    Fusobacterium necrophorum was inoculated into the portal vein of eight cattle. Multiple necrotic foci were found in the livers of three of them which died acutely. The remaining cattle autopsied 12 to 126 days after the inoculation had abscesses in their livers. In the former three cattle, multiple hyperechoic masses were detected in the liver by ultrasonography. The masses corresponded to the necrotic foci. In the latter five cattle, a liver lesion was similarly detected, first on the 5th to 10th day after inoculation. It consisted of a central hyperechoic mass and a surrounding hypoechoic halo, which corresponded to necrotic tissue and granulation tissue, respectively. In some cases the central hyperechoic area of the halo pattern changed later to be hypoechoic. The ultrasonographic lesions appeared as a spherical hypoechoic pattern. Finally, they disappeared from the hepatic images on around the 50th to 70th day after inoculation. These processes corresponded to diminution of the central necrotic mass, the abscess being gradually replaced by the granulation tissue and capsule and to change of the abscess into the scar. The capsule and scar were imaged as a isoechoic or hypoechoic pattern. Some other lesions of the halo pattern remained and were accompained by lateral shadows.

    Serial determination of serum bile acids (SBA) concentration and its fractionation after feeding were performed in 8 healthy dogs. The mean preprandial SBA was 1.07±0.91μM/l. The mean SBA reached a maximum value of 10.35±7.32μM/l at 6 hour with very rapid risd at 1 hour after feeding. There were no significant differences in any of the mean SBA values during the 1 to 8 hour postprandial period. The mean preprandial triglycerides (TG) concentration was 37.07±13.51mg/dl. The mean TG increased rapidly after feeding and reached a maximum value of 73.53±12.01mg/dl at 2 hour. It would appear that postprandial sample for SBA can be taken at 1 hour after feeding. Unfortunately, none of the peaks was identified in the fractionation study. An unidentified peak with a retention time near 6.4min. increased significantly in the 2 hour sample. A new unidentified peak with a retention time near 9.0min. appeared in the 2 hour sample.

    The cytodifferentiation of the cephalic and caudal lobes of the chicken pituitary of different embryonic ages was studied by means of an immunocytochemical technique using antisera to avian and mammalian pituitary hormones. LH-immunoreactive (ir) cells occur first in 4-day embryos in the epithelium of the posterior process of Rathke's pouch, spread in the oral lobe in 6-day embryos and are distributed throughout the pars distalis in 8-day and older embryos. FSH-ir cells occur in the vental aspect of the caudal lobe in 8-day embryos and are distributed throughout the gland after 12-day. ACTH-ir cells occur first in the dorsal part of the cephalic lobe in 7-day embryos and are confined to the cephalic lobe thereafter. TSH-ir cells occur first in the ventral part of the cephalic lobe at 7-day and are confined to the cephalic lobe thereafter. GH-ir cells occur in the ventral portion of the caudal lobe in 8-day embryos, increase remarkably in the later embryonic period and occupy the caudal lobe thereafter. PRL-ir cells occur first in the cephalic lobe in 18-day embryos, and increase remarkably after hatching. The pars tuberalis contains numerous LH-ir cells since it is originated from the lateral lobe in 6-day embryos.

    A method for separation of the acids of the citric acid cycle and related pathways was developed. The acids were separated by high-performance anion-exchange liquid chromatography on a short column of a strong anion-exchange resin (IEX 520AE) which was eluted with 1/30 M potassium phosphate buffer containing 27.5% (v/v) acetonitrile (pH 3.0) at 40°C. Compounds in the column effluent were detected by ultraviolet spectrophotometry at 210nm. This method was used in the analysis of acids of the citric acid cycle of rat brain and liver and of products of the reaction of succinic thiokinase or fumarase with extracts of animal tissues.

    A histopathological screening method was developed to evaluate vaccinal effects on Mycoplasma gallisepticum (MG) infection in chickens. Lesions in the tracheal mucosa were graded by scoring, and mucosal thicknesses were measured and used as an indicator representing the degrees of lesions. There was a close relation between the lesion score and mucosal thickness when examined in vaccinated and challenged or unvaccinated and challenged chickens. The measurements of mucosal thickness in the trachea seemed to be a simple and objective method to estimate the degrees of lesions induced by MG infection.

    The culture supernatants of feline alveolar macrophages stimulated with lipopolysaccharide (LPS) were found to have the ability to enhance mouse thymocyte proliferation in response to phytohemagglutinin A. None of these culture supernatants contained interleukin 2 (IL-2) activity, as determined by the murine IL-2 dependent T cell line (CTLL-2) cell proliferation assay. These results indicate that the thymocyte proliferation is attributed to feline interleukin 1 (IL-1). The maximal IL-1 activity was observed in the supernatants from alveolar macrophages cultured with LPS at concentrations ranging from 5 to 10μg/ml for 12hr. Feline IL-1 activity was sensitive to heat treatment at 70°C for 30min. The molecular weight (m. w.) of the feline IL-1 ranged from 15000 to 20000 daltons, as estimated by Sephacryl S-200 gel filtration, which was within the range of values estimated for IL-1 of humans, mice and rabbits. Feline IL-1 was eluted with three different isoelectric points of 4.1, 4.8 and 5.3, respectively, which resembles murine and human IL-1α.

