The implication of neutrophils-derived oxygen radicals in the formation of ischemia-reinfusion (I-R) injury in the rat stomach was evaluated.
Male SD rats were subjected to 20min hypotension and 20min reinfusion of shed blood as we described previously (Gastroenterology 88: 1162, 1985). The area of gross gastric lesions was measured and scores of histologic damage assessed by our criteria as reported previously (Gastroenterology 94: 1135, 1988). Luminol-dependent chemiluminescence (CL) was assessed in blood samples withdrawn from the portal vein and abdominal aorta immediately before killing rats. In a study prior to the I-R experiment, 3ml of anti-neutrophils monoclonal antibody (ANA) given i.p. was confirmed to significantly reduce the number of neutrophils in blood from the carotid artery with the reduction rate being constant by 6 to 18hr after. On the basis of this results, rats were administered ANA i.p. at the same dose 12hr before I-R. Controls received normal rat serum.
The area of gross gastric lesions in the corpus (69vs. 191mm
2), but not in the antrum (13vs. 20 mm
3), was significantly reduced in the ANA group compared to that in control. The scores of histologic damage was significantly smaller in both the corpus (1.2vs. 2.0) and antrum (0.8vs. 1.5) in the ANA group than in the control. ANA significantly reduced the number of neutrophils and the CL levels in blood from both the portal vein (number, 107vs. 915; CL, 26vs. 235 RLU) and abdominal aorta (number, 52vs. 368; CL, 24vs. 268 RLU) in the ANA groups as compared to controls. There was no significant difference in the CL activity of each neutrophil from both blood samples between the ANA and control groups.
These results suggest that oxygen radicals derived from neutrophils may be implicated in the formation of gastric injury induced by ischemia-reinfusion in the rat.

The objective of this study was to assess, in a controlled experimental system, whether the concentrations of polyamines (Pa) in urine or erythrocytes increase during the process of colon carcinogenesis in rats, and whether such changes reflect tumor volume. Colon carcinoma was induced in Sprague-Dawley rats by subcutaneous injection of 1, -2 dimethylhydrazine (DMH). 24-hour urine and blood samples were collected and analysed for their Pa concent, with highpressure liquid chromatographic method. The levels of any Pa derivatives in either urine or erythrocytes at 32 weeks after the administration of DMH were not significantly high compared with the control group at the same week. However, certain increases of Pa in erythrocytes were observed in the levels of putrescine (1.3 times), spermidine (1.3 times), and spermine (1.5 times) respectively compared with the control group and there were high positive correlations between tumor volume and the levels of each Pa derivatives. On the ground of the results, Pa in erythrocytes seems to be the sensitive parameter for tumor volume compared with Pa in urine.

The sera from patients with various liver diseases were investigated for the antibody against calmodulin (CaM) extracted from bovine brain by the enzymelinked immunosorbent assay.
The specificity and purity of CaM were confirmed by the Western blot technique using anti-CaM antibody (anti-CaM) positive sera.
IgA class antibody was frequently detected in patients with hepatocellular carcinoma (HCC), autoimmune hepatitis (AIH) and chronic active hepatitis (CAH). On the other hand, IgG class antibody was very often present in patients liver cirrhosis, AIH and acute viral hepatitis (AVH). Sixty seven percents of patients with AVH in the acute phase were positive for IgM class anti-CaM and 33% of patients with AVH in the convalescent phase positive respectively. In AVH, the titer of anti-CaM reached its peak on 26.3 days after the onset. The titer of anti-CaM in fulminant hepatitis was higher than that in AVH. Seventy percents of type A hepatitis patients were positive for IgM class anti-CaM, 33% of type B and 33% of type non-A non-B.
These results suggest that the frequency and titer of anti-CaM may depend upon the type of hepatitis and the degree of liver cell injury.

Localization of epidermal growth factor receptor (EGFR) in hepatocellular carcinoma was examined by means of immunohistochemistry. It's localization was observed at plasma membrane of the cancer cells. EGFR was found to be expressed in 7 out of 17 hepatocellular carcinoma. No significant difference was observed between EGFR-positive and EGFR-negative cases in tumor size. AFP, macroscopic and histopathological classification, capsular invasion, and portal invasion. However, in EGFR-positive cases, the number of DNA polymerase-α positive cells and recurrence rate were higher than that in EGFR-negative cases.
These results suggest that EGFR may play an important role in the development and progression of hepatocellular carcinoma.

