Orchidectomized adult rats of the Sprague-Dawley strain were given a transplant of an ovary from immature donors under the abdorminal skin. When sacrificed 40 days after operation, in 57-60% of the animals given no further treatment and of those receiving 10 daily injections of 0.1mg progesterone or a single injection of 3ml of 1% croton oil into a dorsal subcutaneous air pouch, ovarian grafts had some corpora lutea. Ovarian grafts in the rest of the animals contained follicles of varying sizes but no corpora lutea. If the daily dose of progesterone was raised to 0.25mg, incidence of luteinization slightly increased (67%). By contrast, in rats bearing the croton oil injected pouch, daily injections of 0.1 and 0.25mg progesterone elicited luteinization in ovarian grafts in 89 and 100% of the hosts, respectively. Therefore, it seems highly probable that daily progesterone injections and mild stressful stimuli cooperate in inducing luteinization in subcutaneous ovarian grafts in orchidectomized rats.
