In this study, the on-line high-throughput detection technique of a reaction product of a micro-chemical synthesis using microchips was developed by means of an electrospray ionization mass spectrometry (ESIMS) with on-chip purification (extraction) of reaction mixture to reduce any ionization hindrance. This technique is useful for on-line high-throughput qualitative analysis of chemical synthesis products. This technique was applied to monitor the on-chip
N-trifluoroacetylation of 2-phenetylamine. The remaining 2-phenetylamine and trifluoroacetic acid that acted as an ionization hindrance were extracted from the ether solution of the reaction mixture to the phosphate buffer solution using an extraction microchip connected to the micro-synthesis chip. By decreasing the remaining 2-phenetylamine and trifluoroacetic acid, the spectrum of 2,2,2-trifluoro-
N-phenetyl acetamide in the reaction mixture was clearly observed by electrospray ionization mass spectrometer.
