Ito cells are accumulated in the lesion of liver fibrosis in clinical and experimental conditions. However, the detail mechanism of Ito cell accumulation is still unknown. In this study, we examined the effects of basic fibroblast growth factor (bFGF) on Ito cell migration and proliferation using primary cultured rat Ito cells.
Ito cells were prepared from a female Wistar strain rats (bw 200-250g) and were inoculated on plastic culture dish and cultured in DMEM with 10% fetal calf serum for 5 days. Migration and proliferation were assessed with or without bFGF (1-100ng/ml) using time-lapse VTR and BrdU staining. Cultured Ito cells showed spontaneous migration and proliferation. These processes were significantly accelerated by bFGF in a dose-dependent manner. In conclusion, bFGF might play a role in the accumulation of Ito cells in the lesion of liver fibrosis.

Large amount of IL-8 was found to be produced by five human liver tumor cell lines, and the production was augmented by the addition of IL-1β or TNF-α, but not by IFN-γ. Northern blot analysis demonstrated that IL-8mRNA expression was increased in the presence of IL-1β or TNF-α. Reverse transcription polymerase chain reaction showed that all hepatocellular carcinoma (HCC) tissues examined in this study expressed IL-8mRNA. In vitro analyses using human umbilical vein endothelial cells (HUVECs) demonstrated that conditioned medium of PLC/PRF/5 cells as well as recombinant IL-8 stimulated the proliferation of HUVECs in a dose-dependent manner, and the stimulatory effect was partially inhibited by the addition of neutralizing antibody against IL-8. Our present data indicate that IL-8 produced by HCC cell lines has an angiogenic activity.

A case with the giant liver cell adenoma was reported. The patient was an apparently healthy 23-year-old female with no history of drug intake. Physical examination revealed no abnormalities except for the presence of a large mass at the epigastrium. Ultrasonogram, CT and MRI revealed large mass in the left hepatic lobe. Angiographic finding was compatible with liver cell adenoma, with capsular arteries. Liver scintigram showed cold spot in the same area.
The surgical specimen showed large dark red tumor measuring 22×20×12cm. The tumor was composed of cords and sheets of well differentiated clear hepatocyte and these mostly resembled to normal hepatocyte histologically.
Only 53 cases with liver cell adenoma have been reported in Japanese literature till now, and the large one in the left lobe such as over 21cm in diameter was reported only this case.

A 65-year-old man with a history of C viral liver cirrhosis was pointed out 15mm space occupying lesion in the S8 segment of the liver on dynamic CT, demonstrating rapid disappearance associated with arterioportal shunt, which is commonly regarded as a pathognomonic sign of hepatocellular carcinoma. However hepatic angiography showed pooling appearance in the S8 segment compatible with liver hemangioma, which atypically associated with arterioportal shunt. To our knowledge, such an atypical liver hemangioma with arterioportal shunt mimicking hepatocellular carcinoma on dynamic CT is rare in the literature.

A 39-year-old housewife was admitted to our hospital with an asymptomatic tumor in the lateral segment of the live. She had no oral contraceptives and other drugs. Follow up for her on that tumor had been made for three years prior to this admission by abdominal ultrasonography (AUS). The tumor developed from 1.8cm to 2.8cm in diameter with a 572-day doubling time. AUS showed hyperechoic, homogeneous and irregular shaped mass. The tumor was demonstrated as a low density area on plain computed tomogram (CT) and slightly enhanced area on enhanced CT. Magnetic resonance imaging (MRI) showed a high intensity and isointensity area on T1 and T2 weighted images respectively. Angiography indicated that the lesion had faint staining. A laterla segmentectomy of the liver was performed with strong suspicious of a well differentiated hepatocellular carcinoma. The pathological findings revealed it a liver cell adenoma with marked fatty degeneration. The patient has no sign of recurrence now 4 yers after surgery.
