNAs (snRNAs) U1a, U1b, U2, U4, U5, U6 and X (about 90 nucleotides) were recovered from anti-Sm antibody column. Sm snRNPs were also tried to isolate immunochemically from chromatin/nucleolar digest and the post-mitochondrial cytoplasmic fraction. It was demonstrated that selective leakages of any major U-snRNPs to those fractions during aqueous cell fractionation seem unlikely. The high proportion of U1a-snRNP to U1b-snRNP in embryoid bodies and F9 cells was reproduced, using anti-Sm column. X snRNA could not be detected in the Sm snRNP fraction from human KB cell and 129 male mouse liver nuclei. Anti-2, 2, 7-trimethylguanosine (m
2, 2, 73G) antibody column was also demonstrated to be useful for the fractionation of U-snRNPs from embryoid bodies. Only U1a, U1b and U2-snRNP were retained by and eluted from the column. However, when the deproteinized RNA isolated from the nuclear extract of embryoid bodies was loaded onto the column, other U snRNAs (U4 and U5 snRNA) as well as the three U snRNAs were also retained by and eluted from the column. The partially purified U1-snRNP(s) from embryoid bodies was eluted at 0.18 M NH
4Cl and U2+U4+U5 as well as U1-snRNP(s) eluted at approximately 0.3 M NH

In October 1985, a fatally peracute disease of newborn piglets occurred in Kanagawa prefecture, Japan. The disease was characterized by depression, tremor, and yellowish-brown diarrhea. A virus showing cytopathic effect (CPE) in ESK cell culture was isolated from the brain, lungs, kidneys, tonsil, and intestine of al1 4 dead piglets. One of the isolates was cloned and named Kanagawa strain. The virus of the Kanagawa strain was identified as Getah virus, a member of alphavirus of the family Togaviridae, from the results of physicochemical, biological, and serological properties.

A sensitive microassay by reverse passive hemagglutination (RPHA) was carried out for the measurement of bovine myoglobin (Mb). Sheep erythrocytes fixed with glutaraldehyde and treated with tannic acid were coated with anti-bovine Mb antibodies purified by affinity chromatography. The RPHA test was done by a microtiter method and minimal detectable Mb levels were 10ng/ml in serum and 2ng/ml in urine. Mb levels of 15 normal calves and 15 normal cows ranged from <10 to 80ng/ml in serum and from <2 to 16ng/ml in urine. Sixteen calves with nutritional myopathy showed markedly increased Mb levels in serum, 640 to 51, 200ng/ml, and in urine, 640 to 256, 000ng/ml. Serum Mb levels in calves recovered from nutritional myopathy decreased to nearly normal ranges within 3 to 5 days. Although each of the 51 cows at the onset of parturient paresis showed high levels of serum Mb ranging from 320 to 20, 480ng/ml, while about 30 per cent of the cows showed normal serum CPK activities. Serum Mb levels in cows recovered from parturient paresis rapidly decreased to the normal levels after 2 to 4 days. Serum Mb in cases slaughtered continued at high levels until the last treatment of parturient paresis. It would be concluded that RPHA method is useful for Mb determination in evaluating the myolytic state of bovine myopathy and for early detection of the disease.

The Kanagawa strain of Getah virus isolated from newborn piglets which died of peracute disease was examined for pathogenicity in gnotobiotic piglets. Its pathogenicity depended on the route of inoculation. Piglets inoculated intramuscularly on 5 to 18 day old exhibited anorexia, depression, tremor of whole body, red discoloration of the skin, trembling of the tongue, and incoordination of pelvic limbs 20 hrs and died or became moribund 2 to 3 days after inoculation. The virus was recovered at high titer from various organs and serum of dead and moribund piglets.