To evaluate the clinical significance of blood flow in the azygos vein and the oxygen partial pressure in azygos venous blood in portal hypertension, we examined 25 patients with liver cirrhosis, 4 with chronic hepatitis, 4 with idiopathic portal hypertension (IPH) and 16 controls by the continuous thermodilution method and azygos venous blood sampling. The azygos venous flow was significantly higher in the patients with chronic liver diseases than in the controls. There was a significant correlation between azygos venous flow and hepatic venous pressure gradient. In the patients with liver cirrhosis, about one half of the azygos venous flow was assumed to represent upward collacteral flow, as the azygos venous flow was 3.4% of cardiac output in the controls, 6.3% in the liver cirrhosis patients, and 5.1% in the IPH patients. Oxygen partial pressure in azygos venous blood was higher in patients with portal hypertension, especially in the IPH patients, which indicates that part of splenic flow drains into the azygos vein.

In the present study, we have measured the serum levels of laminin and PIIIP in patients with various liver disease, and studied serial changes in serum laminin and PIIIP after TAE.
The following results were obtained.
1) Serum levels of laminin as well as PIIIP were significantly higher in liver cirrhosis, liver cirrhosis with hepatocellular carcinoma and chronic active hepatitis compared to that in healthy controls, and serum levels of laminin revealed significant correlation with serum levels of PIIIP in liver cirrhosis with hepatocellular carcinoma. While serum levels of PIIIP were elevated significantly in acute hepatitis, serum levels of laminin were not elevated.
2) There was no significant difference in the serum levels of laminin between liver cirrhosis and liver cirrhosis with hepatocellular carcinoma. The results suggested that serum levels of laminin as well as PIIIP cannot be a specific marker of hepatocellular carcinoma.
3) In the study of serial changes of these peptides after TAE, changes in the serum levels of laminin in effective cases were different from those in ineffective cases. The results suggested that measurement of serial changes in the serum levels of laminin after TAE was considered to be useful for evaluating the effectiveness of TAE. Also, changes in serum levels of laminin after TAE were different from those observed in serum PIIIP. The results suggest that mechanism of release of laminin into serum in hepatocellular carcinoma may be different from that of PIIIP.

We studied relationship between an involvement of Oddi's sphincter in a choledochopancreatic junction and clinical appearances in 120 patients with anomalous arrangement of the pancreaticobiliary ductal system. The 120 composed of 83 with a sphincteric mechanism at the junction of the common bile and pancreatic duct and 37 without the sphincteric mechanism. We found that influences of sphincteric mechanism at the junction closely related to the length of the common duct between an orifice of the papilla of Vater and a junction of the common bile and pancreatic duct. And in all 83 with a sphincteric mechanism at the junction the length of the common duct showed less than 1.5cm, and in all 37 without a sphincteric mechainsm at the junction the common duct showed more than 1.5cm in length. There was no correlation between a distribution of the sphincter muscle and clinical appearances, association with congenital choledochal cyst, cholelithiasis and bile duct cancer in the 120 patients.

Using sera from patients with pancreatic cancer and chronic pancreatitis, we measured the level of the adenocarcinoma-associated antigen MUSE 11. A comparative study between levels of MUSE 11 and levels of sialyl SSEA-1 antigen (SLX) was also carried out. With respect to the MUSE 11 antigen, positive incidence was found in 17 out of 26 pancreatic cancer patients (65%), and in 1 out of 13 chronic pancreatitis patients (8%).
Similar results were obtained from the assay of SLX levels. However, no correlation was found between the two markers. Out of 9 samples which showed MUSE 11 negative, three were positive for SLX. Out of 13 samples showing SLX negative, seven were positive for MUSE 11. Twenty out of 26 (77%) cases showed positive results for either antigen. These data suggest that the MUSE 11 antigen may be useful for the diagnosis of pancreatic cancers.
