    A home-made electrospray ionization (ESI) source, which generated multiply charged gas-phase ions of proteins under atmospheric pressure, was coupled with a tandem quadrupole mass spectrometer. A detection limit of 18 fmol (1.8 × 10-14 mol) and a precision of 0.01% in the molecular weight measurement were attained using hen-egg lysozyme as a typical protein.
    ESI mass spectrometry (MS) was applied to the prompt identification of the amino acid substitutions in recombinant human lysozymes (HLY). However, the direct analysis of HLY was prevented by the co-existing inorganic salts added to retain its biological activities. On-line coupling of ESI-MS with micro-column liquid chromatography (ESI-LC/MS) was developed to perform direct molecular weight measurements of salt-containing proteins at the picomole level. LC/MS interfacing by ESI required to alleviate the sensitivity losses brought about by the high electric conductivity and the high surface tension of the LC eluent.
    ESI-MS and ESI-LC/MS were successfully applied to the characterization of the post-translational modifications found in recombinant wheat germ agglutinin (WGA2). These methods along with fast atom bombardment (FAB) MS and FAB LC/MS were also applied to the characterization of natural variants of β-casein (β-CA). The possibility and limitation of the present methods for the analysis of complex proteins were discussed.

    Fourier analysis was performed for the oscillatory yield curves obtained in experiments of low energy ion scattering spectroscopy (ISS) to facilitate the understanding of the underlying mechanism of the processes. Major components that appeared in the yield spectral density functions by the Fourier transform were responsible for oscillatory structures arising from the quantum mechanical interaction of the vacant 1 s state of incident He+ ions with the spin-orbit split 4f7/2 and 4f5/2 orbitals of Ta and W target atoms. Two major components at the frequencies of 1.6×106 and 2.8×106 cycles-m/s in the spectral density functions for the Ta target and one major component at the frequency of 1.5×106 cycles-m/s for the W target were respectively assigned as attributable to the ISS quantum interferences.

    With the use of a tunable and coherent vacuum ultraviolet (VUV) radiation around Lyman-α line, a spectroscopy combining the resonantly enhanced multiphoton ionization with the Doppler spectroscopy (REMPI-Doppler spectroscopy) was investigated. The method was applied to the detection of hot hydrogen (H) atoms photodissociated from hydrogen iodine (HI) molecules. Corresponding to whether the transition is parallel or perpendicular, the velocities of recoiling H atoms were measured to be 1.12×104 m/s for parallel transition dissociating to [H(2S1/2)+I*(2P1/2)], and 1.75×104 m/s for perpendicular transition dissociating to [H(2S1/2)+I(2P3/2)], respectively. Atomic H beams formed via purely parallel or perpendicular transition were extracted to measure the velocity spreads of the beam. Experimental velocity distributions were fitted well to the theoretical curves calculated by taking account of appropriate rotational and translational motions of parent HI molecules.

    Fourteen types of corona discharge needles made of different materials were used for high performance liquid chromatography/atmospheric pressure chemical ionizationquadrupole mass spectrometer (LC/APCI-QMS), and after a lapse of one hour and five hours from the start of the equipment, selected-ion monitoring determination of 3,5-di-tert-butyl-4-hydroxytoluene (BHT) was made seven times repeatedly. Suitability as a discharge needle was judged by the total (n=14) relative standard deviation (T-RSD) of the quantitative determination value. T-RSD's of the iron needle, tin-plated iron needle, silver alloy needle, platinum alloy needle and stainless steel needle were 1-3%.
    It could be elucidated from the scanning electron micrograph of the needle surface that suitability as a discharge needle was related to the stability of needle surface material against the corona discharge. The iron needle, having a small T-RSD was judged to be the most suitable as a corona discharge needle for quantitative analysis.
