    To investigate the host specificity of Ophiocordyceps nutans against hemipteran insects in the wild, we determined the relationship between host species and rDNA-internal transcribed spacer (ITS) variation in O. nutans. The analyzed fungal specimens infected 16 host species belonging to four families of Hemiptera. The molecular phylogenetic analysis revealed that O. nutans can be classified into two types corresponding to their host families. The genetic distance values between the two types were very remote (>0.084), and the strains of O. nutans that parasitized Halyomorpha halys and Plautia crossota stali, well-known insect pests, formed a subclade. The results suggest that O. nutans should have host specificity which can be valuable for developing biological control agents against specific hemipteran insects.

    Mating-type genes were cloned and sequenced in the heterothallic phytopathogenic fungus Exserohilum monoceras, a candidate bioherbicide for the control of Echinochloa weeds in rice fields. Using PCR-based methods, we determined both idiomorphs and flanking regions of these genes. The structural organization of the E. monoceras Mat locus was similar to that of other heterothallic ascomycetes. The MAT1-1-1 gene was 1191 bp and encoded a predicted protein of 379 amino acids with an α box domain. The MAT1-2-1 gene was 1093 bp and encoded a predicted protein of 345 amino acids with a high mobility group box domain. Expression of both MAT genes was detected under vegetative and invasive growth conditions. MAT-specific primers were developed to assess the mating-type frequency of E. monoceras field isolates. Both mating types were observed in Japanese field isolates. Analysis of mating types and sexual hybridization of E. monoceras could provide useful approaches for the conventional genetic manipulation of this fungus to produce a more efficient bioherbicide.

    The morphological type of a microorganism generally influences its metabolite production. In the present study, we investigated the effects of the mycelial morphology of shiitake (Lentinula edodes) on the production of 2-mercaptohistidine trimethylbetaine (ergothioneine, ESH) during liquid fermentation. Analyses of the distribution of ESH in mycelial cells of different morphological types revealed that the ESH content of pellets obtained from the liquid fermentation media was much greater than the content in the free mycelia and clumps. The concentration of ESH in pellets on day 15 of liquid fermentation reached 0.79 mg/g dry weight (DW), which is approximately three times the concentration found in mycelia clumps (0.28 mg/g DW) and free mycelia (0.31 mg/g DW). Macroscopic image analysis of the development and morphological changes of the pellets during a liquid fermentation period of up to 25 days indicated that pellet growth showed a highly positive correlation with the increase in ESH concentration (r2 = 0.9851). A reduced agitation rate of 50 rpm for the culture medium was suitable for pellet formation and size enlargement. The results obtained in this work would be helpful in guiding the intentional manipulation of the distribution and enrichment of ESH in L. edodes through changes in liquid fermentation conditions.

    Endophytic fungi from Nyctanthes arbor-tristis were isolated and evaluated for their antimicrobial activity. A total of 19 endophytic fungi were isolated from 400 segments of healthy leaf and stem tissues of N. arbortristis. Eighteen endophytic fungi were obtained from leaf, while only ten from stem. Alternaria alternata had the highest colonization frequency (15.0%) in leaf, whereas Cladosporium cladosporioides ranked first in stem with a colonization frequency of 12%. The diversity and species richness were found higher in leaf tissues than in stem. The similarity indices between leaf and stem were 0.473 for Jaccard's and 0.642 for the Sorenson index, respectively. Of 16, 12 (75%) endophytic fungal extracts showed antibacterial activity against either one or more pathogenic bacteria. The endophytic Nigrospora oryzae showed maximum inhibition against Shigella sp. and Pseudomonas aeruginosa. The leaf endophytes Colletotrichum dematium and Chaetomium globosum exhibited a broad range of anibacterial activity and were active against Shigella flexnii, Shigella boydii, Salmonella enteritidis, Salmonella paratyphi, and P. aeruginosa. Nine out of 16 (56.25%) endophytic fungi exhibited antifungal activity to one or more fungal pathogens. Colletotrichum dematium inhibited 55.87% of the radial growth of the phytopathogen Curvularia lunata. The antimicrobial activity of these endophytic microorganisms could be exploited in the biotechnological, medicinal, and agricultural industries.

    A total of 202 strains of xylariaceous fungi (183 endophytic strains isolated from 25 plant species of 24 genera in 21 families and 19 saprobic strains), segregated into four clades, were examined by nuclear rDNA internal transcribed spacer (ITS) sequence and beta-tubulin coding region analyses to clarify their taxonomic status and species boundaries. Three of the four species clades were assigned to Xylaria cubensis (100 strains), Xylaria grammica (33 strains), and Nemania diffusa (48 strains). Another fungus was tentatively assigned to Nemania cf. bipapillata (21 strains). Comparison of the host plants revealed that X. cubensis inhabited healthy leaves of at least 24 plant species (23 genera of 21 families) as endophytes; N. diffusa was found on 19 plant species (18 genera of 15 families), Nemania cf. bipapillata on 11 species (10 genera of 9 families), and X. grammica on 8 species (8 genera of 7 families). The present results suggest that the major xylariaceous endophytes in tropical plants are likely to be non-host specific, or have a wide range of host plant preferences.