    Using Thoroughbred horses, ponies, Holstein cows, pigs, goats and chickens, the effects of atropine and/or propranolol on heart rate (HR) and T-wave amplitude (TWA) on ECG were examined. HR increased significantly in all animal species after the administration of atropine, and decreased in the pig, goat and chicken after the administration of propranolol. Parasympathetic predominance, estimated from the changes in HR induced by the autonomic blockers, was in the order of Thoroughbred horse > pony > cow > goat whereas the sympathetic predominance was in the order of chicken > pig. The positive ingredient in TWA was increased significantly in the Thoroughbred horse, pony, pig, and goat by atropine whereas decreased only in the pig by propranolol.

    On March 27, a dog kept in Toride, Ibaraki Prefecture, died after manifestation such clinical signs as intense pruritus and self-mutilation. As Aujeszky's disease had been prevalent in swine since 1981 in the area, a canine case was suspected. At autopsy, no significant gross visceral lesion was observed but histopathological observations revealed moderate perivascular cell infiltration, gliosis and neuronophagy in the medulla oblongata and cervical cord. Two rabbits inoculated with a mixture of the brain and spinal cord homogenates and a mixture of the lung and spleen homogenates, respectively, died 4 days after showing pruritus. A cytopathic agent was recovered from the spinal cord of the affected dog in PK-15 cell cultures. The cultures of PK-15 and MA-104 cells inoculated with the agent showed clear cytopathic effect of cell rounding and large syncytia, in which intranuclear inclusion bodies were seen after Giemsa's staining. Electron microscopic observations of the concentrated culture fluid of infected PK-15 cells revealed many particles characteristic of Herpesviridae. The agent was immunologically identified as Aujeszky's disease virus by immunofluorescence.

    Two calves with Arnold-Chiari malformation were examined macroscopically. The brain and cranium appeared to be compressed dorso-ventrally and parts of the cerebellum, medulla oblongata and fourth ventricle herniated through the enlarged foramen magnum to the vertebral foramen of the atlas. The internal cranial base composed of the sphenoid and occipital bones was abnormally flat, so the hypophyseal fossa and dorsum sellae were obscure. It was considered that the malformation probably resulted primarily from an abnormal development of these bones, the osteogenesis of which was induced by the notochord, and the brain secondly showed its abnormal shape and position.

    Circulating immune complex (CIC) levels of sera from cattle with enzootic bovine leukosis (EBL) and bovine leukemia virus (BLV) negative healthy cows were quantified by a polyethylene glycol turbidity method. Sera from EBL cows and those from EBL cows with membranous glomerulonephritis showed significantly higher level of CIC when compared to that of control normal cows (p<0.05). The CIC of sera from EBL cows showed inhibitory effect to mitogen-induced blastogenesis of normal bovine lymphocytes.

    Piliated (P+) and non-piliated (P-) bacteria of Corynebacterium renale strain No. 115 were compared in their susceptibility to phagocytosis by mouse peritoneal macrophages. In the absence of opsonins, P+ bacteria were less phagocytized than P- bacteria (P<0.001). Complement at 20% concentraTion enhanced the uptakes of P+ and P- bacteria (P<0.01). In the presence of anti-pili serum, P+ bacteria were phagocytized at a significantly higher rate than P- bacteria (P<0.01), regardless of the presence of complement. The anti-pili monoclonal antibodies 16/5 and 13/4 were also opsonic for P+ bacteria, regardless of the presence of complement, but there was less effect than that of anti-pili polyclonal antibody. In the presence of anti-P+ bacteria serum and complement, P+ bacteria were more significantly phagocytized than P- bacteria (P<0.05), which was possibly due to presence of anti-pili antibody in the anti-P+ bacteria serum. In the presence of anti-P- bacteria serum, the uptake of P+ bacteria was unexpectedly enhanced and bacame nearly equal to that of P- bacteria. The uptake of P- bacteria was nearly constant in all the experiments, except that it was slightly enhanced by the presence of complement.