Twenty normal healthy mongrel dogs were divided equally into four groups. Acupoints were stimulated bilaterally by insertion of sterilized stainless needles at ST-36. The needles were inserted and left in situ for 30 minutes/day. The single acupuncture group was needled for a day without an electric stimulator, and the repeated acupuncture group was needled once a day for five days. The single electroacupuncture group was stimulated for a day with for electric stimulator (1 volt, 2Hz, 1msec, positive square wave), and the repeated electroacupuncture group were stimulated once a day for five days. Cellular immunological responses and hematological findings were determined after repeated acupuncture stimulations. ANAE positive cells stained by alpha naphthyl acetate esterase, the stimulation index of lymphocytes blastformed by incubation with phytohemagglutinin and the Y-globulin of serum protein all decreased within several days after repeated electroacupuncture stimulations, representing a decrease in basal immunological response. After the repeated electroacupuncture stimulation on the ST-36, it was suggested that in healthy dogs the depression of the immunological responses was due chiefly to the T-cells.

The karyotype of a Holstein-Friesian bull with unilateral cryptorchidism was investigated according to the G-, C- and Cd-banding techniques. By G-banded karyotyping, it was shown that this bull had 1/26 Robertsonian translocation. Furthermore, there were 2 dark dots at the centromere of 1/26 translocated chromosome by C- and Cd-banded karyotyping. Although the chromosomes of the parents of this bull were not examined, 12 half-sib cows from different sires or dams did not show any chromosomal aberrations. The semen quality of the father and the conception rate of the parents were within the normal range in Japan. From the results obtained, it appears that this translocation resulted from a centric fusion at the early embryonic stage and had occurred de novo.

The effects of β-carotene and vitamin A on progesterone production by cultured granulosa cells of cows were studied. The granulosa cells were isolated from medium-sized follicles (8-10mm) by the needle aspiration. When granulosa cells were treated with 50μg/dl of β-carotene for 4 days, progesterone production was increased slightly. In contrast, treatment with the same amount of vitamin A was associated with a significant (P<0.01) increase in progesterone production of the cells. Stimulation of progesterone production by vitamin A was noted in dose-dependent manners. Proliferation of granulosa cells was suppressed by vitamin A treatments, and some morphological changes were observed in cultures grown in media with vitamin A. These results suggest that β-carotene, mediated by vitamin A may stimulate steroidogenesis of granulosa cells.

The prevalence of antibodies against Toxoplasma gondii, Brucella abortus, Leptospira interrogans (serovars; icterohaemorrhagiae, autumnalis, hebdomadis, australis and canicola), Chlamydia psittaci and Japanese encephalitis virus was determined in serum samples collected from hunter-killed wild Japanese serows in Gifu Prefecture during the winter months of 1981 to 1984. Seropositive rates for each agent were 10.7% (36/335) for Chlamydia psittaci, 10.4% (42/404) for Leptospira interrogans and 5.4% (41/765) for Toxoplasma gondii, in descending order. Antibodies to Brucella abortus and Japanese encephalitis virus were not detected in 718 and 208 serum samples, respectively. The results obtained suggest that wild Japanese serows have serologic evidence of infection with some of the zoonosis agents tested.

A hairworm was vomited by a domestic cat. The worm specimen is white with a yellowish tint and 585mm in length and 1.2mm in maximum width. Morphological examinations revealed that the specimen is almost identical to Gordius ogatai Inoue, 1979 although not closely identified because of the damage of the cuticle. The cat would be infected by ingesting host insects parasitized by a well-developed young hairworm.

Neurotoxic effects of clofenotane (DDT) and dieldrin were analyzed pharmacologically in mice. Oral (p. o.) or intravenous (i. v.) administration of DDT caused the tremor, while dieldrin caused the tonic or clonic convulsion. Intracerebroventricular (i. c. v.) injection of both agents also caused the neurotoxic symptoms which were qualitatively similar to those shown after p. o. or i. v. administration. These effects may be central nervous system in origin since i. v. injection of either insecticides had no direct effect on sciatic nerve-gastrocnemius muscle system after the nerve had been sectioned. The tremor due to DDT was aggravated by dopaminergic antagonist but unaffected by muscarinic antagonist. The convulsion due to dieldrin was alleviated by muscarinic or dopaminergic antagonist but aggravated by adrenergic or GABA ergic antagonist. These differences suggest that DDT and dieldrin develop their neurotoxic symptoms through different neural pathway(s).

