  • (第2報) Aspergillus屬菌類の分類學的位置に就て

    1. The classification of the Aspergilli, isolated from tl different forest soils reported in the 1st report (On the fungus flora of the different forest soils.) was studied.
    2. In the forest soils examined by us Aspergilli are not generally dominant in the fungus flora.
    3. Among them, 3 species, A. niger VAN TIEGHEM, A. japonicus SAITO, and A. oryzae var. microsporus SAKAGUCHI et YAMADA f. sp. flavo-viridis were relatively widely distributed and they account for 1 to 4 % of the total no. of colonies in some forest soils.
    4. 9 other species of Aspergilli were occasionally isolated. A. giganteus WEHMER, A. fumigates, F. FRES. f. sp. floeeosus and f. sp. veltinus, A. sydowi BAIN. et SART. A. sojae SAKAGUEHI et YAMADA f. sp. flavo-viridis, A. oryzae var. microsporus SAKAGUCHI et YAMADA f. sp. fuivescens, A. ustus BAIN., A. ochraceus var. microsporus TIRABOSHI and A. ochraceus var. microsporus TIRABOSHI f. Sp. floccosus.

    1. 低壓負荷状態下に於けるしろねずみの飼育試瞼驗に於て,Sporobolomyces屬赤色酵母を經口的に投與すれば麥酒酵母を投與せる場合と同様に良好なる成育を示すが,赤色酵母と麥酒酵母とは其の組成上大いに異るので成育上好結果を與えたる原因は夫々全く異るものと考えられる.
    2. 當赤色酵母中には普通酵母中に存する血糖降下性物質グルコキニンと類似した作用物質を看することを家兎に就て證明し得た.而して其の血糖降下作用は赤色酵母菌體其の儘を新鮮物として或は乾燥物として與えたる場合何れも有效であるが,普通酵母では全く作用が認められない.又家兎に對し低壓を負荷する場合に現れる過血糖は赤色酵母を投與することにより抑制せられ,低壓負荷状態下に於けるしろねずみの成育に好影響を與えたる原因は過血糖を抑制するためではないかと考えられる.
    3. 當赤色酵母の菌體及び培養液には絲状菌特に黒黴の繁殖を抑制する作用があるが,其の様な靜黴作用のある場合には同一區分に就て血糠降下作用をも認められ,從つて靜黴作用を以て血糖降下作用の本態を迫及して行く場合の一指標となし得るのではないかと考えられる.

    Sporobolomyces赤色酵母培養申血糖降下作用性物質及び靜黴作用性物質の生産される時期と分布及びその相互隨伴關係を明らかにするため,その増殖生理と培養液並に細胞の化學的變化を竝行的に追及した結果次の事を知つた.
    1) 赤色酵母の培養初期に於て培養液中に酸度曲線と略並行して焦性葡萄酸及びメチール・グリオキザールが少量一時停滞する.然し之等物質は酵母の最高増殖期には既に殆んど消失している.
    2) 靜黴作用性物質には溜出性及び非溜出性の2種類があり,前者は上記還元性物質と同じく培養初期に於て産生され,後者は本來は細胞内にあり培養後期に於て自己消化の結果培養液に移行するものと言える.
    3) 血糖降下作用性物質は非溜出性靜黴作用物質とその消長ほぼ並行し,自己消化期迄は菌體内にあり,自己消化の結果培養液に移行する.

    We examined the color reaction of leaves of glutinous rice, dahlia, and Jerusalem artichoke by Sachs' iodine test and observed that all these leaves colored blue with iodine. So we attempted to isolate the assimilation starch from boiling-water extract of leaves by Schoch's butanol-pre-cipitating method or thymol-precipitating method of Haworth et al. Using leaves of squash or Jerusalem artichoke we successed in isolating the assimilation starch as microscopic crystals. By comparing the precipitability and X-ray diffraction pattern of the assimilation starch with those of corn amylose, it was confirmed that the assimilation starch was very similar to amylose.

    In the studies on yeasts by electron microscope the following results were obtained.
    1. It was observed that all kinds of yeasts that I used had mucous substance around their bodies. But from differences of their figures on the screen, it was surmised that there were differences in quality and quantity of mucous substance among kinds of yeasts.
    2. It was ascertained that floating yeasts in liquid-media during fermentation were not simple naked ones. In some cases (Fig.9) mucous substance including gas bubbles was noti-ced around the floating yeasts, and in other cases (Fig. 10) there were many fragments which were composed of only mucous substance including gas bubbles.

    In the study on turbidity of liquid-media during cultivation of yeast the following results were obtained.
    1. In more than one half of the kinds of yeasts that I used (14 Genera, 53 species) turbidity of media was found. It was usually milky, powdery or scum-like.
    2. Except in one case, the turbidity of media had a close connection to generation of gas bubbles by yeast, namely they both occurred in company, increased and decreased in parallel, and came to the end about the same time.
    3. From the relation between generation of gas bubbles and turbidity of media, all kinds of yeasts that I used can be classed into the following groups:-

    1. An antibiotic substance, active against both gram-positive and gram-negative bacteria, was obtained from the culture filtrate of Actinomyces 700.
    2. It was extracted and purified by the same method as streptothricin and streptomycin.
    3. In our opinion this substance is one of the streptothricin-like substances.

    In Otaki area of Kiso, there remain salt-less pickles of ancient origin. The author isolated Leuconosioc meesenteroides and Streptococcus faecalis. The former is to be supposed as the principal organism of the pickles.

    The data were reporte_??_as a type of fermentation and of nutritional loss in the silage of med_??_um quality.

    1. For the purpose of making proteinous foods (including raw food material) Aspergillus oryzae was cultured on following media: (1) dehydrated sweet-potato flakes; (2) sweet-potato powder; and (3) sweet-potato powder with rice-bran. As the nutrients of the mould-fungi ammonium sulphate and calcium carbonate were added.
    2. The increase of protein is the greatest in medium (3) when 2 per cent. of ammonium sulphate was added successively five times (Sample No.6, Table 14), and it was 1.09g for 10g of sweet-potato powder used.
    3. The maximum content of protein in koji is obtained in medium (2) when 3 per cent of ammonium sulphate was added repeatedly twice (Sample. No. 5, Table 10). The content of protein reached 19.64 per cent in koji.