    Fusobacterium necrophorum was inoculated into the portal vein of seven cattle. Multiple necrotic foci were found in the livers of the two cattle which died on 10th and 14th day after inoculation. The remaining cattle autopsied on 12th to 126th day after inoculation were affected with hepatic abscesses. Some of them were affected also with extrahepatic abscesses found mainly in the lung and intercostal space. Serum sialic acid concentration was not elevated even 10 days after the inoculation in one of the two cattle affected with the multiple necrotic lesions in the liver. In the second it began to rise 8 days after the inoculation. In cattle affected with the hepatic abscesses, it began to rise above preinoculation values 3 to 5 days and reached a peak values 7 to 9 days after the inoculation. They remained at this level for 2 or 3 days and then either decreased gradually toward preinoculation values or remained somewhat elevated for more than 4 months. The effects of both hepatic and extrahepatic abscesses on the serum sialic acid level were discussed. Hepatic abscess formation correlated significantly with the rise in serum sialic acid level.

    The ethidium bromide (EB) fluorescence assay was applied to evaluate blastogenesis of swine lymphocytes by mitogens. The optimal responses were observed in cultures of 1.0×106 cells for 72hr with the final concentration of PHA-P of 5μl/ml, Con A of 25μg/ml or PWM of 0.25μg/ml. At the optimal conditions, values of stimulation index by the EB fluorescence assay (SI-FA) correlated highly with those of stimulation index by the radioisotope incorporation assay (SI-RI) (r=0.752, n=21, P<0.01). The correlation coefficients were r=0.863 in PHA-P and Con A (n=7, P<0.05) and r=0.710 in PWM (n=7, P>0.05). From these results, this EB fluorescence assay is thought to be possible to be used in place of the radioisotope incorporation assay for evaluation of blastogenic response of swine lymphocytes. The SI-FA values of blood lymphocytes of healthy pigs at about 6 months old were 1.54±0.26 to PHA-P, 1.60±0.23 to Con A and 1.56±0.28 to PWM.

    Histopathogenesis of intestinal tumors in 178 rats induced by oral administration of bracken was studied. The tumors was noted to be epithelial and non-epithelial being mainly localized at ileum (93.2%). The epithelial ones occurred at a high rate, and they were diagnosed as tubular adenocarcinoma (178 cases), papillary adenocarcinoma (22 cases) and adenoma (6 cases). Tubular adenocarcinoma with most frequences was presumed to occur from the proliferative zone in crypts of the small intestine. On the other hand, the non-epithelial tumors occurred being closely related to the stroma or adjacent tissue of carcinoma. They were noted to be leiomyosarcoma (62 cases), fibrosarcoma (14 cases), fibroma (10 cases), leiomyoma (8 cases), lymphangioendothelioma (2 cases), hemangioma (1 case) and hemangioendothelioma (1 case).

    Thirteen monoclonal antibodies against intraerythrocytic merozoites (piroplasms) of Theileria sergenti were prepared for analysis of their structural proteins. All monoclonal antibodies stained the piroplasm but not uninfected erythrocytes by indirect fluorescent antibody test. Western blot analysis showed that 6 hybridomas tested recognized polypeptides with molecular weights of 32, 000 or 23, 000.

    Between April and June 1986, the tonsils of 126 healthy pigs were examined for the presence of staphylococci. All of the pigs examined harboured staphylococci in the tonsils. The most predominant Staphylococcus species was S. hyicus subsp. hyicus (92.9%), followed by S. aureus (57.1%), S. hominis (40.5%), S. simulans (22.2%), S. epidermidis (3.2%), S. xylosus (2.4%), S. haemolyticus (1.6%) and S. hyicus subsp. chromogenes (0.8%). Unidentifiable species were isolated from 10 (7.9%) of the 126 tonsils examined. The combinations of S. hyicus subsp. hyicus and S. aureus with or without other species were most frequently encountered, finding in 53.2% of all the tonsils examined. The main phage patterns of 69 S. hyicus subsp. hyicus strains typable with S. hyicus subsp. hyicus phage set were S13 (n=27), S9/S39/S188 (n=12), S9/S188 (n=11) and S9 (n=6). The main phage patterns of 63 S. aureus strains typable with the human S. aureus phage set were 3A/3C (n=36) and 3C (n=16). Sixty-one coagulase-negative staphylococcal strains typable with Pulverer's phage set were differentiated into 25 phage patterns.

    The present paper is concerned with in vitro fertilization (IVF) and embryo transfer (ET) in the pig. Follicular and tubal oocytes were recovered 39 to 40hr post hCG from PMSG-hCG treated prepuberal gilts and were inseminated with ejaculated spermatozoa capacitated in vitro. The 95% (25/26) oocytes were fertilized in vitro 6 to 7hr after insemination. There was the variability in polyspermic fertilization between males. About half of IVF embryos reached 2-cell stage at 27 to 28hr and the cleavage rates, as determined by the number of embryos developed 2- to 8-cell stage at 27 to 72hr after insemination, were 83% (43/52) in follicular oocytes and 67% (18/27) in tubal ones, respectively. Seventeen 2-cell stage embryos which originated from IVF of follicular oocytes were transfered into the oviduct of a recipient at 29hr after insemination. This recipient farrowed 4 healthy piglets at 113 days after ET. The results demonstrate the feasibility of IVF and ET in the pig.