A total of 266 calves, 154 from areas with a history of the outbreaks of congenital anomalies caused by bovine viral diarrhea-mucosal disease (BVD-MD) virus and 112 from a clean area, were examined for persistent infection with BVD-MD virus. Non-cytopathogenic BVD-MD virus was isolated from sera and leukocytes obtained from 18 calves in the epidemic areas. The second blood samples were collected from 13 of 18 calves positive for virus isolation 1 to 2 months after the initial sampling, and the virus was again isolated from 12 calves, indicating that they were persistently infected with BVD-MD virus. Viral titers in sera and leukocytes of persistently infected calves ranged between 10

50/ml. None of persistently infected calves has serum neutralizing antibody to BVD-MD virus. No persistently infected calves were found in the clean area. Gas chromatography (GC) was tried for rapid presumptive diagnosis of clostridial infection in domestic animals. Propionic, butyric, and acetic acids were detected by GC in specimens collected from lesions in the liver and spleen of a pig died of gas gangrene. The volatile fatty acid (VFA) pattern was very similar to that of a culture of Clostridium novyi isolated from these lesions. Emulsions of the lesions in the liver and spleen of the two cows died of blackleg exhibited VFA patterns resembling that of a culture of Clostridium chauvoei isolated from these lesions. The results of GC analyses of subcutaneous lesions of mice and guinea pigs experimentally infected with C. novyi and those with C. chauvoei were very similar to those of the pig and the cows, respectively. The VFA pattern of lesions of domestic animals may be useful for rapid presumptive diagnosis of a certain clostridial infections such as that with C. novyi. It may be difficult, however, to distinguish among infections with Clostridium tetani, Clostridium septicum, and C. chauvoei by GC analysis.

Fecal microflora of wild Japanese monkeys in a snowy area was compared with that of laboratory-reared Japanese monkeys. The wild animals in winter fed mainly on the bark of certain broad-leaf trees and the reared animals were fed the commercial monkey diet and raw sweat potatoes. The total bacterial count of the wild animals was significantly lower than that of the reared animals. Significant qualitative differences in the numbers of all aerobes in both groups were observed, whereas the numbers of all anaerobes in both animals did not differ. The numbers of Bacteroidaceae, Eubacterium spp., Peptococcaceae, Bifidobacterium spp., Veillonella spp., Sarcina ventriculi, Entero-bacteriaceae and Streptococcus spp. in the wild monkeys were significantly lower than those in the reared monkeys. Also, a significant reduction of Bacteroides spp. isolated from bacteroides-selective agar (NBGT) was observed in the feces of the wild animals. However, the numbers of Lactobacillus Spp., anaerobic curved rods and spiral shaped rods did not differ in both animals. Lecithinase-negative Clostridium spp. was predominant fecal bacteria in the wild monkeys, although the incidence was low. The results show that the "bark-eating" habits may cause remakable changes in the microbial population and proportions in the feces in the wild Japanese monkeys in snowy areas.

Monoclonal antibody (MoAb) c143 has been shown to react with tumor-associated antigen on neoplastic cells from cattle with enzootic bovine leukosis (EBL). The c143 showed suppressive effect on tumor growth of EBL cell line implanted in nude mice as evidenced by smaller size of tumor nodules in the antibody-treated group than that of the control animals (P<0.05). The histopathological changes on inoculated sites of mice treated with c143 were examined. In c143-treated mice, proliferation of macrophage was observed at 3 days and was remarkable between 7 to 14 days after tumor cells inoculation. Cellular immune responses by macrophages might be involved in the anti-tumor effects of c143-treated nude mice.

