    The purposes of this study were to clarify morphological characteristics of tissue diversity of salivary pleomorphic adenomas (PA), and to examined cell adhesion and formation of mesenchyma-like tissue using histopathological and immunohistochemical methods. Histopathologically, tumor parenchyma showed ductal epithelium-like cells on the inner side which enclosed the lumen (first layer), surrounded by stratified myoepithelium-like cells (second layer). In the outer part of the second layer, myoepithelium-like cells were dispersed (third layer) toward the mesencyma-like tissue. Immunohistochemically, E-, N-cadherins and β-catenin positive tumor cell rates decreased as they transitioned from the first to third layers, and the ratio of N-cadherin positive cells remained higher than that of E-cadherin in the third layer. Further, vimentin, actin, and p63 positive cell rates increased and collagen type IV positive basement membrane tended to disappear as they transitioned from the first to third layers. These findings suggested that the epithelial tumor cells changed their characteristics to mesenchymal cells.

    Using pathological and immunohistochemical approaches, we investigated whether lingual injection of B16 cells spontaneously induces cervical lymph node metastasis and whether the extent of tumor lymphangiogenesis is related to the incidence of lymph node metastasis during the local progression of a tumor. Primary tumors developed into unilateral (phase I), separated (phase II), and fused (phase III) patterns. The rate of cervical lymph node metastasis increased in both phases II and III. Intratumoral lymphatic vessel density (LVD) was significantly higher in phase III, whereas increase in peritumoral LVD started in phase II. In this study, we demonstrate the development of cervical lymph node metastasis induced by the progression of melanoma in the tongue, and provide unique evidence that an increased LVD may heighten the risk of lymph node metastasis. In particular, the peritumoral vessels may play an important role in the propensity for early metastatic spread to regional lymph nodes.

    Odontogenic myxoma, originating from the odontogenic ectomesenchyme, is an intraosseous benign neoplasm characterized by the presence of stellate and spindleshaped cells proliferating in an abundant myxoid or mucoid extracellular matrix. We experienced a giant case of this type of neoplasm. The patient was a 40-year-old Cambodian female with a presenting complaint of painless swelling of her gums since which had been present for 15 yaers. We examined the histopathological features and compared the mesenchymal tumoral tissue characteristics of this case with those of an ameloblastic fibroma with respect to Notch1 signaling. By immunohistochemical examination of Notch1 intracellular domain (NICD), we found that unlike the ameloblastic fibroma, NICD positive products were not observed in the current case of giant odontogenic myxoma. This negative staining result suggests that the ectomesenchymal tumoral proliferating cells of this neoplasm differ in their development stage from those of an ameloblastic fibroma.