In order to investigate whether heartworm migration was induced or not, metoprolol, an anti-hypertensive agent with a cardioselective β-adrenoceptor blocking action (β
1-blocker), was administered to heartworm-infected dogs under general anesthesia. After about 8mg/kg body weight of metoprolol administration, heart rate and right cardiac output reduced significantly in all 6 dogs. Heartworms migrated from the pulmonary artery to the right atrium in 5 dogs, and hurdled the pulmonary valve transiently in 1 dog on two-dimensional echocardiogram. However, the worms returned to the pulmonary artery in a short time in 4 cases. The pressures at the right ventricle and the pulmonary artery and stroke volume did not show uniform changes after the administration. It was considered that the decrease of heart rate and cardiac output, namely, decreased blood flow volume and velocity at the pulmonary artery by metoprolol administration induced the heartworm migration in the direction opposite blood flow.

A sero-epizootiological study was conducted on horses which showed clinical pyrexia at two training centers, each of which maintained 1, 200 to 2, 100 racehorses for training, of the Japan Racing Association. It continued from 1980 to 1985 to clarify the cause of pyrexia, so that measures might be considered for the prevention of infectious disease. A total of 3, 849 horses were found to be affected with pyrexia for the 6 years. Of them, 2, 852 horses were tested to equine herpesvirus type 1 (EHV-1), equine rhinovirus type 1 (ERhV-1), rotavirus and equine adenovirus (EAdV) by collecting paired serum samples. As a result, 669 (23.5%) horses showed seroconversion to these viruses; that is, 491 horses (73.4%) to EHV-1, 91 (13.6%) to ERhV-1, 64 (9.6%) to rotavirus and 23 (3.4%) to EAdV. EHV-1, ERhV-1 and rotavirus exhibited a seasonal characteristic in spread. EHV-1 and rotavirus spread in winter and early spring. ERhV-1 spread not only in the cold season, but in changing times of season, such as early spring and early autumn. The percentage of 3 years old in the horses showing seroconversion to EHV-1 was two times higher than those in the horses showing seroconversion to other viruses. All of the horses showing seroconversion to ERhV-1 and EAdV were under 4 or 3 years old, respectively. The mean age of the horses showing seroconversion to EHV-1, rotavirus, ERhV-1 and EAdV were 2.96, 2.73, 2.44 and 2.25, respectively.

Inactivated Sendai virus vaccines including chemically virus-core-modified ones and virus-disrupted ones were examined immunohistologically in mice for their nasal immunogenicity. With five virus-disrupter (deoxycholate, phenol, urea, alkali and thymol) -treated vaccines, no immunoprophylactic effect in the entire respiratory tracts was observed. Of eight vaccines inactivated by chemical modification of viral core RNA, diethylsulphate, N-methyl-N'-nitroso-guanidine, and crystal-violet vaccines had no marked protection, whereas brilliant-green, rhodamine-B, and Hofmann's violet vaccines induced strong protection. Marked effects were brought about with hydroxylamine-treated vaccine, but not with O-methylhydroxylamine-treated vaccine. High nasal immunogenicity was elicited by such inactivated vaccines having partially modified core RNA and intact superficial layers of the virion.

A new strain of feline infectious peritonitis virus (FIPV), FIPV-HR, was isolated from a diseased household cat using fcwf-4 cells after ten passages in cats. The maximum titers of cell-associated and cell-free viruses scored 4.8×10
7 and 2.3×10
7PFU/ml, respectively. Half time of the reduction of viral infectivity at 37°C was 180min. The in vitro grown virus induced exudative peritonitis in kittens.

Pulsed Doppler examinations combined with M-mode and/or two-dimensional echocardiography (pulsed Doppler echocardiography) were performed on 10 dogs and 9 calves which were clinically normal, and 3 cases of valvular regurgitation. The Doppler technique is described. The blood flow patterns obtained at all four valve areas in normal animals and the abnormal pulsed Doppler echocardiographic findings in clinical cases are reported. The findings suggested that pulsed Doppler echocardiography can be useful not only for diagnosis, but also for assessing the degree of severity of any type of valvular regurgitation in dogs and calves.
